Abstract
Die Erfindung betrifft ein neues Verfahren zur Herstellung von (2E)-2-(Hydroxyphenyl)-2-(alkoxyimino)-N-methylacetamiden.
Description
Verfahren zur Herstellung von 2-(HydroxyphenvD-2-(alkoxyimino)- N-methylacetamid-Derivaten
Die Erfindung betrifft Verfahren zur Herstellung von 2-(Hydroxyphenyl)-2-
(alkoxyimino)-N-methylacetamid-Derivaten, neue Zwischenprodukte zu deren Herstellung, sowie Verfahren zur Herstellung dieser Zwischenprodukte.
Es ist bereits bekannt geworden, dass man Dichlorbenzofuranon-3 durch Chlorierung von Benzofuranon-3 erhält (vergleiche z.B. Chem. Ber. 1912, S. 161). Die aus dem
Stand der Technik bekannte Umsetzung wird im geschlossenen Gefäß durchgeführt und ist daher im großtechnischen Maßstab nicht durchführbar. Dichlorbenzofuranon- 3 wird außerdem als Nebenprodukt bei der Herstellung von 2-Mefhoxybenzoylchlo- rid erhalten (vgl. DE-A-2040186). Es ist bisher jedoch kein im großtechnischen Maßstab anwendbares Verfahren zur gezielten Herstellung von Dichlorbenzofura- non-3 bekannt. Auch die Ringöffnung von Dichlorbenzofuranon-3 ist nicht literaturbekannt.
Es ist bereits bekannt geworden, dass man 2-(2-Hydroxyphenyl)-2-(methoxyimino)- N-methylacetamid (C) erhält, wenn man 2-(2-Hydroxyphenyl)-2-(methoxyimino)- essigsäuremethylester (A) oder l-Benzofüran-2,3-dion-3-(O-methyloxim) (B), mit Methylamin umsetzt (vergleiche z. B. EP-A 398 692 oder WO 95/24396):
Der Nachteil dieser Verfahren liegt in der schlechten Zugänglichkeit der Ausgangsstoffe.
In WO 01/38294 werden zwei Verfahren zur Herstellung von 2-(2-Hydroxyphenyl)- 2-(alkoxyimino)-N-methylacetamid-Derivaten ausgehend von 2-(2-Hydroxyphenyl)- 2-(hydroxyimino)-N-methylacetamid bzw. ausgehend von 2-(2-Hydroxyphenyl)-2- (methylimino)-N-methylacetamid beschrieben.
Es wurde nun gefunden, dass man 2-(Hydroxyphenyl)-2-(alkoxyimino)-N-methyl- acetamid-Derivate der allgemeinen Formel (I)
in welcher
R1 und R2 für C i -C4-Alkyl stehen,
dadurch erhält, dass man
a) eine Verbindung der Formel (II)
mit Chlorgas gegebenenfalls in Gegenwart eines Verdünnungsmittels umsetzt;
b) die dabei entstehende Verbindung der Formel (III)
mit einem Amin der Formel (IV),
R'-NH2 (IV)
in welcher
R1 die oben angegebene Bedeutung hat,
- oder einem Säureadditionskomplex davon
gegebenenfalls in Gegenwart eines Verdünnungsmittels und gegebenenfalls in Gegenwart eines Säureakzeptors, zu einer Verbindung der Formel (V) umsetzt;
diese
cl) mit Hydroxylamin - oder einem Säureadditionskomplex davon - gegebenenfalls in Gegenwart eines Verdünnungsmittels und gegebenenfalls in Gegenwart eines Puffermediums zu Verbindungen der Formel (VI) umsetzt;
und
c2) mit einem Alkylierungsmittel der Formel (VII),
R2-X (VII) in welcher
R die oben angegebene Bedeutung hat, und
X für Halogen, -O-CO-O-R2, -O-SO2-R2, oder -O-SO2-O-R2 steht, wobei R die oben angegebene Bedeutung hat,
gegebenenfalls in Gegenwart eines Verdünnungsmittels und gegebenenfalls in Gegenwart eines Säureakzeptors, gegebenenfalls in Gegenwart eines Phasen- transferkatalysators, umsetzt;
oder
d) mit Alkoxyaminen der Formel (VIII),
R2-O-NH2 (VIII) in welcher
R die oben angegebene Bedeutung hat,
oder einem Säureadditionskomplex davon -
gegebenenfalls in Gegenwart eines Verdünnungsmittels, und gegebenenfalls in Gegenwart eines Puffermediums, umsetzt.
