Abstract

Die vorliegende Erfindung betrifft ein Herstellungsverfahren für Mumbaistatin-Derivate (I), wobei das Anthrachinon-Grundgerüst über eine Diels-Alder-Reaktion und die zentrale Methylenbrücke über eine Übergangsmetall-katalysierte Reaktion aufgebaut wird, sowie die in diesem Verfahren verwendeten Intermediate.

Verfahren zur Herstellung von Mumbaistatin-Derivaten
Die vorliegende Erfindung betrifft Herstellungsverfahren für Mumbaistatin und Mumbaistatin-Derivate sowie die in diesen Verfahren verwendeten Intermediate.
Mumbaistatin ist ein aromatisches Diketoderivat, das als Glukose-6-phosphatase Translokase Inhibitor in der Behandlung von Diabetes mellitus verwendet werden kann. Mumbaistatin (A) ist aus dem Mikroorganismus DSM11641 isolierbar (Vertesy et al., WO99/67408 und J. Antibiot. 2001 , 54, 354-363). WO01/30736 beschreibt die Mumbaistatin-Derivate (B) - (D) sowie Ester und Ether davon. Mumbaistatin liegt in einem Äquilibrium einer offenen Form (A) und einer Hemiacetalform (B) vor und kann in Abhängigkeit vom pH-Wert in die Verbindungen (B) und (D) umgewandelt werden.
Krohn et al. (Tetrahedron 2006, 62, 1223-1230) beschreiben eine 7-9-stufige Synthese der tri-o/ϊΛo-substituierten 2'-Dealkyl-Mumbaistatin-Derivate der Formel (E), wobei die Oxidation der benzylischen Position mittels Br2 in einem CCI4-
Wasser-Gemisch unter Bestrahlung gelang,
und wobei das Anthrachinon-Grundgerüst durch eine mehrstufige Aldol- Kondensation einer durch Lewis-Säure katalysierte Michael-Addition hergestellten Dien-Zwischenstufe aufgebaut wird, und die Gesamtausbeute ausgehend vom Naphthylderivat (F) 2,8 bis 14,1 % beträgt.
Kaiser et al. (J. Org. Chem. 2002, 67, 9248-9256; Tetrahedron 2003, 59, 3201-3217) beschreiben die 9-stufige Synthese des tri-orfΛo-substituierten 3'-Deoxy-2- decarboxy-Mumbaistatin-Derivats (G),
wobei das Anthrachinon-Grundgerüst (H) über eine Arin-Phthalid-Anellierung hergestellt wurde, und die Gesamtausbeute ausgehend vom käuflichen 3- Hydroxybenzaldehyd 3,7 % beträgt. Aufgabe der vorliegenden Erfindung ist es, einen effizienten Syntheseweg zu Mumbaistatin-Derivaten bereitzustellen.
Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung von Mumbaistatin- Derivaten der Formel (I) oder von Salzen davon,
wobei
R1 und R2 unabhängig voneinander ein H, (C-|-C6)Alkyl oder Benzyl sind, R3, R4 und R5 unabhängig voneinander OH, O-(C-|-C6)Alkyl, O-Benzyl oder
O-Acyl sind,
R6 OH, Halogen, (C -| -CO)AI kyl, O-(C-|-C6)Alkyl, Phenyl, Benzyl, O-Phenyl,
O-Benzyl, O-Acyl ist,
X1 und X2 unabhängig voneinander O, NH, N(C-I -CβJAlkyl oder S sind, und m, n, q und r unabhängig voneinander O oder 1 sind,
wobei in Schritt (1 ) eine Verbindung der Formel (II)
(H) in der Y eine Abgangsgruppe ausgewählt aus der Gruppe HaI, OTs, OTf oder OMs ist, und vorzugsweise Chlor oder Brom ist, mit einer Verbindung der Formel

in Schritt (3.1 ) die Verbindung (V) mit einer metallorganischen Verbindung (VI)
in der M Sn[(Ci-C6)Alkyl]3, B(OH)2, B(OR)2, BF3 ", ZnHaI oder MgHaI bedeutet, und (OR)2 gleich [O-(C-|-C6)Alkyl]2 oder ein 1 ,2-vicinaler Dialkohol, beispielsweise
Pinakol, Cathechol bedeutet, wobei ein aus dem vicinalen Dialkohol abgeleiteter Boronsäureester entsteht, unter Übergangsmetall-Katalyse, vorzugsweise unter Pd-, Ni- oder Fe-Katalyse zu einer Verbindung der Formel (VII) umgesetzt wird,
und anschließend die Verbindung (VII) zu einer Verbindung (VIII) oxidiert wird,
oder alternativ in einem Schritt (3.2) die Verbindung (V) mit einer metallorganischen Verbindung (Vl')
wobei M und R6 wie für Verbindung (VI) definiert sind, und Z O-(CrC6)Alkylen-O, vorzugsweise O-(CH2-CH2)-O bedeutet, unter Übergangsmetall-Katalyse, vorzugsweise unter Pd-, Ni- oder Fe-Katalyse zu einer Verbindung der Formel (VIT) umgesetzt wird,

