Description
NOUVEAUX LIMONOIDES UTILES EN PARTICULIER DANS LE TRAITEMENT DU CANCER ET LEUR PROCEDE DE PREPARATION
La présente invention concerne de nouveaux limonoïdes, leur procédé de purification, ainsi que de préparation, et leur utilisation, en particulier dans le traitement du cancer, ou le traitement du paludisme.
Ces nouveaux limonoïdes peuvent être isolés à partir des organes végétaux de certaines plantes, en particulier du genre Harrisonia, mais aussi de toute autre genre, qui pourrait les contenir. On trouve les plantes du genre Harrisonia, appartenant à la famille des simaroubacées principalement en Asie du Sud-Est. Par exemple, l'espèce Harrisonia perforata Merr. se rencontre au Viêt-nam, en Chine du Sud, au Laos, au Cambodge, en Thaïlande, aux Philippines, et en Indonésie. Cette espèce s'est montrée particulièrement apte à être utilisée en tant que matériel de départ dans la présente invention, mais il est évident que les procédés mis en œuvre dans la présente invention pourront être utilisés avec d'autres espèces, de la même famille botanique, ou d'autres familles.
Les limonoïdes sont des composés déjà décrits dans la littérature, et peuvent être trouvés dans de nombreuses espèces végétales. Quelques représentants de ce type de composés ont été signalés dans des plantes du genre Harrisonia (Kubo et al.
Heterocycles, 1976, 5, 485-98 ; Rajab et al. J. Nat. Prod, 1997, 60, 822-5 ; Koike et al. Tetrahedron, 1993, 49, 2209-16 ; Mitsunaga et al. Phytochemistry, 1994, 37,
1443-6) et des limonoïdes différents de ceux décrits dans la présente invention ont été isolés d 'Harrisonia perforata (Byrne et al. Austr. J Chem. 1991, 44, 165-9 ; Wei et al. Jiegou Huaxue, 1985, 4, 281-7 ; TranVan et al. Phytochemistry, 1995,
38, 213-5 ; Kamiuchi et al. Heterocycles, 1996, 43, 653-64).
C'est un des objets de la présente invention que de caractériser les nouveaux liminoïdes isolés de plantes d'un tel genre, ainsi que de fournir un procédé d'extraction.
Ces nouveaux liminoïdes peuvent être modifiés, et les produits obtenus par les modifications chimiques sont également des objets de la présente invention, de même que les procédés de modification.
Il est connu que les liminoïdes peuvent avoir un effet contre certains types de cancer, ou dans le traitement du paludisme. La Demanderesse a montré que certains des produits selon l'invention possèdent une activité cytotoxique remarquable contre la prolifération cellulaire. Ainsi, l'utilisation des composés selon l'invention dans le traitement du cancer ou du paludisme, ou pour la préparation de médicaments pour une telle utilisation est également un des objets de l'invention.
Les composés selon l'invention sont obtenus à partir de plantes du genre Harrisonia, de manière préférée Harrisonia perforata, suivant un procédé simple et bon marché.
Le procédé de l'invention consiste essentiellement à traiter des organes végétaux de Harrisonia perforata, en particulier les feuilles desséchées, réduites en poudre au préalable, par une solution hydroalcoolique, ce qui fournit une solution hydroalcoolique de principes actifs amers. Cette solution est ensuite traitée par une solution aqueuse d'acétate de plomb et le précipité formé est séparé par filtration. Le filtrat obtenu est traité par une solution aqueuse de sulfate de sodium. Après décantation et filtration on obtient un filtrat hydroalcoolique de principes amers que l'on peut traiter de deux façons différentes.
1. Le filtrat hydroalcoolique est traité, sous agitation, par du charbon actif qui absorbe les principes amers. On recueille le charbon par filtration. Ce charbon est desséché à 50° et réduit en poudre. Les principes amers retenus par cette poudre de charbon sont extraits par du chloroforme.
L'extrait chloroformique est évaporé à sec. On obtient l'extrait brut de principes actifs.
2. Le filtrat hydroalcoolique est évaporé à sec sous vide. L'extrait brut est extrait au chloroforme. La solution chloroformique, évaporée à sec, fournit l'extrait brut de principes actifs
L'extrait brut ainsi obtenu, contenant un mélange de nombreux principes actifs est également un des objets de l'invention, ainsi que les composés susceptibles d'être isolés à partir de cet extrait brut.
On effectue alors un traitement complémentaire de cet extrait brut pour en isoler les limonoïdes purs par chromatographie, soit sur colonne soit sur plaque de silice, en utilisant différents solvants, en particulier les toluène, cyclohexane, heptane, chloroforme, éther, acétate d'éthyle, éthanol, méthanol, chlorure de méthylène.
Les limonoïdes ainsi obtenus peuvent ensuite être modifiés en adaptant des procédés chimiques connus de l'homme du métier, afin de substituer certains radicaux, dans le but, par exemple, d'obtenir des composés possédant des stéréochimies différentes, ou une activité biologique améliorée. Ainsi, les exemples 7 et 10 décrivent certaines modifications connues de l'homme du métier.
Les composés selon l'invention possèdent tous des centres d'asymétrie et peuvent donc exister sous forme d'isomères optiques. La présente invention comprend aussi bien tous ces isomères, soit séparément, soit en tant que mélange.