In der Definition von R
stoffketten jeweils geradkettig oder verzweigt. Insbesondere steht Cι-C4-Alkyl für Methyl, Ethyl, n- oder i-Propyl, n-, s- oder i- Butyl.
R1 steht insbesondere für Methyl, Ethyl oder n-Propyl.
R1 steht besonders bevorzugt für Methyl.
R2 steht insbesondere für Methyl, Ethyl oder n-Propyl.
R2 steht besonders bevorzugt für Methyl.
Die oben aufgeführten allgemein oder in Vorzugsbereichen angegebenen Restedefinitionen gelten sowohl für die Endprodukte der Formel (I) als auch entsprechend für die jeweils zur Herstellung benötigten Ausgangsstoffe bzw. Zwischenprodukte.
Die in den jeweiligen Kombinationen bzw. bevorzugten Kombinationen von Resten im einzelnen für diese Reste angegebenen Restedefinitionen werden unabhängig von der jeweilig angegebenen Kombination der Reste, beliebig auch durch Restedefinitionen anderer Vorzugsbereiche ersetzt.
Die erfindungsgemäßen Verbindungen können gegebenenfalls als Mischungen verschiedener möglicher isomerer Formen, insbesondere von Stereoisomeren, wie z.B. E- und Z-, vorliegen. Es werden sowohl die E- als auch die Z-Isomeren, wie auch beliebige Mischungen dieser Isomeren, beansprucht.
Aus dem Stand der Technik ist die Chlorierung von Enolacetaten nicht bekannt. Es ist als ausgesprochen überraschend zu bezeichnen, dass beim erfmdungsgemäßen Verfahren in Reaktionsschritt a) die Chlorierung des Enolacetats der Formel (II) unter Abspaltung der Acetatgruppe erfolgt. Auch die Ringöffnung von Dichlorbenzo- furanon-3 mit Aminen ist nicht literaturbekannt.
Das erfindungsgemäße Verfahren weist eine Reihe von Vorteilen auf. Das erfindungsgemäße Verfahren wird insbesondere zur Herstellung von 2-(2-Hydroxyphe- nyl)-2-(alkoxyimino)-N-methylacetamid-Derivaten verwendet, die wichtige Zwischenprodukte für die Herstellung von Schädlingsbekämpfungsmitteln sind (vergleiche z.B. EP-A 398 692 und EP-A-937 050). Durch das erfindungsgemäße
Verfahren wird ein neuer synthetischer Zugang zu 2-(2-Hydroxyphenyl)-2-(alkoxy- imino)-N-methylacetamid-Derivaten eröffnet, der sich durch die leichte Zugänglichkeit der Edukte gute Ausbeuten der einzelnen Reaktionsstufen und gute technische Durchführbarkeit der einzelnen Reaktionsschritte auszeichnet.
Dieses neue Verfahren ist in Reaktionsschema 1 dargestellt.
Reaktionsschema 1
Die zur Durchführung des erfindungsgemäßen Verfahrens im Reaktionsschritt a) als Ausgangsstoff benötigte Verbindung der Formel (II) ist bereits bekannt und lässt sich
nach bekannten Methoden herstellen (vergleiche z.B. J. Chem. Soc. Perkin Trans 1, 1984, 1605-1612, WO 01/36405).
Das weiterhin zur Durchführung des erfindungsgemäßen Verfahrens im Reaktions- schritt a) als Ausgangsstoff benötigte Chlor ist eine kommerziell erhältliche Synthe- sechemikalie. Es kann sowohl als Gas, als auch als alkoholische oder wässrige Lösung in einem Verdünnungsmittel eingesetzt werden.
Als weiterhin zur Durchführung des erfindungsgemäßen Verfahrens im Reaktions- schritt b) als Ausgangsstoffe benötigte Amine der Formel (IV) oder deren Säureadditionskomplexe sind kommerziell erhältliche Synthesechemikalien. Sie können sowohl gegebenenfalls als Gas, als auch als alkoholische oder wässrige Lösung eingesetzt werden. Bevorzugte Amine der Formel (IV) sind Ethylamin und Methylamin. Besonders bevorzugt ist Methylamin. Bevorzugte Säureadditionskomplexe sind die Hydrochloride, Hydrogensulfate oder Sulfate.
Als weiterhin zur Durchführung des erfindungsgemäßen Verfahrens im Reaktionsschritt cl) als Ausgangsstoffe benötigte Hydroxylamin oder dessen Säureadditionskomplexe sind kommerziell erhältliche Synthesechemikalien. Bevorzugte Säure- additionskomplexe sind die Hydrochloride, Hydrogensulfate oder Sulfate.