und anschließend die Verbindung (VIT) zu einer Verbindung (VIII) durch Umsetzung mit einer geeigneten Säure hydrolysiert wird,
oder alternativ in einem Schritt (3.3) die Verbindung (VI) mit einer Verbindung der Formel (V), in der X2R2 gleich OH ist, zu einer Verbindung der Formel (VM") verestert wird,
und anschließend die Verbindung (VN") unter Übergangsmetall-Katalyse, vorzugsweise unter Pd-, Ni- oder Fe-Katalyse zu einer Verbindung der Formel (VH'") umgesetzt wird,
und die Verbindung (VN'") zu einer Verbindung der Formel (VIII) umgesetzt wird, indem unter basischen oder unter sauren Bedingungen, vorzugsweise unter basischen Bedingungen hydrolysiert, anschließend der entstandene Benzylalkohol zum Aldehyd oxidiert und optional die entstandene Benzoesäure-Funktion verestert oder amidiert wird,
in Schritt (4) die Verbindung (VIII) mit einer Verbindung der Formel (IX)
zu einer Verbindung der Formel (X) umgesetzt wird,
in Schritt (5) die Verbindung (X) unter Hydrierung der Dreifach-Bindung zu einer Einfachbindung und Abspaltung der [(C-|-C6)Alkyl]3Si-Gruppe in eine Verbindung der Formel (Xl) umgesetzt wird,
(Xi). in Schritt (6) die Verbindung (Xl) zu einer Verbindung der Formel (XII) oxidiert wird,
in Schritt (7) die Verbindung (Xl) oder die Verbindung (XII) zu einer Verbindung der Formel (I) oxidiert wird, in der X1 R1 = OH ist, und
in Schritt (8) optional die in Schritt (7) erhaltene Verbindung zu einer Verbindung der Formel (I) umgesetzt wird, in der X1 NH, N(Ci-C6)Alkyl oder S ist, und R1 gleich
(C-|-C6)Alkyl oder Benzyl ist, oder einem Salz davon.
Im Zuge der Synthese von Verbindungen der Formel (I) werden vorzugsweise die OH-Funktionen an R3, R4, R5 und/oder R6, sofern vorhanden, vorübergehend mit einer geeigneten Schutzgruppe versehen, beispielsweise mit einer Methyl-, Methoxymethyl-, Benzyl- oder p-Methoxybenzyl-Gruppe.
Im Zuge der Synthese der Verbindungen der Formel (I) kann es notwendig oder wünschenswert sein, intermediäre Verbindungen enthaltend C(O)XI R1- und/oder C(O)X2R2- Gruppen, soweit in den Verbindungen der Formeln (I), (IV), (V), (VII), (VIII), (VII1), (X), (Xl) oder (XII) vorhanden, ineinander umzuwandeln. Dies kann auf dem Fachmann an sich bekannten Wegen geschehen. Zum Beispiel können Ester unter wässrigen sauren bzw. basischen Bedingungen zu freien Säuren hydrolysiert werden. Die freien Carbonsäuren lassen sich dann mit Basen leicht in Salze überführen. Geeignete Basen sind zum Beispiel Alkalimetallhydroxide wie NaOH oder KOH. Weiterhin können zur Herstellung von alternativen Carbonsäurederivaten Ester oder Carbonsäuren, nach üblicher Aktivierung z.B. mit Thionylchlorid, in Gegenwart einer Base mit Nukleophilen (beispielsweise NH2(Ci-C6)Alkyl, NH2-
Benzyl, NH[(C-|-C6)Alkyl]2, NHt(C1 -C6)Alkyl][benzyl], [(C-|-C6)Alkyl]-SH, Benzyl-SH) umgesetzt werden. Dabei ist es möglich, soweit beide vorhanden sind, entweder beide Säure(dehvat)funktionalitäten gemeinsam umzusetzen, selektiv nur eine allein oder beide unabhängig voneinander nacheinander.
Der Ausdruck 'Halogen' und 'HaI' bedeutet Fluor, Chlor, Brom oder lod, bevorzugt Chlor oder Brom.
Der Ausdruck '(C-] -Cs)AI kyl' bedeutet eine geradkettige oder verzweigte, optional mit
1 , 2, 3 oder 4 Substituenten substituierte (C-]-C6)Alkyl-Gruppe, wobei die Substituenten ausgewählt sind aus der Gruppe OH, (C-|-C4)Alkyl, (C-|-C4)Alkoxy, (C-i-C4)Alkylthio, Carbamoyl, Carboxyl, Trifluormethyl, Cyano, Nitro, Amino, (C-|-C4)Alkylamino, Di-((C-|-C4)alkyl)amino, Amidino, Phenyl, O-Phenyl, O-Benzyl, NH-Phenyl, NH-Benzyl oder Halogen, -Phenyl-(C-|-C4)Alkyl, -Phenyl-O- (C-|-C4)Alkyl, vorzugsweise eine (C-|-C4)Alkyl-Gruppe. Beispielsweise ist '(C-|- CßJAlkyl' Methyl, Ethyl, n-Propyl, Isopropyl, n-Butyl, Isobutyl, tert-Butyl,
Methoxymethyl-, Benzyl- oder p-Methoxybenzyl.
Der Ausdruck 'Acyl' bedeutet -C(O)-(C-|-C6)Alkyl, beispielsweise Acetyl oder
Propanoyl; -C(O)-(C6-C-|rj)Aryl oder -C(O)-(Ci -C4)Alkylen-(C6-C<ιo)Aryl, wobei (C6-C-|o)Aryl eine 6 bis 10 Kohlenstoffatome enthaltende aromatische Gruppe bedeutet, beispielsweise Phenyl oder ToIyI; oder -C(O)-(C5-C6)Heteroaryl, wobei (C5-Cß)Heteroaryl einen 5-6-gliedrigen Ring mit 1 , 2, 3 oder 4 Heteroatomen bedeutet, beispielsweise Pyridyl, Furyl, Pyrrolyl, Thienyl, Thiazolyl oder Oxazolyl.
Ein Salz der Verbindung der Formel (I) enthält als Kation beispielsweise ein anorganisches Metallion oder ein Ammoniumion. Insbesondere bedeutet der Ausdruck 'Kation' ein pharmazeutisch verträgliches Alkali- oder Erdalkali-Metallion, beispielsweise Natrium, Kalium, Kalzium oder Magnesium, ferner NH4+, ein mit (C-|-C6)Alkyl-alkyliertes Ammoniumion wie beispielsweise Tetraethylammonium, Diethanolammonium, Morpholinonium oder Benzylammonium, oder protonierte Formen von Procain, L-Arginin oder L-Lysin.
Eine bevorzugte Verbindung der Formel (I) ist eine Verbindung der Formel (I-A)
wobei R1 bis R6, X1 , X2, m und n unabhängig voneinander die oben genannte allgemeine oder bevorzugte Bedeutung haben.
Bevorzugte Definitionen der Substituenten der Verbindung der Formel (I) oder (I-A) sind wie folgt: R1 und R2 sind bevorzugt unabhängig voneinander ein H oder (Ci-Cß)Alkyl, besonders bevorzugt H oder (C-|-C4)Alkyl.
R2 ist besonders bevorzugt H. R3 ist bevorzugt OH.
R4, R5 und R6 sind bevorzugt unabhängig voneinander OH oder -O-(C-|-C6)Alkyl. X1 und X2 sind bevorzugt O. m ist bevorzugt 0. q ist bevorzugt 1. r ist bevorzugt 1. Speziell bevorzugt ist eine Verbindung der Formel (I-A), in der X1 R1 gleich OH oder O-(Ci-Ce)Alkyl ist,
X2R2 gleich OH ist, R3 gleich OH ist, R4 gleich OH oder O-(Ci -C6)Alkyl ist,
R5 gleich OH oder O-(C-i-C6)Alkyl ist,
R6 gleich OH oder 0-(C-I-Ce)AIkVl ist, und m und n unabhängig voneinander O oder 1 sind.
Schritt (1 ) setzt sich aus zwei experimentell separaten Teilreaktionen zusammen, einer [4+2]-Cycloaddition und einer nachfolgenden Aromatisierung (via Eliminierung). Vorzugsweise bedeutet X2R2 in der Verbindung (III) eine in der späteren Reaktionsfolge unter milden Bedingungen, z.B. mittels TMS-OTf, abzuspaltende Gruppe, beispielsweise O-tert-Butyl. Verbindungen vom Typ (II) können hergestellt werden nach literaturbekannten
Verfahren (Rapoport et al., J. Org. Chem. 1979, 44(13), 2153-2158; Cameron et al., Aust. J. Chem. 1981 , 34, 1513-1522).
Der [4+2]-Cycloadditionschritt zu Verbindungen vom Typ (IV) wird durchgeführt durch Erhitzen der Komponenten (II) und (III) über einen Zeitraum von 1 bis 48 h, vorzugsweise 2-24 h, bei einer Temperatur von 80-1800C ohne Lösungsmittel oder in einem geeigneten inerten Lösungsmittel oder in einem Gemisch von inerten Lösungsmitteln, beispielsweise einem aliphatischen, alicyclischen oder aromatischen Kohlenwasserstoff, z.B. Toluol, XyIoI oder Benzol. Optional wird Schritt (1 ) in Gegenwart einer Lewis-Säure durchgeführt, beispielsweise TiCI4, Ti(Z-Pr-O)4,
Ti(Z-Pr-O)2CI2, AICI3 oder Sc(OTf)3. Die Reaktion wird prinzipiell durch Zusatz von Lewis-Säuren beschleunigt und kann dann auch bei niedrigeren Reaktionstemperaturen (<110 0C) durchgeführt werden, wobei im Einzelfall aber Nebenreaktionen auftreten können. Schritt (1) kann optional mittels eines Mikrowellenreaktors durchgeführt werden, ferner ist die Verwendung von ionischen Flüssigkeiten als Reaktionsmedium möglich. Sofern Nebenreaktionen auftreten, könne diese durch die Verwendung von Zusätzen, beispielsweise organische oder anorganische Basen, Protonenschwämme oder Radikalfänger, unterdrückt werden, beispielsweise mittels Alkalimetallcarbonaten.
Die Aromatisierung des primären [2+4]-Cycloadditionsproduktes wird anschließend durchgeführt, indem es mit einer geeigneten Säure, z.B. Kieselgel, versetzt wird und bei 15-700C für 0,5 bis 24 Stunden gerührt wird. Vorzugsweise wird das Lösungsmittel des ersten Teilschritts durch Wasser und ein mit Wasser mischbares organisches Lösungsmittel, beispielsweise THF, ersetzt. Falls die Gruppe R' gleich (CrC6)Alkyl ist, wird diese unter den sauren Aromatisierungsbedingungen i. A. nicht abgespalten, so dass direkt Verbindungen vom Typ (IV) mit R" = (C-i-CβJAlkyl entstehen. Falls die Gruppe R' gleich [(C-ι-C6)Alkyl]3silyl ist, kann diese unter den sauren Reaktionsbedingungen abgespalten werden. Hierfür geeignet sind verdünnte wässrige anorganische oder organische Säuren, beispielsweise Salzsäure, Schwefelsäure, Essigsäure oder p-Toluolsulfonsäure. Besonders geeignet ist
Kieselgel, welches gegebenenfalls vorab mit einer dieser Säuren behandelt wurde.
In Schritt (2) wird zunächst nach dem Fachmann an sich bekannten Verfahren die optionale Veretherung bzw. Veresterung der OH-Gruppe von (IV) durchgeführt, beispielsweise durch Umsetzen mit einem (C-|-C6)Alkylhalogenid, vorzugsweise
Methyliodid, Benzylhalogenid oder einem aktivierten Carbonsäurederivat, beispielsweise einem Acylchlorid oder -anhydrid, in Gegenwart einer organischen oder anorganischen Base, z.B. Kaliumcarbonat, in einem geeigneten polaren organischen Lösungsmittel, beispielsweise Aceton, DMSO oder DMF, bei einer Temperatur von 0-800C und Reaktionszeiten von 0,5-12 h.
Die weitere Umsetzung zu (V) erfolgt durch radikalische Halogenierung der Methylgruppe unter dem Fachmann an sich bekannten, photochemischen und/oder thermischen Reaktionsbedingungen, wobei im letzteren Fall ein Radikalstarter verwendet wird. HaI in der Verbindung (V) bedeutet bevorzugt Brom. Die Umsetzung wird beispielsweise durchgeführt durch 5 bis 24-stündiges Bestrahlen mit Licht geeigneter spektraler Zusammensetzung (z. B. Sonnenlicht oder Scheinwerfer mit relativ hohem UV-Anteil) von (IV) und Λ/-Brom-Succinimid in einem inerten Lösungsmittel, beispielsweise ein perhalogenierter Kohlenwasserstoff, vorzugsweise Tetrachlormethan. Vorzugsweise werden in bestimmten zeitlichen Abständen (z.B. 15-180 min) Anteile eines Radikalstarters, vorzugsweise Dibenzoylperoxid oder Azo- b/s-isobutyronitril (AIBN), zugesetzt.
Schritt (3.1 ) wird durchgeführt nach dem Fachmann an sich allgemein bekannten Methoden für Übergangsmetall-katalysierte Kreuzkupplungsreaktionen (Nobre & Monteiro, Tetrahedron Lett. 2004, 45 8225-8228; Phopase et al., Tetrahedron Lett. 2004, 45, 6959-6962), beispielsweise wird die Verbindung (V) mit einer metallorganischen Verbindung vom Typ (VI), in der gegebenenfalls die OH-Funktion mit einer geeigneten Schutzgruppe versehen ist, in einem Temperaturbereich von 0- 150 0C, vorzugsweise 5-4O0C, und einer Reaktionszeit von 1-72 h umgesetzt. Geeignete Schutzgruppen sind beispielsweise [(Ci-C6)Alkyl]3silyl], (C-i-CβJAlkanoyl oder Acetal Z, wobei im Fall von Z bei der Herstellung von (Vl') zunächst nach bekannten Methoden oxidiert und anschließend acetalisiert wird. Bevorzugt wird die OH-Funktion mit einer [(Ci-C6)Alkyl]3silyl]-Gruppe wie Triethylsilyl- , Tri(/so- propyl)silyl- oder einer terf-Butyl-dimethylsilyl-Gruppe geschützt. Die Kupplungsreaktion kann optional in einem Mikrowellenreaktor durchgeführt werden, wobei die Reaktionstemperatur in einem Bereich von 15-300 0C liegt und die Reaktionszeit von 1 Minute bis 120 Minuten beträgt. Schritt (3.1 ) wird in einem oder mehreren inerten Lösungsmitteln, beispielsweise unpolare aliphatische, alicyclische oder aromatische Kohlenwasserstoffe wie z. B. Toluol, XyIoI, Benzol, Verbindungen schwacher Polarität wie z. B. THF oder Dibutylether, oder polare Verbindungen wie z. B. NMP1 DMF oder DMSO, vorzugsweise THF und/oder NMP, vorzugsweise unter einer Inertgasatmosphäre durchgeführt.
In Schritt (3.1 ) wird nach der Kupplungsreaktion durch dem Fachmann an sich bekannte Methoden der benzylische Alkohol oxidiert, beispielsweise durch Dess- Martin-Oxidation unter Verwendung von Dess-Martin-Periodinan in DCM als Lösungsmittel, oder vorzugsweise unter den Bedingungen einer Swern-Oxidation (Reagenzien: DMSO, z. B. Oxalylchlorid, Triethylamin; inertes Lösungsmittel: DCM; Temperaturbereich: -78 0C bis 40 0C; Reaktionszeit: 1-24 h).
In Schritt (3.2) kann der Kupplungsschritt entsprechend zu Schritt (3.1 ) durchgeführt werden, wobei eine Verbindung der Formel (Vl') eingesetzt wird, die eine geschützte Aldehydfunktion Z gleich O-(Ci-C6)Alkylen-O, vorzugsweise O-(CH2-CH2)-O enthält. Die Schutzgruppe Z wird nach der Kupplungsreaktion in einem geeigneten wasserhaltigen Lösungsmittel, vorzugsweise einem Gemisch von Wasser und einem polaren organischen Lösungsmittel, beispielsweise Aceton, bei einer Temperatur von 0-100 0C, über einen Zeitraum von 0,5-48 h mit einer geeigneten Säure, beispielsweise PTS oder vorzugsweise PPTS behandelt, wobei die Acetal-Funktion zum benzylischen Aldehyd gespalten wird.
In dem alternativen Reaktionsschritt (3.3) erfolgt der Kupplungsschritt in einer entropisch begünstigten intramolekularen Weise, wobei zunächst die
Kupplungspartner unter an sich bekannten Bedingungen miteinander zu einer Verbindung der Formel (VM") verestert werden, beispielsweise mittels einer Mitsunobu-artigen Veresterung von (VI) mit einer Verbindung (V), in der X2R2 gleich OH ist. Hierfür geeignet ist eine Reagenzkombination bestehend aus einem Azodicarbonsäurediester, beispielsweise Diethylazodicarboxylat (DEAD) oder Diisopropylazodicarboxylat (DIAD), und einem Phosphan, beispielsweise Triphenylphosphan, Tributylphosphan oder Diphenyl(2-pyridyl)phosphan. Die Umsetzung erfolgt in einem inerten Lösungsmittel (z.B. THF) in einem Temperaturbereich von -20 0C bis 100 0C bei Reaktionszeiten von 1-24 h.
Die Hydrolyse der Verbindung (VM'") nach erfolgter Kupplung erfolgt unter basischen oder unter sauren Bedingungen, vorzugsweise unter basischen Bedingungen, bevorzugt mittels wässriger NaOH- oder KOH-Lösung. Der so entstandene Benzylalkohol wird nach dem Fachmann an sich bekannten Methoden zum Aldehyd oxidiert, indem beispielsweise mittels einer Swern-Oxidation oder einer Dess-Martin- Periodinan Oxidation oxidiert wird. Die intermediär entstandene Benzoesäure- Funktion wird nach dem Fachmann an sich bekannten Methoden verestert oder amidiert, indem nach Aktivierung der Benzoesäurefunktion, beispielsweise mit Thionylchlorid, mit dem entsprechenden Nukleophil (z.B. HO(Ci-C6)Alkyl,
NH2(Ci -C6)Alkyl, NH2-Benzyl, NHt(C1-Ce)AIkYl]2, NHt(C1 -C6)Alkyl][benzyl], [(C-|-C6)Alkyl]-SH, Benzyl-SH) umgesetzt wird.
Bevorzugt wird in den Schritten (3.1 ), (3.2) und (3.3) eine Verbindung (VI) oder (Vl') mit M gleich Sn(Bu)3 eingesetzt, deren Herstellung von Meyer & Seebach (Chem. Ber. 1980, 113, 1304-1319) beschrieben ist.
In den Schritten (3.1 ), (3.2) und (3.3) wird die Kupplungsreaktion in Gegenwart einer geeigneten Übergangsmetallverbindung als Katalysator oder Katalysatorvorstufe durchgeführt, wobei Pd-, Ni- oder Fe-Verbindungen bevorzugt sind, und ferner sub- stöchiomethsche Mengen eingesetzt werden. Als Liganden der Übergangsmetallverbindungen eignen sich beispielsweise alkylierte oder arylierte Phosphor-, Stickstoff- oder Arsen-Verbindungen, sowie N-heterocyclische Carbene, welche ein- oder zweizähnige Liganden sein können. Geeignete Liganden sind beispielsweise P(Ph)3, P(cyclohexyl)3l P(2-furyl)3, AsPh3, dppe, dppp, dppf, dba, P(terf.-Bu)3 oder Ibiox7, vorzugsweise AsPh3 oder dba. Ein bevorzugter Pd-Katalysator ist Pd2(dba)3. Dem Reaktionsgemisch können optional bestimmte, reaktionsfördemde Additive, wie CsF, LiCI oder Kupfer(l)halogenide zugesetzt werden, bevorzugt Kupfer(l)iodid.
Schritt (4) wird durchgeführt, indem die Verbindung (VIII) bei einer Temperatur von -78 0C bis 25 0C1 vorzugsweise -60 0C bis -20 0C, mit einer Metallorganyl- Verbindung (IX') umgesetzt wird,
in der M' gleich K, Na, Li, Ag, Cu, ZrCp2HaI, CeCI2, oder Ti[O(d-C6)alkyl]3 ist, vorzugsweise ZrCp2CI oder Ti(Z-Pr-O)3, und welche aus der Verbindung (IX) durch Deprotonierung, bevorzugt mit einem Metallorganyl (beispielsweise Butyllithium) und nachfolgende Umsetzung mit dem entsprechenden Metallreagenz, beispielsweise ZrCp2Cl2 oder bevorzugt CITi(Z-Pr-O)3 in einem inerten Lösungsmittel, vorzugsweise einem Ether, z.B. THF, und in einem Temperaturbereich von -30 0C bis 20 0C über einen Zeitraum von 2-60 Minuten erhältlich ist.
In Schritt (5) wird nach dem Fachmann an sich bekannten Methoden die Verbindung (X) homogen-katalytisch oder heterogen-katalytisch hydriert, wobei die Verbindung (X) oder die desilylierte Verbindung (IX) vide infra unter einer Wasserstoffgas- Atmosphäre bei Drücken von 1-50 bar bei einer Reaktionszeit von 15 Minuten bis 72 h und einem Temperaturbereich von 15-80 0C in einem geeigneten Lösungsmittel in Gegenwart eines vorzugsweise in substöchiometrischen Mengen vorliegenden Katalysators, vorzugsweise eines fein verteilten Übergangsmetalls, beispielsweise Pd, oder eines Übergangsmetall-Komplexes, beispielsweise Wilkinson-Katalysator, umgesetzt wird. Geeignete Lösungsmittel sind vorzugsweise polare, aprotische Lösungsmittel, beispielsweise Essigsäureethylester.
Schritt (5) beinhaltet ferner die Abspaltung der terminalen [(C-|-C6)Alkyl]3Si-Gruppe unter Freilegung der terminalen Alkohol-Funktion der Verbindung (Xl) nach dem Fachmann an sich bekannten Methoden, wobei die Verbindung (X) oder die hydrierte Verbindung (IX) beispielsweise mit einem fluorhaltigen Reagenz, beispielsweise TBAF oder HF-Pyridin in einem geeigneten Lösungsmittel, beispielsweise einem Gemisch von Wasser und THF über einen Zeitraum von 30 Minuten bis 4 d und einer Reaktionstemperatur von -40 °C und 70 0C umgesetzt wird.
Die Reihenfolge der Teilschritte in Schritt (5) ist beliebig.
In Schritt (6) wird nach dem Fachmann an sich bekannten Methoden die Verbindung (Xl) zu einer Verbindung (XII) oxidiert, beispielsweise lässt sich diese Reaktion unter den typischen Bedingungen einer „Jones-Oxidation" (Cr(VI )-vermittelt) erreichen, wobei zu der Verbindung (Xl) in einem geeigneten Lösungsmittel, vorzugsweise Aceton im Gemisch mit Wasser, in einem Temperaturbereich von -40 0C bis 60 0C, Jones-Reagenz (Chromtrioxid/ Schwefelsäure) gegeben und die Mischung für einen Zeitraum von 10 Minuten bis 24 h gerührt wird. Des weiteren können als Oxidationsmethoden die Umsetzung mit Kaliumpermanganat, optional katalysiert durch eine Lewis-Säure (Lai & Lee, Tetrahedron 2002, 58, 9879), Behandlung mit einem Ru^lO-salz/Metallperiodat-Gemisch oder eine Behandlung mit einem RuCI3/ 'BuOOH-Gemisch verwendet werden.
In Schritt (7) wird die Verbindung (Xl) oder die Verbindung (XII) in eine Verbindung der Formel (I) umgesetzt, beispielsweise durch Umsetzung mit Kaliumpermanganat, optional katalysiert durch eine Lewis-Säure, oder Oxidation mittels eines trifluorierten 1 ,1-Dimethyldioxirans, oder durch die Anwendung von molekularem Sauerstoff in Gegenwart eines Katalysators, beispielsweise von Aktivkohle (Kawabata & Hayashi, Tetrahedron Lett. 2004, 58, 5457), oder bevorzugt durch Behandlung mit einem Ru(lll)-salz/Metallperiodat-Gemisch, Behandlung mit einem RuCI3/ 4BuOOH- Gemisch oder Oxidation mittels TEMPO, oder besonders bevorzugt mittels Br2 in einem Gemisch von CCI4 und Wasser unter Bestrahlung mit Licht, beispielsweise mittels einer Wolfram-Lampe bei einer Temperatur von 0-70 0C nach Krohn et al. (Tetrahedron 2006, 62, 1223-1230).
In Schritt (8) wird die in Schritt (7) erhaltene Verbindung unter dem Fachmann an sich bekannten Bedingungen optional entweder durch Umsetzen mit einer geeigneten Base, beispielsweise NaOH oder KOH, zu einem Salz umgesetzt, oder die intermediär entstandene Säure-Funktion wird nach dem Fachmann an sich bekannten Methoden verestert oder amidiert, indem nach Aktivierung der Säurefunktion, beispielsweise mit Thionylchlorid, mit dem entsprechenden Nukleophil (NH2(C1 -CßJAlkyl, NH2-Benzyl, NH[(C-|-C6)Alkyl]2, NH[(C-|-C6)Alkyl][benzyl], [(C<|-C6)Alkyl]-SH, Benzyl-SH) umgesetzt wird. Die vorliegende Erfindung betrifft ferner Zwischenprodukte in dem oben beschriebenen Verfahren zur Herstellung von Mumbaistatin-Derivaten der Formel (I), insbesondere die Zwischenprodukte der Formeln (IV), (V), (VII), (VII1), (VII"), (VII1"), (VIII), (IX), (X), (Xl) und (XII), wobei solche Verbindungen der Formeln (VII), (VII1), (VII"), (VII1"), (VIII), (IX), (X), (Xl) und (XII) bevorzugt sind, die den R6-Substituenten ortho zur Benzophenon-Methylenbrücke tragen. Diese Verbindungen werden im Folgenden als (VII-A), (Vll'-A), (Vll"-A), (Vir'-A), (VIII-A), (IX-A), (X-A), (Xl-A) und (XII-A) bezeichnet, z.B. hat die Verbindung (XII-A) die Formel
Besonders bevorzugt ist eine Verbindung der Formel (IV), wobei X2R2 OH oder O-(C-|-C6)Alkyl ist, R4 OH oder O-(C-|-C6)Alkyl ist, und n und q unabhängig voneinander O oder 1 sind. Besonders bevorzugt ist eine Verbindung (IV)1 in der X2R2 OH oder O-(C-| -CO)AI kyl ist, R4 OH oder O-(Ci-Ce)Alkyl ist, n gleich O oder 1 ist, und q gleich 1 ist.
Besonders bevorzugt ist eine Verbindung der Formel (V), wobei X2 O ist, R2 H oder (C-|-C6)Alkyl ist, R4 und R5 unabhängig voneinander OH oder O-(C<|-C6)Alkyl sind, n und q unabhängig voneinander O oder 1 sind, und HaI Chlor oder Brom bedeutet. Besonders bevorzugt ist eine Verbindung (V), in der X2 O ist, R2 H oder (C-|-C6)Alkyl ist, R4 und R5 O-(C-|-C6)Alkyl sind, n gleich O oder 1 ist, q gleich 1 ist, und HaI Brom bedeutet. Besonders bevorzugt ist eine Verbindung der Formel (VII) oder (VII-A), wobei X2 O ist, R2 H oder (C-i-CßJAlkyl ist, R4, R5 und R6 unabhängig voneinander OH oder
O-(C-|-C6)Alkyl sind, und n, q und r unabhängig voneinander 0 oder 1 sind.
Besonders bevorzugt ist eine Verbindung (VII), in der X2 O ist, R2 H oder (C<|-C6)Alkyl ist, R4, R5 und R6 O-(C-|-C6)Alkyl sind, n gleich 0 oder 1 ist, und q und r gleich 1 sind.
Besonders bevorzugt ist eine Verbindung der Formel (VIT) oder (Vll'-A), wobei X2R2 OH oder O-(C-|-Cö)Alkyl ist, und ferner R4, R5 und R6 unabhängig voneinander OH oder O-(C-|-C6)Alkyl sind, und n, q und r unabhängig voneinander 0 oder 1 sind. Speziell bevorzugt ist X2R2 OH, ferner R4, R5 und R6 gleich O-(Ci-C6)Alkyl, n gleich O oder 1 ist, und q und r gleich 1. Z ist bevorzugt O-(CH2-CH2)-O.
Besonders bevorzugt ist eine Verbindung der Formel (VN") oder (Vll"-A), wobei R4, R5 und R6 unabhängig voneinander OH oder O-(C-| -CO)AI kyl sind, HaI Chlor, Brom oder lod ist, und n, q und r unabhängig voneinander O oder 1 sind. Speziell bevorzugt ist R4, R5 und R6 O-(C<|-C6)Alkyl, HaI Chlor oder Brom, n gleich O oder 1 , und q und r gleich 1.
Besonders bevorzugt ist eine Verbindung der Formel (VN'") oder (VH'"-A), wobei R4, R5 und R6 unabhängig voneinander OH oder O-(C-|-C6)Alkyl sind, und n, q und r unabhängig voneinander O oder 1 sind. Speziell bevorzugt sind R4, R5 und R6 O-(C-i-C6)Alkyl, n gleich O oder 1 , und q und r gleich 1.
Besonders bevorzugt ist ferner eine Verbindung (VIII) oder (VIII-A), wobei X2 O ist, R2 H oder (C-|-C6)Alkyl ist, R4, R5 und R6 unabhängig voneinander OH oder
O-(C-]-C6)Alkyl sind, und n, q und r unabhängig voneinander O oder 1 sind.
Besonders bevorzugt ist eine Verbindung (VIII), in der X2 O ist, R2 H oder (C-i-CßJAlkyl ist, R4, R5 und R6 O-(C<|-C6)Alkyl sind, n gleich O oder 1 ist, und q und r gleich 1 sind. Die Verbindung der Formel (IX) kann racemisch oder als Enantiomer eingesetzt werden, beispielsweise als (R)- oder als (S)-Form.
Besonders bevorzugt ist eine Verbindung der Formel (Xl) oder (Xl)1 in der X2R2 OH oder O-(C<|-C6)Alkyl ist, R4, R5 und R6 unabhängig voneinander OH oder O-(C-|-C6)Alkyl sind, und n, q und r unabhängig voneinander 0 oder 1 sind.
Besonders bevorzugt ist eine Verbindung der Formel (XII) oder (XII-A), wobei X2R2 OH oder O-(C-|-C6)Alkyl ist, R4, R5 und R6 unabhängig voneinander OH oder O-(C-|-C6)Alkyl sind, und n, q und r unabhängig voneinander O oder 1 sind.
Über das vorliegende Verfahren sind Mumbaistatin-Derivate ausgehend von der Verbindung (II) in einer 7-8-stufigen Synthese zugänglich. Der beschriebene Syntheseweg zeichnet sich durch eine verbesserte Effizienz und Ausbeute im Aufbau des hochsubstituierten Anthrachinon-Bausteins der Verbindung der Formel (I) über eine Diels-Alder-Cycloaddition ohne die Notwendigkeit von aufwendigen Schutzgruppenmanipulationen aus. Desweiteren bietet die Anwendung einer Übergangsmetall-katalysierten Reaktion erstmalig einen Zugang zu tetra-ortho- substituierten Benzophenonen in Kombination mit einem Anthrachinon-Gerüst in den Mumbaistatin-Derivaten (I) und deren Intermediaten.

