La camptothécine (CPT), alcaloïde isolé de Camptothéca accuminata, est un agent anticancéreux possédant un large spectre d'activité. Un autre problème provient du manque de stabilité du cycle E. En effet, la fonction lactone du cycle E, est en équilibre dans les milieux physiologiques avec sa forme ouverte hydroxy-acide. Cette dernière est inactive et semble avoir une toxicité particulière intrinsèque [Cancer Research., 49, 1465 (1989); ibid, 49, 5077 (1989)]. Des tentatives de modification de ce cycle, afin de le rendre plus stable, ont été menées, en particulier l'atome d'oxygène cyclique a été remplacé par un atome d'azote ou de soufre, mais dans chaque cas il y a perte de l'activité pharmacologique confirmant ainsi l'importance de la lactone [Journal of Médicinal Chemistry, 32, 715 (1989)]. D'autres modifications structurales du cycle E de la CPT ont été décrites par la suite, en particulier dans le brevet EP 1101765.