Als weiterhin zur Durchführung des erfindungsgemäßen Verfahrens im Reaktions- schritt c2) als Ausgangsstoffe Alkylierungsmittel der Formel (VII) kommen alle chemischen Verbindungen infrage, die Alkylgruppen übertragen können. Bevorzugt sind Chlormethan, Brommethan, Iodmethan, Dimethylsulfat und Dimethylcarbonat.
Besonders bevorzugt sind Dimethylsulfat und Chlormethan.
Die weiterhin zur Durchführung erfindungsgemäßen Verfahrens im Reaktionsschritt d) als Ausgangsstoffe benötigten Alkoxyamine der Formel (V) bzw. deren Säure- additionskomplexe sind handelsübliche Synthesechemikalien. Bevorzugte Säureadditionskomplexe sind die Hydrochloride, Hydrogensulfate oder Sulfate.
Gegenstand der Erfindung ist auch ein Verfahren zur Herstellung von Verbindungen der Formel (V),
in welcher
R1 für Cι-C4-Alkyl steht,
dadurch gekennzeichnet, dass man
a) eine Verbindung der Formel (II)
mit Chlorgas gegebenenfalls in Gegenwart eines Verdünnungsmittels umsetzt und
b) die dabei entstehende Verbindung der Formel (III)
mit einem Amin der Formel (IV),
R -NH2 (IV) in welcher
R1 die oben angegebene Bedeutung hat,
- oder einem Säureadditionskomplex davon -
gegebenenfalls in Gegenwart eines Verdünnungsmittels und gegebenenfalls in Gegenwart eines Säureakzeptors umsetzt.
Ein weiterer Gegenstand der Erfindung sind die neuen Verbindungen der allgemeinen Formel (I),
in welcher
R1 für C i -C4- Alkyl steht und R2 für C2-C4- Alkyl steht.
In den neuen Verbindungen der Formel (I) R2 steht bevorzugt für Ethyl oder n-Pro- pyl, R2 steht besonders bevorzugt für Ethyl.
Reaktionsschritt a)
Als Verdünnungsmittel zur Durchführung des erfindungsgemäßen Verfahrens kommen in Rektionsschritt a) halogenierte Kohlenwasserstoffe, beispielhaft und vor-
zugsweise Chlorbenzol, Dichlorbenzol, Dichlormethan, Chloroform, Tetrachlormethan, Dichlorethan oder Trichlorethan; Nitrile, wie beispielhaft und vorzugsweise Acetonitril, Propionitril, n- oder i-Butyronitril oder Benzonitril; Eisessig; Ester wie beispielhaft und vorzugsweise Essigsäuremethylester oder Essigsäureethylester oder Alkohole, wie beispielhaft und vorzugsweise Methanol, Ethanol, n- oder i-Propanol, n-, i-, sek- oder tert-Butanol, Ethandiol, Propan-l,2-diol, Ethoxyethanol, Methoxy- ethanol, Diethylenglykolmonomethylether, Diethylenglykolmonoethylether, deren Gemische mit Wasser oder reines Wasser in Betracht. Bevorzugt wird in Reaktionsschritt a) Eisessig als Verdünnungsmittel verwendet.
Die Reaktionstemperaturen können bei der Durchführung des Reaktionsschritts a) des erfindungsgemäßen Verfahrens in einem größeren Bereich variiert werden. Im Allgemeinen arbeitet man bei Temperaturen von 15 °C bis 80°C, vorzugsweise bei Temperaturen von 15°C bis 60°C.
Reaktionsschritt a) des erfmdungsgemäßen Verfahrens wird im Allgemeinen unter Normaldruck durchgeführt. Es ist jedoch auch möglich, unter erhöhtem oder vermindertem Druck - im Allgemeinen zwischen 0,1 bar und 10 bar - zu arbeiten.
Zur Durchführung des Reaktionsschritts a) des erfindungsgemäßen Verfahrens zur
Herstellung der Verbindungen der Formel (I) setzt man pro Mol der Verbindung der Formel (II) im Allgemeinen 2 bis 5 Mol, vorzugsweise 2 bis 3 Mol Chlor ein.
Zur Durchführung des Reaktionsschritts a) des erfindungsgemäßen Verfahrens zur Herstellung der Verbindungen der Formel (I) geht man im' Allgemeinen wie folgt vor. Die Verbindung der Formel (II) (3-Acetoxybenzofuran) wird in dem Verdünnungsmittel, insbesondere in Eisessig, vorgelegt und mit Chlor versetzt. Man rührt gegebenenfalls bei erhöhter Temperatur bis die Umsetzung vollständig ist. Nach beendeter Reaktion wird in üblicher Weise aufgearbeitet.