Die oben genannten Reaktionsschritte und die im Folgenden beschriebenen Reaktionen werden an Luft oder unter Schutzgas, vorzugsweise unter Schutzgas durchgeführt.
Beispiel 1 - Herstellung von 3-Trimethylsilanyloxy-2-(1-trimethylsilanyloxy-vinyl)-but- 2-ensäure-tert.-butylester:
34.2 ml (0.21 mol) 3-Oxo-butansäure-tert.-butylester wurden in 300 ml Dichlormethan gelöst und mit 20 g (0.21 mol, 1.0 eq.) wasserfreiem
Magnesiumchlorid versetzt. Zu der lebhaft gerührten Suspension wurden bei 0 0C 33.9 ml (0.42 mol, 2.0 eq.) trockenes Pyridin zugesetzt. Nach 15 Minuten wurden tropfenweise 14.3 ml (0.21 mol, 1.0 eq.) Acetylchlorid zugegeben und die Mischung 1 Stunde bei RT gerührt. Überschüssiges Pyridin wurde bei O 0C mit verdünnter Salzsäure (37 ml konz. Salzsäure in 100 ml Wasser) neutralisiert und die organische Phase abgetrennt. Die wässrige Phase wurde mit MTBE (3 x 100 ml) extrahiert, die vereinigten organischen Phasen mit gesättigter NaCI-Lösung gewaschen und über MgSO4 getrocknet. Nachdem das Lösungsmittel entfernt worden war, wurden 41.2 g (0.206 mol, 98 %) 2-Acetyl-3-oxo-butansäure-tert-butylester in Form eines blassgelben Öls erhalten.
40.2 g (0.20 mol) des so erhaltenen 2-Acetyl-3-oxo-butansäure-tert-butylester wurden ohne weitere Aufreinigung in 50 ml Toluol mit 107.0 ml (0.44 mol, 2.2 eq.) BSA versetzt und 2 d bei RT gerührt. Nachdem das Lösungsmittel am Rotationsverdampfer entfernt worden war, wurde das Produkt einer Vakuumdestillation unterworfen (0.36 mbar, 75 0C), wobei 60.6 g (0.176 mol, 88 %) des gewünschten 3-Trimethylsilanyloxy-2-(1 -trimethylsilanyloxy-vinyl)-but-2-ensäure- tert-butylester als gelbliches Öl erhalten wurde. In einem weiteren Experiment wurden Ausbeuten von 97 bzw. 84 % erhalten.