Par « dérivés », on entend tous les intermédiaires métaboliques produits lors la synthèse biochimique de ces composés par le végétal, c'est-à-dire les
composés qui peuvent être obtenus en aval de la voie métabolique, ainsi que les composés présents en amont de cette même voie, et les composés qui en découlent.
Par « sels pharmaceutiquement acceptables », on entend les sels pouvant être formés à partir de ces composés, en particulier par association avec des bases classiques et non toxiques, organiques ou minérales. Les sels dérivés de bases minérales incluent les sels d'aluminium, de cuivre, de fer ferreux ou ferrique, de lithium, de manganèse, de zinc, et plus particulièrement les sels d'ammonium, de calcium, de potassium, de magnésium et de sodium. Les sels dérivés de bases organiques incluent les sels d'aminés primaires, secondaires ou tertiaires, d'aminés substitués, d'aminés cycliques ou de résines échangeuses d'ions basiques, comme l'arginine, la bétaïne, la caféine, la choline, l'éthylènediamine, la diéthylamine, la triéthylamine, la tripropylamine, la triméthylamine, la trométhamine, la lysine, l'arginine, la méthylglucamine, la pipérazine, la pipéridine, les résines de polyamine, les purines, la N-éthylpipéridine, la glucamine, ou la glucosamine. On préférera tout particulièrement les composés de formule (I), ayant pour formule particulière :

Parmi les dérivés des composés de formule générale (I), on entend comprendre les composés qui peuvent également être extraits du végétal considéré, font partie de la même voie métabolique que les composés de formule générale I, et font donc partie de l'invention.
Les exemples montreront que les composés suivants, qui possèdent la même structure que les composés de formule générale (I), peuvent être isolés et caractérisés à partir de l'extrait brut. Ils font donc partie de l'invention.