Reaktionsschritt b)
Als Verdünnungsmittel zur Durchführung des erfindungsgemäßen Verfahrens kommen in Reaktionsschritt b) Ether, wie beispielhaft und vorzugsweise Diethylether, Diisopropylether, Methyl-t-butylether, Methyl-t-Amylether, Dioxan, Tetrahydrofu- ran, 1,2- Dimethoxyethan, 1,2-Diethoxyethan oder Anisol oder Alkohole, wie beispielhaft und vorzugsweise Methanol, Ethanol, n- oder i-Propanol, n-, i-, sek- oder tert-Butanol, Ethandiol, Propan-l,2-diol, Ethoxyethanol, Methoxyethanol, Diethylen- glykolmonomethylether, Diethylenglykolmonoethylether, deren Gemische mit Wasser oder reines Wasser in Betracht.
Der Reaktionsschritt b) des erfindungsgemäßen Verfahrens wird gegebenenfalls in Gegenwart eines geeigneten Säureakzeptors durchgeführt. Hierzu gehören beispielhaft und vorzugsweise Erdalkalimetall- oder Alkalimetallhydroxide, -carbonate oder -hydrogencarbonate, wie beispielsweise Natriumhydroxid, Kaliumhydroxid,
Natriumacetat, Kaliumacetat, Natriumcarbonat, Kaliumcarbonat, Kaliumhydrogen- carbonat oder Natriumhydrogencarbonat.
Die Reaktionstemperaturen können bei der Durchführung des Reaktionsschritts b) des erfindungsgemäßen Verfahrens in einem größeren Bereich variiert werden. Im
Allgemeinen arbeitet man bei Temperaturen von 15°C bis Rückflusstemperatur, vorzugsweise bei Temperaturen von 20°C bis 40°C.
Der Reaktionsschritt b) des erfindungsgemäßen Verfahrens wird im Allgemeinen unter Normaldruck durchgeführt. Es ist jedoch auch möglich, unter erhöhtem oder vermindertem Druck - im Allgemeinen zwischen 0,1 bar und 10 bar - zu arbeiten.
Zur Durchführung des Reaktionsschritts b) des erfindungsgemäßen Verfahrens zur Herstellung der Verbindungen der Formel (I) setzt man pro Mol der Verbindung der Formel (III) im Allgemeinen 4 bis 10 Mol, vorzugsweise 4 bis 8 Mol des Amins der
Formel (IV) ein.
Zur Durchführung des Reaktionsschritts b) des erfindungsgemäßen Verfahrens geht man im Allgemeinen wie folgt vor. Das Alkylamin der Formel (IV) wird zu der Verbindung der Formel (III), vorzugsweise in Gegenwart eines Verdünnungsmittels gegeben. Man rührt in einem Verdünnungsmittel gegebenenfalls bei erhöhter Tem- peratur bis die Umsetzung vollständig ist. Nach beendeter Reaktion wird in üblicher
Weise aufgearbeitet.
Reaktionsschritte cl) und d)
Als Verdünnungsmittel zur Durchführung der Reaktionsschritte cl) und d) des erfmdungsgemäßen Verfahrens kommen alle inerten organischen Lösungsmittel in Betracht. Hierzu gehören beispielhaft und vorzugsweise Ether, wie Diethylether, Diisopropylether, Methyl-t-butylether, Mefhyl-t-Amylefher, Dioxan, Tetrahydro- furan, 1,2- Dimethoxyethan, 1,2-Diethoxyethan oder Anisol; Nitrile, wie Acetonitril, Propionitril, n- oder i-Butyronitril oder Benzonitril; Amide, wie N,N-Dimethylform- amid, N,N-Dimethylacetamid, N-Methylformanilid, N-Methylpyrrolidon oder Hexa- methylphosphorsäuretriamid; Ester wie Essigsäuremethylester oder Essigsäureethylester; Sulfoxide, wie Dimethylsulfoxid; Sulfone, wie Sulfolan; Alkohole, wie Methanol, Ethanol, n- oder i-Propanol, n-, i-, sek- oder tert-Butanol, Ethandiol, Propan-l,2-diol, Ethoxyethanol, Methoxyethanol, Diefhylenglykolmonomethylether,
Diethylenglykolmonoethy lether, Bevorzugte Verdünnungsmittel sind Alkohole, deren Gemische mit Wasser oder reines Wasser.