Beispiel 2 - Herstellung von 3-Hydroxy-1-methyl-9,10-dioxo-9,10-dihydro-anthracen- 2-carbonsäure-tert-butylester:
27.9 g (81.2 mmol, 2.0 eq.) 3-Trimethylsilanyloxy-2-(1-trimethylsilanyloxy-vinyl)-but- 2-ensäure-tert-butylester (Beispiel 1 ) und 7.8 g (40.6 mmol) 2-Chlor-1 ,4- naphthochinon (Gupta & Franck, Synlett 1990, 355-357) wurden in 325 ml wasserfreiem Toluol gelöst und 24 Stunden zum Rückfluß erhitzt. Danach wurde das Lösungsmittel am Rotationsverdampfer entfernt und der braune, ölige Rückstand mit 250 ml THF versetzt. Nachdem 10 ml Wasser zugesetzt worden waren wurde 1 d bei RT gerührt. Dann wurden ca. 50 ml Kieselgel zugesetzt, wobei sich das Gemisch erwärmte, und anschließend das Lösungsmittel entfernt. Das auf das Kieselgel aufgezogene Rohprodukt wurde nun in eine mit Kieselgel gefüllte Fritte gefüllt und mit DCM als Laufmittel eluiert. Die gelbe Produkt-Bande wurde separat aufgefangen, und nach Entfernen des Lösungsmittels wurden 7.7 g (22.8 mmol, 55 %) 3-Hydroxy- 1-methyl-9,10-dioxo-9,10-dihydro-anthracen-2-carbonsäure-tert-butylester erhalten, der noch geringe Mengen an Verunreinigungen aufwies, welche jedoch auf der nächsten Synthesestufe vollständig entfernt werden konnten. In einem weiteren Experiment wurde eine Ausbeute von 48 % erhalten.