Die Reaktionsschritte cl) und d) des erfindungsgemäßen Verfahrens werden gege- benenfalls in Gegenwart eines Puffermediums durchgeführt. Als Puffermedien kommen alle üblichen Säure/Salzgemische infrage, die den pH- Wert im Bereich von 1 bis 7 puffern. Bevorzugt ist das Gemisch Essigsäure/Natriumacetat oder kein Puffeimedium.
Die Reaktionstemperaturen können bei der Durchführung der Reaktionsschritte cl) und d) des erfindungsgemäßen Verfahrens in einem größeren Bereich variiert wer-
den. Im Allgemeinen arbeitet man bei Temperaturen von -20°C bis 150°C, vorzugsweise bei Temperaturen von 0°C bis 80°C.
Die Reaktionsschritte cl) und d) des erfindungsgemäßen Verfahrens werden im Allgemeinen unter Normaldruck durchgeführt. Es ist jedoch auch möglich, unter erhöhtem oder vermindertem Druck - im Allgemeinen zwischen 0,1 bar und 10 bar - zu arbeiten.
Zur Durchführung des Reaktionsschritts cl) des erfindungsgemäßen Verfahrens zur Herstellung der Verbindungen der Formel (VI) setzt man pro Mol der Verbindung der Formel (V) im Allgemeinen 1 bis 15 Mol, vorzugsweise 1 bis 2 Mol Hydroxylamin oder eines Säureadditionskomplexes davon ein.
Zur Durchführung des Reaktionsschritts d) des erfindungsgemäßen Verfahrens zur Herstellung der Verbindungen der Formel (I) setzt man pro Mol der Verbindung der
Formel (V) im Allgemeinen 1 bis 15 Mol, vorzugsweise 1 bis 2 Mol Alkoxyamin der Formel (VIII) oder eines Säureadditionskomplexes davon ein.
Zur Durchführung des Reaktionsschritts cl) des erfindungsgemäßen Verfahrens geht man im Allgemeinen wie folgt vor. Die Verbindung der Formel (V) wird vorzugsweise in Gegenwart eines Verdünnungsmittels mit Hydroxylamin, bzw. dessen Säureadditionskomplex, und gegebenenfalls dem Puffermedium versetzt und erwärmt. Nach beendeter Reaktion wird in üblicher Weise aufgearbeitet.
Zur Durchführung des Reaktionsschritts d) des erfindungsgemäßen Verfahrens geht man im Allgemeinen wie folgt vor. Die Verbindung der Formel (V) wird vorzugsweise in Gegenwart eines Verdünnungsmittels mit Alkoxyamin, bzw. dessen Säureadditionskomplex, und gegebenenfalls dem Puffermedium versetzt und erwärmt. Nach beendeter Reaktion wird in üblicher Weise aufgearbeitet.
Reaktionsschritt c2)
Als Verdünnungsmittel zur Durchführung des Reaktionsschritts c2) des erfindungsgemäßen Verfahrens kommen in Verfahrensschritt c2) alle inerten organischen Lösungsmittel in Betracht. Hierzu gehören beispielhaft und vorzugsweise aliphati- sche, alicyclische oder aromatische Kohlenwasserstoffe, wie beispielsweise Petrol- efher, Hexan, Heptan, Cyclohexan, Methylcyclohexan, Benzol, Toluol, Xylol oder Decalin; halogenierte Kohlenwasserstoffe, wie beispielsweise Chlorbenzol, Dichlor- benzol, Dichlormethan, Chloroform, Tetrachlormethan, Dichlorethan oder Trichlor- ethan; Ether, wie beispielsweise Diethylether, Diisopropylether, Methyl-t-butylether,
Methyl-t-Amylether, Dioxan, Tetrahydrofuran, 1,2- Dimethoxyethan, 1,2-Diethoxy- ethan oder Anisol; Ketone, wie beispielsweise Aceton, Butanon, Methyl-isobutyl- keton oder Cyclohexanon; Nitrile, wie beispielsweise Acetonitril, Propionitril, n- oder i-Butyronitril oder Benzonitril; Amide, wie N,N-Dimethylformamid, N,N- Dimethylacetamid, N-Methylformanilid, N-Methylpyrrolidon oder Hexamefhyl- phosphorsäuretriamid; Ester wie Essigsäuremethylester oder Essigsäureethylester; Sulfoxide, wie Dimefhylsulfoxid; Sulfone, wie Sulfolan; Alkohole, wie Methanol, Ethanol, n- oder i-Propanol, n-, i-, sek- oder tert-Butanol, Ethandiol, Propan-l,2-diol, Ethoxyethanol, Methoxyethanol, Diethylenglykolmonomethylether, Diethylengly- kolmonoethylether, deren Gemische mit Wasser oder reines Wasser.
Der Reaktionsschritt c2) des erfindungsgemäßen Verfahrens wird gegebenenfalls in Gegenwart eines geeigneten Säureakzeptors durchgeführt. Als solche kommen alle üblichen anorganischen oder organischen Basen infrage. Hierzu gehören beispielhaft und vorzugsweise Erdalkalimetall- oder Alkalimetallhydroxide, -alkoholate, -acetate,
-carbonate oder -hydrogencarbonate, wie beispielsweise Natriummethanolat, Na- triumethanolat, Kalium-tert.-butanolat, Natriumhydroxid, Kaliumhydroxid, Natrium- acetat, Kaliumacetat, Natriumcarbonat, Kaliumcarbonat, Kaliumhydrogencarbonat oder Natriumhydrogencarbonat, sowie tertiäre Amine, wie beispielsweise Trimefhyl- amin, Triethylamin, Tributylamin, N,N-Dimethylanilin, N,N-Dimefhyl-benzylamin,
Pyridin, N-Methylpiperidin, N-Methylmorpholin, N,N-Dimethylaminopyridin,
Diazabicyclooctan (DABCO), Diazabicyclononen (DBN) oder Diazabicycloundecen (DBU). Bevorzugt werden in Verfahrensschritt c2) Kaliumcarbonat, Natriumhydroxid oder Kaliumhydroxid verwendet.
Der Reaktionsschritt c2) des erfindungsgemäßen Verfahrens wird gegebenenfalls in
Gegenwart eines geeigneten Phasentransferkatalysators durchgeführt. Hierzu gehören beispielhaft und vorzugsweise quartäre Ammoniumsalze, wie beispielsweise Tetra- butylammoniumbromid, -chlorid, -hydrogensulfat oder -sulfat, Methyl-trioctyl- ammoniumbromid oder -chlorid, -hydrogensulfat oder -sulfat oder 4-Dimethyl- amino-N-(2-ethyl-hexyl)pyridiniumbromid oder -chlorid, -hydrogensulfat oder - sulfat, quartäre Phosphoniumsalze, wie beispielsweise Tributyl-tetradecyl-phospho- niumbromid oder -chlorid, Tetraphenylphosphoniumbromid oder -chlorid, Kronenether, wie beispielsweise Dibenzo-18-krone-6, Guanidiniumsalze, wie beispielsweise Hexaalkylguanidiniumchlorid, sowie Polyethylenglycolderivate. Bevorzugt wird in Verfahrensschritt c2) Tetrabutylammoniumbromid oder Tetrabutylamrnoniumchlorid verwendet.
Die Reaktionstemperaturen können bei der Durchführung des Reaktionsschritts c2) des erfindungsgemäßen Verfahrens in einem größeren Bereich variiert werden. Im Allgemeinen arbeitet man bei Temperaturen von -20°C bis Rückflusstemperatur, vorzugsweise bei Temperaturen von 0°C bis 60°C.
Der Reaktionsschritt c2) des erfindungsgemäßen Verfahrens wird im Allgemeinen unter Normaldruck durchgeführt. Es ist jedoch auch möglich, unter erhöhtem oder vermindertem Druck - im Allgemeinen zwischen 0,1 bar und 10 bar - zu arbeiten.
Zur Durchführung des Reaktionsschritts c2) des erfindungsgemäßen Verfahrens zur Herstellung der Verbindungen der Formel (I) setzt man pro Mol der Verbindung der Formel (VI) im Allgemeinen 1 bis 15 Mol, vorzugsweise 1 bis 2 Mol Alkylierungs- mittel der Formel (VII) ein.
Zur Durchführung des Reaktionsschritts c2) des erfindungsgemäßen Verfahrens geht man im Allgemeinen wie folgt vor. Die Verbindung der Formel (VI) wird vorzugsweise in Gegenwart eines Verdünnungsmittels mit einer Base und gegebenenfalls einem Phasentransferkatalysators versetzt. Hierzu gibt man das Alkylierungsmittel der Formel (VII) und rührt, gegebenenfalls bei erhöhter Temperatur bis die Umsetzung vollständig ist. Nach beendeter Reaktion wird in üblicher Weise aufgearbeitet.
Die nachfolgenden Beispiele dienen zur Erläuterung der Erfindung. Die Erfindung ist jedoch nicht auf die Beispiele limitiert.