Beispiel 3 - Herstellung von 3,8-Dihydroxy-1-methyl-9,10-dioxo-9,10-dihydro- anthracen-2-carbonsäure-tert-butylester:
20.1 g (58.5 mmol, 2.0 eq.) 3-Trimethylsilanyloxy-2-(1-trimethylsilanyloxy-vinyl)-but- 2-ensäure-tert-butylester und 5.05 g (24.3 mmol) 2-Chlor-8-hydroxy-1 ,4- naphthochinon (Giles & Roos, J. Chem. Soc Perkin I 1976, 2057-2060) wurden in 125 ml wasserfreiem Toluol gelöst und 24 Stunden zum Rückfluss erhitzt. Danach wurde das Lösungsmittel am Rotationsverdampfer entfernt und der braune, ölige Rückstand mit 150 ml THF versetzt. Nachdem 10 ml Wasser zugesetzt worden waren wurde 1 d bei RT gerührt. Dann wurden ca. 50 ml Kieselgel zugesetzt und das Gemisch vom Lösungsmittel befreit. Das auf das Kieselgel aufgezogene Rohprodukt wurde nun in eine mit Kieselgel gefüllte Fritte gefüllt und mit DCM als Laufmittel eluiert. Die einfach zu isolierende stark gelbgefärbte Produkt-Bande wurde separat aufgefangen, und nach Entfernen des Lösungsmittels konnten 3.8 g (10.9 mmol, 45 %) des braunen 3,8-Dihydroxy-1-methyl-9,10-dioxo-9,10-dihydro- anthracen-2-carbonsäure-tert-butylesters isoliert werden, welcher noch geringe Mengen an Verunreinigungen enthielt, die aber auf der nächsten Synthesestufe vollständig entfernt werden konnten.

Beispiel 4 - Herstellung von 3-Methoxy-1-methyl-9,10-dioxo-9,10-dihydro-anthracen- 2-carbonsäure-tert.-butylester:
3.5 g (10.4 mmol) 3-Hydroxy-1-methyl-9,10-dioxo-9,10-dihydro-anthracen-2- carbonsäure-tert-butylester wurden in 70 ml Aceton mit 2.88 g (20.8 mmol, 2.0 eq.) wasserfreiem K2CO3 und 5.2 ml (83.2 mmol, 8.0 eq.) Methyl iodid versetzt und 3 Stunden zum Rückfluß erhitzt. Nach Abkühlung wurde das Aceton am Rotationsverdampfer abdestilliert. Das verbleibende Gemisch wurde in DCM aufgenommen und gegen Wasser extrahiert. Nach Trocknen über MgSO4, Waschen mit gesättigter NaCI-Lösung und Entfernen des Lösungsmittels wurden 3.49 g (9.9 mmol, 95 %) des methylierten Produktes 3-Methoxy-1-methyl-9,10-dioxo-9,10- dihydro-anthracen-2-carbonsäure-tert.-butylester in analytisch reiner Form als bräunlicher Feststoff erhalten. In einem weiteren Experiment wurde eine Ausbeute von 92 % erhalten.

Beispiel 5 - Herstellung von 3,8-Dimethoxy-1-methyl-9,10-dioxo-9,10-dihydro- anthracen-2-carbonsäure-tert.-butylester:
1.87 g (5.28 mmol) 3,8-Dihydroxy-1-methyl-9,10-dioxo-9,10-dihydro-anthracen-2- carbonsäure-tert.-butylester wurden in 40 ml Aceton mit 2.92 g (20.8 mmol, 4.0 eq.) wasserfreiem K2CO3 und 5.26 ml (84.5 mmol, 16.0 eq.) Methyliodid versetzt und 8 Stunden zum Rückfluß erhitzt. Nach Abkühlung wurde das Aceton am Rotationsverdampfer abdestilliert. Das verbleibende Gemisch wurde in DCM aufgenommen und gegen Wasser extrahiert. Nach Waschen mit gesättigter NaCI- Lösung und Trocknen über MgSO4 wurde über einen Silica-Plug filtriert und das Lösungsmittel entfernt. Es wurden 1.92 g ( 5.02 mmol, 95 %) des methylierten Produktes 3,8-Dimethoxy-1-methyl-9,10-dioxo-9,10-dihydro-anthracen-2- carbonsäure-tert.-butylester in analytisch reiner Form als ockerfarbener Feststoff erhalten.

Beispiel 6 - Herstellung von 1-Brommethyl-3-methoxy-9,10-dioxo-9,10-dihydro- anthracen-2-carbonsäure-tert.-butylester:
Es wurden 3.49 g (9.9 mmol) 3-Methoxy-1-methyl-9,10-dioxo-9,10-dihydro- anthracen-2-carbonsäure-tert.-butylester in 370 ml trockenem und entgastem CCU gelöst und mit 2.29 g (12.9 mmol, 1.3 eq.) NBS versetzt. Die Lösung wurde zum Rückfluss erhitzt und unter Bestrahlung mit Licht (150 W Lampe, hoher UV-Anteil) 2 x 8 Stunden lang bei dieser Temperatur gehalten. Dabei wurden in 5 Portionen alle 3 Stunden je 72 mg (0.297 mmol, 3 mol%) Dibenzoylperoxid (gelöst in je 1 ml CCI4) als Radikalstarter zugesetzt. Nachdem das Reaktionsgemisch abgekühlt war, konnte das unlösliche Succinimid abfiltriert und das Lösungsmittel entfernt werden. Es konnten 4.23 g (9.8 mmol, 99 %) des benzylisch bromierten Anthrachinons 1- Brommethyl-S-methoxy-Θ.IO-dioxo-Θ.IO-dihydro-anthracen^-carbonsäure-tert.- butylester analytisch rein in Form eines gelblich-braunen Feststoffes erhalten werden. In einem weiteren Experiment wurde eine Ausbeute von 92 % erhalten.