Herstellungsbeispiele:
Beispiel 1
2,2-Dichloro-l -benzofuran-3(2H)-on
Reaktionsschritt a)
5.0 g 3-Acetoxybenzofuran werden in 15 ml Eisessig bei 50°C vorgelegt und innerhalb 1.5 Stunden mit 5.0 g Chlor versetzt. Man rührt 0.5 Stunden bei 50°C nach und destilliert anschließend das Lösungsmittel ab. Der Rückstand wird in Toluol aufgenommen und im Vakuum bis zur Trockene eingeengt. Man erhält 5.7 g 2,2-Dichloro- l-benzofuran-3(2H)-on (GC: 98.1 %; Ausbeute: 98.4 % d. Th.) als Feststoff.
(2Z)-2-(2-Hydroxyphenyl)-2-(methoxyimino)-N-methylethanamid:
Verfahrensschritt b) und d) (Eintopfverfahren)
5.39 g 2,2-Dichloro-l-benzofuran-3(2H)-on (GC: 97.9 %) werden in 60 ml Methyl- tert.-buty lether vorgelegt und bei Raumtemperatur innerhalb von 10 Minuten mit 10.09 g 40%iger wässriger Methylaminlösung versetzt. Man rührt 3 Stunden nach und gibt bei Raumtemperatur innerhalb von 5 Minuten eine Lösung aus 8.8 ml
Wasser, 2.61 g Methoxyaminhydrochlorid und 2.56 g Natriumacetat hinzu. Es wird 9 Stunden nachgerührt, das Lösungsmittel im Vakuum abdestilliert und der Rückstand mit 150 ml Eiswasser versetzt. Das Produkt wird abgesaugt, mit Wasser gewaschen und bei 50°C im Vakuum über Nacht getrocknet. Man erhält (2Z)-2-(2- hydroxyphenyl)-2-(methoxyimino)-N-methylethanamid als Feststoff (4.6 g, HPLC-
STD: 91.5 %, 77.8 % d. Th.).
Beispiel 2
2,2-Dichloro-l-benzofuran-3(2H)-on
Reaktionsschritt a)
5.0 g 3-Acetoxybenzofuran werden in 15 ml Eisessig bei 50°C vorgelegt und innerhalb 1.5 Stunden mit 5.0 g Chlor versetzt. Man rührt 0.5 Stunden bei 50°C nach und destilliert anschließend das Lösungsmittel ab. Der Rückstand wird in Toluol aufgenommen und im Vakuum bis zur Trockene eingeengt. Man erhält 5.7 g 2,2-Dichloro- l-benzofuran-3(2H)-on (GC: 98.1 %; Ausbeute: 98.4 % d. Th.) als Feststoff.
(2Z)-2-(Hydroxyimino)-2-(2-hydroxyphenyl)-N-methylethanamid:
Verfahrensschritte b) und cl)
5.36 g 2,2-Dichloro-l-benzofuran-3(2H)-on (GC: 98.5 %) werden in 14 ml Methanol suspendiert und bei Raumtemperatur innerhalb von 15 Minuten mit 10.09 g 40 %iger methanolischer Methylaminlösung versetzt. Man rührt 1 Stunde nach und gibt bei Raumtemperatur innerhalb von 10 Minuten eine Lösung aus 5.0 ml Wasser und
2.59 g Hydroxylammoniumsulfat hinzu. Es wird 1 Stunde nachgerührt, das Lösungsmittel im Vakuum abdestilliert, der Rückstand mit Wasser versetzt und pH 5 eingestellt. Man extrahiert dreimal mit Essigester, trocknet die vereinigten organischen Extrakte über Natriumsulfat und engt bis zur Trockene ein. Man erhält (2Z)-2- (hydroxyimino)-2-(2-hydroxyphenyl)-N-methylethanamid als Feststoff (4.2 g,
HPLC-STD: 85.2 %, Z/E = 81/19, 70.9 % d. Th.).
Beispiel 3
2-(2-Hydroxyphenyl)-N-methyl-2-(methylimino)ethanamid:
Reaktionsschritt b)
5.0 g 2,2-Dichloro-l-benzofuran-3(2H)-on werden in 20 ml Methyltertbutylether vorgelegt und bei Raumtemperatur innerhalb von 30 Minuten mit 14.0 g Methylamin (41 %-ig in Wasser) versetzt. Man rührt 4.5 Stunden bei Raumtemperatur nach, extrahiert den Ansatz dreimal mit Essigester, wäscht die vereinigten organischen Extrakte mit Wasser, trocknet die organische Phase mit Natriumsulfat und destilliert das Lösungsmittel ab. Man erhält 4.4 g 2-(2-Hydroxyphenyl)-N-methyl-2-(methyl- imino)-ethanamid (GC: 93.0 %; Ausbeute: 86.5 % d. Th.) als Feststoff.
Claims
Patentansprüche
Verfahren zur Herstellung von Verbindungen der allgemeinen Formel (I),
in welcher
R . 11 , u.„ndJ τ R>2 für Cι-C4- Alkyl stehen,
dadurch gekennzeichnet, dass man
a) eine Verbindung der Formel (II)
mit Chlorgas gegebenenfalls in Gegenwart eines Verdünnungsmittels umsetzt;
b) die dabei entstehende Verbindung der Formel (III)
mit einem Amin der Formel (IV),
R -NH2 (IV)
in welcher
R1 die oben angegebene Bedeutung hat,
- oder einem Säureadditionskomplex davon -
gegebenenfalls in Gegenwart eines Verdünnungsmittels und gegebenenfalls in Gegenwart eines Säureakzeptors, zu einer Verbindung der Formel (V) umsetzt;
diese
cl) mit Hydroxylamin - oder einem Säureadditionskomplex davon - gegebenenfalls in Gegenwart eines Verdünnungsmittels und gegebenenfalls in Gegenwart eines Puffermediums zu Verbindungen der Formel (VI) umsetzt;
und
c2) mit einem Alkylierungsmittel der Formel (VII),
R2-X (VII)
in welcher
R die oben angegebene Bedeutung hat, und
X für Halogen, -O-CO-O- R2, -O-SO2- R2, oder -O-SO2-O- R2 steht, wobei R die oben angegebene Bedeutung hat,
gegebenenfalls in Gegenwart eines Verdünnungsmittels und gegebenenfalls in Gegenwart eines Säureakzeptors, gegebenenfalls in Gegenwart eines Phasentransferkatalysators, umsetzt;
oder
d) mit Alkoxyaminen der Formel (VIII)
R2-O-NH2 (VIII),
in welcher
R2 die oben angegebene Bedeutung hat,
- oder einem Säureadditionskomplex davon -
gegebenenfalls in Gegenwart eines Verdünnungsmittels, gegebenen- falls in Gegenwart eines Säureakzeptors und gegebenenfalls in
Gegenwart eines Puffermediums, umsetzt.
2. Verfahren gemäß Anspruch 1, dadurch gekennzeichnet, dass die Reaktionsschritte a) und b) bei 20 °C bis Rückflusstemperatur durchgeführt werden.
3. Verfahren gemäß mindestens einem der Ansprüche 1 bis 2, dadurch gekennzeichnet, dass als Verdünnungsmittel in Reaktionsschritt a) Eisessig verwendet wird.
4. Verfahren gemäß mindestens einem der Ansprüche 1 bis 3, dadurch gekenn- zeichnet, dass R1 und R2 für Methyl stehen.
5. Verfahren mindestens einem der Ansprüche 1 bis 4, dadurch gekennzeichnet, dass in Reaktionsschritt a) 2 bis 5 Mol Chlor bezogen auf die Verbindung der Formel (II) verwendet werden.
6. Verfahren mindestens einem der Ansprüche 1 bis 5, dadurch gekennzeichnet in Reaktionsschritt b) 4 bis 8 Mol Alkylamin der Formel (IV) bezogen auf die Verbindung der Formel (III) verwendet werden.
7. Verfahren gemäß mindestens einem der Ansprüche 1 bis 6, dadurch gekennzeichnet, dass im Reaktionsschritt b) als Amin der Formel (IV) Methylamin verwendet wird.
8. Verfahren gemäß mindestens einem der Ansprüche 1 bis 7, dadurch gekennzeichnet, dass die Reaktionsschritte b) und d) als Eintopfreaktion durchgeführt werden.
9. Verfahren gemäß mindestens einem der Ansprüche 1 bis 7, dadurch gekennzeichnet, dass die Reaktionsschritte b) und cl) als Eintopfverfahren durchgeführt werden.
0. Verfahren zur Herstellung von Verbindungen der Formel (V),
in welcher
R1 für Cι-C4-Alkyl steht
dadurch gekennzeichnet, dass man
a) eine Verbindung der Formel (II)
mit Chlorgas gegebenenfalls in Gegenwart eines Verdünnungsmittels umsetzt;
b) die dabei entstehende Verbindung der Formel (III)
mit einem Amin der Formel (IV),
R -NH2 (IV),
in welcher
R1 die oben angegebene Bedeutung hat,
- oder einem Säureadditionskomplex davon -
gegebenenfalls in Gegenwart eines Verdünnungsmittels und gegebenenfalls in Gegenwart eines Säureakzeptors umsetzt.
11. Verbindungen der allgemeinen Formel (I),
in welcher
R1 für C i -C4- Alkyl steht und
R2 für C2-C4- Alkyl steht.