Beispiel 7 - Herstellung von 1-Brommethyl-3,8-dimethoxy-9,10-dioxo-9,10-dihydro- anthracen-2-carbonsäure-tert.-butylester:
Es wurden 1.9 g (4.97 mmol) 3,8-Dimethoxy-1-methyl-9,10-dioxo-9,10-dihydro- anthracen-2-carbonsäure-tert.-butylester in 110 ml trockenem und entgastem CCI4 gelöst und mit 1.15 g (6.46 mmol, 1.3 eq.) NBS versetzt. Die Lösung wurde zum Rückfluss erhitzt und unter Bestrahlung mit Licht (150 W Lampe, hoher UV-Anteil) zweimal 8 Stunden lang bei dieser Temperatur gehalten. Dabei wurden in 5
Portionen alle 3 Stunden je 36 mg (0.15 mmol, 3 mol%) Dibenzoylperoxid (gelöst in je 1 ml CCI4) als Radikalstarter zugesetzt. Nachdem das Reaktionsgemisch abgekühlt war, konnte das unlösliche Succinimid abfiltriert und das Lösungsmittel entfernt werden. Das Reaktionsgemisch wurde säulenchromatographisch aufgereinigt. Es konnten 1.54 g (3.33 mmol, 67 %) des benzylisch bromierten
Anthrachinons 1-Brommethyl-3,8-dimethoxy-9,10-dioxo-9,10-dihydro-anthracen-2- carbonsäure-tert.-butylester in Form eines rötlich-gelben Feststoffes erhalten werden.

Beispiel 8 - Herstellung von 3-Methoxy-2-(thbutylstannyl)-benzylalkohol:
OMe 2.00 g (14.5 mmol) 3-Methoxybenzylalkohol wurden in 40 ml trockenem Benzol gelöst. Bei 0 0C wurden 19.91 ml (31.9 mmol, 2.2 eq.) einer 1.6 molar n-BuLi-Lösung langsam zugetropft, wobei sich die nach Zugabe eines Äquivalents n-BuLi gebildete weisse Suspension bei Zugabe des zweiten Äquivalents dunkelrot färbte. Es wurde noch 2 Stunden bei RT weitergerührt und dann bei 0 0C mit 5.1 ml (18.8 mmol, 1.3 eq.) Tributylzinnchlorid versetzt. Nachdem über Nacht bei RT gerührt worden war, wurden 40 ml gesättigte NH4CI-Lösung zugesetzt und mit dreimal 70 ml EE extrahiert. Nach Waschen der organischen Phase mit gesättigter NaCI-Lösung und Trocknen über MgSO4 wurde das Lösungsmittel entfernt. Das Rohprodukt wurde säulenchromato-graphisch gereinigt (reversed-phase silica RP-18, MeCN) und man erhielt 4.83 g (11.3 mmol, 78 %) 3-Methoxy-2-(tributylstannyl)-benzylalkohol als farbloses Öl. In einem weiteren Experiment wurde eine Ausbeute von 60 % erhalten.

Beispiel 9 - Herstellung von 1-(2'-Hydroxymethyl-6'-methoxy-benzyl)-3-methoxy- 9,10-dioxo-9,10-dihydro-anthracen-2-carbonsäure-tert.-butylester:
100 mg (232 μmol) 1-Brommethyl-3-methoxy-9,10-dioxo-9,10-dihydro-anthracen-2- carbonsäure-tert.-butylester wurden in einem Schlenkrohr mit Rückflußkühler in einer Mischung aus 2.0 ml THF und 2.0 ml NMP gelöst. Danach wurden 3.5 mg (20 μmol, 8 mol%) Kupfer(l)iodid, 11.6 mg (37 μmol, 16 mol%) Triphenylarsin und 8.9 mg (9 μmol, 4 mol%) Pd2(dba)3 unter Rühren zugesetzt. Die braun-rote Lösung wurde zweimal am Vakuum entgast und mit Argon gesättigt. Dann wurden 119 mg (278 μmol, 1.2 eq.) 3-Methoxy-2-(tributylstannyl)-benzylalkohol durch den Hahn zugespritzt. Das Reaktionsgemisch wurde für 20 Stunden auf eine Temperatur von 80 0C erhitzt, wobei sich die Lösung langsam von braun nach schwarz verfärbte. Nach Abkühlung wurden 15 ml H2O zugesetzt und das Gemisch mit 3 x 50 ml EE extrahiert. Trocknen über MgSO4 und Entfernen des Lösungsmittels führte zu einem braunen, schleimigen Feststoff, der säulenchromatographisch (Cyclohexan/EE 2:1 ) gereinigt wurde. Es konnten 49.8 mg (100 μmol, 43 %) des Stille- Kupplungsproduktes 1 -(2'-Hydroxymethyl-6'-methoxy-benzyl)-3-methoxy-9, 10-dioxo- 9,10-dihydro-anthracen-2-carbonsäure-tert.-butylester in Form eines gelben Feststoffs erhalten werden. In einem weiteren Experiment wurde eine Ausbeute von 17 % erhalten.

Beispiel 10 - Herstellung von 1-(2'-Formyl-6'-methoxybenzyl)-3-methoxy-9,10-dioxo- 9,10-dihydro-anthracen-2-carbonsäure-tert.-butylester:
Es wurden 100 mg (204 μmol) 1-(2'-Hydroxymethyl-6'-methoxy-benzyl)-3-methoxy- 9,10-dioxo-9,10-dihydro-anthracen-2-carbonsäure-tert.-butylester in 6 ml DCM gelöst und auf -78 0C gekühlt. Dann wurden 290 μ\ (4.0 mmol, 20 eq.) DMSO (über 4A- Molsieb getrocknet) zugegeben und nach 5 Minuten 172 //I (2.0 mmol, 10 eq.) Oxalylchlorid. Nach weiteren 60 Minuten wurden 1.42 ml (10.2 mmol, 50 eq.) Triethylamin (wasserfrei) zugesetzt und das Reaktionsgemisch über Nacht langsam auf RT erwärmt. Nach Zugabe von 20 ml H2O wurden die Phasen getrennt und die wässrige Phase noch mit 3 x 30 ml EE extrahiert. Nach Waschen der organischen Phase mit gesättigter NaCI-Lösung und Trocknen über MgSO4 wurde das Lösungsmittel entfernt. Ohne weitere Aufreinigung konnten in reiner Form 97 mg (200 /ymol, 98 %) des freien Aldehyds 1-(2'-Formyl-6'-methoxy-benzyl)-3-methoxy- 9,10-dioxo-9,10-dihydro-anthracen-2-carbonsäure-tert.-butylester in Form eines leuchtend gelben Feststoffs erhalten werden.

Beispiel 11 - Herstellung von 5-(tert.-Butyldimethylsiloxy)-pent-1-in:
5.00 g (59.4 mmol) Pent-1-in-5-ol wurden in 50 ml trockenem DMF gelöst und mit 10.1O g (149 mmol, 2.5 eq.) Imidazol und 9.40 g (62.4 mmol, 1.1 eq.) TBS-CI versetzt. Nachdem 12 Stunden bei RT gerührt worden war, wurden 250 ml Wasser zugesetzt und mit 3 x 150 ml MTBE extrahiert. Die organische Phase wurde noch 3 x mit je 100 ml Wasser gewaschen und schließlich über MgSO4 getrocknet. Nach säulenchromatographischer Aufreinigung wurden 11.10 g (53.5 mmol, 90 %) 5-(tert.- Butyldimethylsiloxy)-pent-1-in als farbloses Öl erhalten.

Zu einer Lösung von 5-(tert.-Butyldimethylsiloxy)-pent-1-in (1.44 mmol) in THF (4 ml) wurden bei O 0C 0.8 ml n-BuLi (1.23 mmol, 1.6 molar in Hexan) gegeben. Nach 15 Minuten wurde auf -60 0C abgekühlt und 350 mg CITi(OZ-Pr)3 (1.31 mmol) zugesetzt. Nach 90 Minuten wurde eine Lösung von 200 mg 1-(2'-Formyl-6'- methoxybenzylJ-S-methoxy-Θ.IO-dioxo-Θ.IO-dihydro-anthracen^-carbonsäure-tert.- butylester (0.4 mmol) in 6 ml THF zugespritzt und weitere 30 Minuten gerührt. Nach Erwärmung auf —20 0C wurde 15 Stunden weitergerührt und schließlich mit gesättigter NH4CI-Lösung versetzt. Die wässrige Phase wurde 2 x mit CH2CI2 extrahiert und die vereinigten organischen Phasen über MgSO4 getrocknet. Nach säulenchromatographischer Aufarbeitung (CHCI3 / EE 95:5) konnte das Additionsprodukt 1 -{2'-[6"-(tert-Butyldimethylsilanyloxy)-1 "-hydroxy-hex-2"-inyl]-6'- methoxybenzylJ-S-methoxy-Θ.IO-dioxo-Θ.IO-dihydroanthracen^-carbonsäure-tert.- butylester in Form eines gelben Öls erhalten werden (260 mg, 0.38 mmol, 93 %).

Beispiel 13 - Herstellung von tert.-Butyl-(3-methoxymethoxy-pent-4-inyloxy)- dimethylsilan:
0.5 g 5-(tert.-Butyldimethylsilanyloxy)-pent-1-in-3-ol (2.33 mmol) wurden in 15 ml DCM gelöst, woraufhin 1.22 ml 1Pr2EtN (7.00 mmol) und 0.53 ml Chlormethyl- methylether (7.00 mmol) bei 25 0C zugegeben wurden. Nach 15 Stunden wurde die Raktion durch Zugabe von 10 ml Wasser abgebrochen und mit 10 ml DCM extrahiert. Nach Trocknung der organischen Phase, Entfernen des Lösungsmittels und säulenchromatographischer Aufreinigung konnten 0.45 g (1.75 mmol, 75%) des Produktes tert.-Butyl-(3-methoxymethoxy-pent-4-inyloxy)-dimethylsilan als farbloses Öl erhalten werden.

Beispiel 14 - Herstellung von 6"-(tert.-Butyldimethylsilanyloxy)-1 "-hydroxy-4"- methoxymethoxy-hex-2"-inyl]-6'-methoxybenzyl}-3-methoxy-9,10-dioxo-9,10- dihydroanthracen-2-carbonsäure-tert.-butylester
Entsprechend Beispiel 12 wurde 1-(2'-Formyl-6'-methoxybenzyl)-3-methoxy-9,10- dioxo-9,10-dihydroanthracen-2-carbonsäure-tert.-butylester mit tert.-Butyl-(3- methoxymethoxy-pent-4-inyloxy)-dimethylsilan umgesetzt. Nach säulenchromato- graphischer Aufarbeitung über Silicagel (CHCI3 / EE 95:5) konnte das Additionsprodukt 6"-(tert.-Butyldimethylsilanyloxy)-1 "-hydroxy-4"-methoxymethoxy-hex-2"- inyl]-6'-methoxybenzyl}-3-methoxy-9,10-dioxo-9,10-dihydroanthracen-2- carbonsäure-tert.-butylester in Form eines gelben Öls erhalten werden (230 mg, 0.31 mmol, 75 %).

Beispiel 15 - Herstellung von 1-{2'-[6"-(tert.-Butyldimethylsilanyloxy)-1 "- hydroxyhexyll-δ'-methoxybenzylJ-S-methoxy-Θ.IO-dioxo-Θ.I O-dihydro-anthracen^- carbonsäure-tert.-butylester:
Zu einer Lösung von 0.12 g (0.18 mmol) 1-{2'-[6"-(tert.-Butyldimethylsilanyloxy)-1- hydroxy-hex-2"-inyl]-6'-methoxybenzyl}-3-methoxy-9,10-dioxo-9,10- dihydroanthracen-2-carbonsäure-tert.-butylester (Beispiel 12) in 40 ml EE wurde Palladium auf Kohle (5 Gew.%) gegeben (Gewichtsverhältnis 1 :10 Katalysator/Substrat). Nachdem unter einer ^-Atmosphäre 2 Stunden lang gerührt worden war, wurde der Katalysator abfiltriert, das Lösungsmittel entfernt und das Produkt säulenchromatographisch aufgereinigt. Es konnten 0.11 g (0.16 mmol, 90%) des Produktes 1-{2'-[6"-(tert.-Butyldimethylsilanyloxy)-1 "-hydroxyhexyl]-6'-methoxy- benzylJ-S-methoxy-Θ.IO-dioxo-Θ.IO-dihydro-anthracen^-carbonsäure-tert.-butylester als gelbes Öl erhalten werden.

Beispiel 16 - Herstellung von 1-[2'-(5"-Carboxypentanoyl)-6'-methoxybenzyl]-3- methoxy-9,10-dioxo-9,10-dihydro-anthracen-2-carbonsäure-tert.-butylester via Jones-Oxidation:
Zu einer Lösung von 47 mg (0.07 mmol) 1-{2'-[6"-(tert.-Butyldimethylsilanyloxy)-1 "- hydroxyhexyl]-6'-methoxybenzyl}-3-methoxy-9,10-dioxo-9,10-dihydroanthracen-2- carbonsäure-tert.-butylester (Beispiel 15) in 4 ml Aceton wurden bei 0 °C tropfenweise 5 Äquivalente Jones-Reagenz in 5 Minuten zugetropft. Die Mischung wurde zunächst 1 Stunde bei 0 0C, dann 3 Stunden bei RT gerührt. Überschüssiges Oxidationsmittel wurde durch Zugabe von 2-Propanol zerstört. Nach Verdünnen mit 10 ml Wasser wurde mit 3 x 10 ml EE extrahiert. Die organischen Phasen wurden über Na2SO4 getrocknet und das Lösungsmittel entfernt. Nach säulenchromatographischer Aufreinigung konnten 21 mg (0.04 mmol, 52 %) des Produktes 1-[2"-(5"-Carboxypentanoyl)-6'-methoxybenzyl]-3-methoxy-9,10-dioxo- 9,10-dihydroanthracen-2-carbonsäure-tert.-butylester als gelbliches Öl erhalten werden.

Beispiel 17 - Herstellung von 1-{2'-[6"-(tert.-Butyldimethylsilanyloxy)-hexanoyl]-6'- methoxybenzyl}-3-methoxy-9 , 10-d ioxo-9 , 10-d i hyd roanthracen-2-carbonsäu re-tert. butylester via Swem-Oxidation:
Zu einer Lösung von 27 mg (0.21 mmol) Oxalylchlorid in 2 ml DCM wurden bei -78 0C 33 mg DMSO (0.42 mmol) gegeben. Nach 5 Minuten wurden 48 mg (0.07 mmol) 1 -{2'-[6"-(tert.-Butyldimethylsilanyloxy)-1 "-hydroxyhexyl]-6-methoxybenzyl}-3- methoxy-9,10-dioxo-9,10-dihydroanthracen-2-carbonsäure-tert.-butylester, gelöst in 3 ml DCM, zugesetzt. Nach weiteren 15 Minuten wurden schließlich 85 mg (0.84 mmol) NEt3 zugegeben, die Lösung auf RT erwärmt und 15 Stunden lang gerührt. Nach Zugabe von 10 ml gesättigter NH4CI-Lösung wurde die organische Phase abgetrennt, über Na2SO4 getrocknet, filtriert und das Lösungsmittel entfernt. Nach säulenchromatographischer Aufreinigung konnten 34 mg (0.05 mmol, 72 %) des Produktes 1-{2'-[6"-(tert.-Butyldimethylsilanyloxy)-hexanoyl]-6'-methoxybenzyl}-3- methoxy-9,10-dioxo-9,10-dihydroanthracen-2-carbonsäure-tert.-butylester als gelbes Öl erhalten werden.

Claims

Patentansprüche:
1. Verfahren zur Herstellung von Mumbaistatin-Derivaten der Formel (I) oder von Salzen davon,
wobei
R1 und R2 unabhängig voneinander ein H, (C-|-C6)Alkyl oder Benzyl sind, R3, R4 und R5 unabhängig voneinander OH, O-(C<|-C6)Alkyl, O-Benzyl oder
O-Acyl sind,
R6 OH, Halogen, (C<|-C6)Alkyl, O-(C<|-C6)Alkyl, Phenyl, Benzyl, O-Phenyl,
O-Benzyl, O-Acyl ist,
X1 und X2 unabhängig voneinander O, NH, N(C-|-C6)Alkyl oder S sind, und m, n, q und r unabhängig voneinander 0 oder 1 sind,
wobei in Schritt (1 ) eine Verbindung der Formel (II)
(H) in der Y eine Abgangsgruppe ausgewählt aus der Gruppe HaI, OTs, OTf oder OMs ist, und vorzugsweise Chlor oder Brom ist, mit einer Verbindung der Formel (III),
in der X eine Elektronen-schiebende Gruppe, beispielsweise [(Ci-C6)Alkyl]3silyloxy, vorzugsweise [Methyl]3silyloxy ist, und R' (CrCβJAlkyl oder [(CrC6)Alkyl]3silyl bedeutet, in einer [2+4]-Cycloaddition und anschließender Umsetzung mit einer geeigneten Säure zu einer Verbindung der Formel (IV) umgesetzt wird,
wobei R" = H oder (CrC6)Alkyl bedeutet,
in Schritt (2), sofern q = 1 und R" = H ist, die phenolische OH-Gruppe optional unter dem Fachmann an sich bekannten Bedingungen mit einer (C<|-C0)Alkylhalogen- oder Benzylhalogen-Verbindung verethert wird oder mit einer Acyl- Halogenverbindung verestert wird, und anschließend die Verbindung (IV) oder die so erhaltene veretherte oder acylierte Verbindung (IV) zu einer Verbindung der Formel (V) halogeniert wird,
in Schritt (3.1 ) die Verbindung (V) mit einer metallorganischen Verbindung (VI)
in der M Sn[(Ci-C6)Alkyl]3, B(OH)2, B(OR)2, BF3 ", ZnHaI oder MgHaI bedeutet, und (OR)2 gleich [O-(C-|-C6)Alkyl]2 oder ein 1 ,2-vicinaler Dialkohol, beispielsweise
Pinakol, Cathechol, unter Übergangsmetall-Katalyse, vorzugsweise unter Pd-, Ni- oder Fe-Katalyse zu einer Verbindung der Formel (VII) umgesetzt wird,
und anschließend die Verbindung (VII) zu einer Verbindung (VIII) oxidiert wird,
oder alternativ in einem Schritt (3.2) die Verbindung (V) mit einer metallorganischen Verbindung (Vl') wobei M und R6 wie für Verbindung (VI) definiert sind, und Z 0-(C-ι-C6)Alkylen-0, vorzugsweise O-(CH2-CH2)-O bedeutet, unter Übergangsmetall-Katalyse, vorzugsweise unter Pd-, Ni- oder Fe-Katalyse zu einer Verbindung der Formel (VIT) umgesetzt wird,
und anschließend die Verbindung (VIT) zu einer Verbindung (VIII) durch Umsetzung mit einer geeigneten Säure hydrolysiert wird,
oder alternativ in einem Schritt (3.3) die Verbindung (VI) mit einer Verbindung der Formel (V), in der X2R2 gleich OH ist, zu einer Verbindung der Formel (VH") verestert wird,
und anschließend die Verbindung (VM") unter Übergangsmetall-Katalyse, vorzugsweise unter Pd-, Ni- oder Fe-Katalyse zu einer Verbindung der Formel (VII'") umgesetzt wird,
und die Verbindung (VN'") zu einer Verbindung der Formel (VIII) umgesetzt wird, indem unter basischen oder unter sauren Bedingungen, vorzugsweise unter basischen Bedingungen hydrolysiert, anschließend der entstandene Benzylalkohol zum Aldehyd oxidiert und optional die entstandene Benzoesäure-Funktion verestert oder amidiert wird,
in Schritt (4) die Verbindung (VIII) mit einer Verbindung der Formel (IX) zu einer Verbindung der Formel (X) umgesetzt wird,
in Schritt (5) die Verbindung (X) unter Hydrierung der Dreifach-Bindung zu einer Einfachbindung und Abspaltung der [(C-|-C6)Alkyl]3Si-Gruppe in eine Verbindung der Formel (Xl) umgesetzt wird,
(Xi). in Schritt (6) die Verbindung (Xl) zu einer Verbindung der Formel (XII) oxidiert wird,
in Schritt (7) die Verbindung (Xl) oder die Verbindung (XII) zu einer Verbindung der Formel (I) oxidiert wird, in der X1 R1 = OH ist, und
in Schritt (8) optional die in Schritt (7) erhaltene Verbindung zu einer Verbindung der Formel (I) umgesetzt wird, in der X1 NH, N(C-|-C6)Alkyl oder S ist, und R1 gleich
(C-|-C6)Alkyl oder Benzyl ist, oder einem Salz davon.
2. Verfahren gemäß Anspruch 1 , wobei die Verbindung der Formel (I) durch eine Verbindung der Formel (I-A)
gekennzeichnet ist.
3. Verfahren gemäß Anspruch 2, wobei in der Verbindung (I-A) X1 R1 gleich OH oder O-(Ci-Ce)Alkyl ist,
X2R2 gleich OH ist,
R3 gleich OH ist,
R4 gleich OH oder O-(C<|-C6)Alkyl ist,
R5 gleich OH oder O-(C-|-C6)Alkyl ist,
R6 gleich OH oder O-(C-|-C6)Alkyl ist, und m und n unabhängig voneinander O oder 1 sind.
4. Verbindung der Formel (IV)
wobei R" = H oder (C-ι-C6)Alkyl bedeutet, und R2, R4, X2, n und q wie in Anspruch 1 definiert sind.
5. Verbindung der Formel (IV) gemäß Anspruch 4, wobei X2R2 OH oder O-(C<|-C6)Alkyl ist, R4 OH oder O-(C-|-C6)Alkyl ist, und n und q unabhängig voneinander O oder 1 sind.
6. Verbindung der Formel (IV) gemäß Anspruch 5, wobei X2R2 = OH oder O-(Ci-Ce)Alkyl ist, R4 = OH oder O-(C-|-C6)Alkyl, n = O oder 1 , und q = 1 ist.
7. Verbindung der Formel (V),
(V)1 wobei R2, R4, R5, X2, HaI, n und q wie in Anspruch 1 definiert sind.
8. Verbindung der Formel (V) gemäß Anspruch 7, wobei X2R2 OH oder O-(C-|-C6)Alkyl ist, R4 und R5 unabhängig voneinander OH oder O-(C-|-C6)Alkyl sind, n und q unabhängig voneinander O oder 1 sind, und HaI Chlor, Brom oder lod bedeutet.

12. Verbindung der Formel (VII) gemäß einem der Ansprüche 10 oder 11 , wobei X2R2 OH oder O-(C<|-C6)Alkyl ist, ferner R4, R5 und R6 unabhängig voneinander OH oder O-(C-|-C6)Alkyl sind, und n, q und r unabhängig voneinander 0 oder 1 sind.
13. Verbindung der Formel (VII) gemäß einem der Ansprüche 10 bis 12, wobei X2R2 OH oder O-(C-|-C6)Alkyl ist, ferner R4, R5 und R6 O-(C-|-C6)Alkyl sind, n gleich 0 oder 1 ist, und q und r gleich 1 sind.
14. Verbindung der Formel (VIT)
wobei Z O-(CrC6)Alkylen-O bedeutet, und R2, R4, R5, R6, X2, n, q und r wie in Anspruch 1 definiert sind.
15. Verbindung der Formel (VIT) gemäß Anspruch 14, gekennzeichnet durch eine Verbindung der Formel (Vll'-A)
16. Verbindung der Formel (VIT) gemäß einem der Ansprüche 14 oder 15, wobei X2R2 OH oder O-(C-|-C6)Alkyl ist, ferner R4, R5 und R6 unabhängig voneinander
OH oder O-(C-|-C6)Alkyl sind, und n, q und r unabhängig voneinander O oder 1 sind.
17. Verbindung der Formel (VIT) gemäß einem der Ansprüche 14 bis 16, wobei X2R2 OH ist, ferner R4, R5 und R6 O-(C<|-C6)Alkyl sind, n gleich O oder 1 ist, und q und r gleich 1 sind.
18. Verbindung der Formel (VM') gemäß einem der Ansprüche 14 bis 17, wobei Z O-(CH2-CH2)-O bedeutet.
19. Verbindung der Formel (VII") wobei HaI, R4, R5, R6, n, q und r wie in Anspruch 1 definiert sind.
20. Verbindung der Formel (VII") gemäß Anspruch 19, gekennzeichnet durch eine Verbindung der Formel (Vll"-A)
21. Verbindung der Formel (VH") gemäß einem der Ansprüche 19 oder 20, wobei R4, R5 und R6 unabhängig voneinander OH oder O-(C-i-Cß)Alkyl sind, HaI Chlor, Brom oder lod ist, und n, q und r unabhängig voneinander 0 oder 1 sind.
22. Verbindung der Formel (VM") gemäß einem der Ansprüche 19 bis 21 , wobei R4, R5 und R6 O-(C-|-C6)Alkyl sind, HaI Chlor oder Brom ist, n gleich 0 oder 1 ist, und q und r gleich 1 sind.
23. Verbindung der Formel (VII'") wobei R4, R5, R6, n, q und r wie in Anspruch 1 definiert sind.
24. Verbindung der Formel (VM'") gemäß Anspruch 23, gekennzeichnet durch eine Verbindung der Formel (Vll'"-A)
25. Verbindung der Formel (VII'") gemäß einem der Ansprüche 23 oder 24, wobei R4, R5 und R6 unabhängig voneinander OH oder O-(C-| -Ce)AI kyl sind, und n, q und r unabhängig voneinander 0 oder 1 sind.
26. Verbindung der Formel (VH'") gemäß einem der Ansprüche 23 bis 25, wobei R4, R5 und R6 O-(C-|-C6)Alkyl sind, n gleich 0 oder 1 ist, und q und r gleich 1 sind.
27. Verbindung der Formel (VIII), wobei R2, R4, R5, R6, X2, n, q und r wie in Anspruch 1 definiert sind.
28. Verbindung der Formel (VIII) gemäß Anspruch 27, gekennzeichnet durch eine Verbindung der Formel (VIII-A)
29. Verbindung der Formel (VIII) gemäß einem der Ansprüche 27 oder 28, wobei X2R2 OH oder O-(C-|-Ce)Alkyl ist, R4, R5 und R6 unabhängig voneinander OH oder O-(C-|-C6)Alkyl sind, und n, q und r unabhängig voneinander O oder 1 sind.
30. Verbindung der Formel (VIII) gemäß einem der Ansprüche 27 bis 29, wobei X2R2 OH oder O-(C-|-C6)Alkyl ist, und R4, R5 und R6 O-(C-|-C6)Alkyl sind, n gleich
O oder 1 ist, und q und r gleich 1 sind.
31. Verbindung der Formel (Xl) wobei R2, R3, R4, R5, R6, X2, m, n, q und r wie in Anspruch 1 definiert sind.
32. Verbindung der Formel (Xl) gemäß Anspruch 31 , gekennzeichnet durch eine Verbindung der Formel (Xl-A)
33. Verbindung der Formel (Xl) gemäß einem der Ansprüche 31 oder 32, wobei X2R2 OH oder O-(C<| -Cg)AIkVl ist, R4, R5 und R6 unabhängig voneinander OH oder O-(C-|-C6)Alkyl sind, und n, q und r unabhängig voneinander O oder 1 sind.
34. Verbindung der Formel (XII) wobei R2, R3, R4, R5, R6, X2, m, n, q und r wie in Anspruch 1 definiert sind.
35. Verbindung der Formel (XII) gemäß Anspruch 34, gekennzeichnet durch eine Verbindung der Formel (XII-A)