Les compositions pharmaceutiques contenant un composé selon l'invention, et des excipients pharmaceutiquement acceptables font également partie de l'invention. Ces compositions peuvent être formulées pour l'administration aux mammifères, y compris l'homme. On pourra ainsi utiliser toute sorte de d'agents liant, tensioactif, d'enrobage, de dispersion, de mouillage afin de favoriser la stabilité, la conservation, et la libération de ces composés. En particulier, on utilisera des excipients classiques, par exemple la gélatine, l'amidon, le lactose, le stéarate de magnésium, le talc, la gomme arabique, ou analogues. On pourra ainsi définir toutes sortes de compositions pharmaceutiques, en particulier injectables, ou permettant une prise orale. Les compositions permettant une prise orale pourront être des comprimés, des gélules, des poudres, des granules ou des suspensions ou solutions orales. Elles comprennent également les formes d'administration sublinguale et buccale. Les compositions injectables pourront l'être par voie sous-cutanée, intramusculaire, intraveineuse, intranasale ou intraoculaire.
L'utilisation d'un composé selon l'invention pour la préparation d'un médicament, en particulier destiné au traitement du cancer ou du paludisme fait également partie de l'invention. La posologie varie selon le traitement et selon l'affection en cause.
Les exemples qui suivent servent à illustrer l'invention, sans qu'ils ne représentent une quelconque limitation à l'invention. Il est entendu que les méthodes d'extraction chromatographiques peuvent être optimisées. En particulier, l'homme du métier reconnaîtra qu'un changement dans les solvants décrits dans ces exemples pourra éventuellement permettre d'obtenir les mêmes composés.
* EXEMPLE 1 :
1 000 g de feuilles séchées et pulvérisées de Harrisonia perforata sont mis en macération avec de l'éthanol aqueux titrant 25°, à la température du laboratoire. On extrait ensuite, par lixiviation, avec au total, 10 L d'alcool titrant 25°. Le lixiviat hydroalcoolique est traité par 400 ml de solution aqueuse d'acétate de plomb à 30%.
Il se forme un précipité que l'on élimine par filtration.
Le filtrat obtenu est ensuite traité par 120 ml d'une solution aqueuse de sulfate de sodium à 15%. Le précipité de sulfate de plomb qui s'est formé est éliminé par filtration. On obtient ainsi une solution hydroalcoolique de principes actifs. Cette solution est traitée, en trois fois, par agitation avec du charbon végétal (en tout 30 g de charbon ) qui retient les principes actifs. Le charbon est séparé par filtration et séché à 50° pendant 24 h. Ce charbon renfermant les principes actifs est introduit dans un Soxhlet et extrait par du chloroforme.
On évapore l'extrait chloroformique à sec sous vide pour obtenir 2,42 g d'extrait sec, nommé extrait N° 1. Le rendement en extrait sec est de 0,25%. * EXEMPLE 2 :
L'extrait N°l obtenu selon l'exemple 1 (175 g) est dissous dans du toluène (400 ml) et soumis à un traitement chromatographique sur colonne de silice 60 H.
On effectue une élution de la colonne de silice (1 kg) par des fractions de 1 L d'un mélange de toluène-éther (1 :1). Chaque fraction est évaporée à sec sous vide et pesée. Les différentes fractions sont analysées par chromatographie en couche mince de silice (CCM).
La première fraction Fi (4 L) par évaporation sous vide à sec fournit un résidu de 74,80 g nommé extrait N° 2.
Par cristallisation dans le méthanol ce résidu fournit l'haperforine A (HA, composé II.2) (1,30 g) qui présente les caractéristiques suivantes :

La deuxième fraction F (7 L) est évaporée à sec sous vide et redissoute dans le méthanol. Les cristaux formés séparés et purifiés fournissent 12,05 g d'haperforine E (HE, composé IV)

* EXEMPLE 3 :
L'extrait N°2 (91 g) de l'exemple 2 est dissous dans le toluène et soumis à un traitement chromatographique sur colonne de silice 60 H (1 kg). La colonne de silice est éluée par des fractions de solvants de 1 L (toluène-éther (1 :1).
La troisième fraction de 1 L est évaporée à siccité sous pression réduite et conduit à un résidu N°3 ( 52 g ) qui sera décrit dans l'exemple 4.
La quatrième fraction (4 L) par cristallisation du méthanol fournit l'haperforine E ( 6,20 g) décrite dans l'exemple 2.
* EXEMPLE 4 :
Les résidus des chromatographies des exemples 2 et 3 (127 g) sont dissous dans du toluène (400 ml) et soumis à un traitement chromatographique sur colonne de silice 60 H (1kg).
La colonne de silice est éluée par fraction de 1 000 ml d'un mélange de toluène-éther (7 : 3). Chaque fraction est évaporée à sec sous vide et pesée. La fraction 1 (2 L) fournit un résidu de 4 g, que l'on dissout dans du méthanol.
Les cristaux, formés après concentration et repos à basse température (3°) sont séparés et purifiés par recristallisation dans ce même solvant, fournissent l'haperforine A (225 mg), décrite dans l'exemple 2.
La fraction 3 ( 7 L) évaporée à sec ( 91 g) est traitée selon l'exemple 5.
* EXEMPLE 5 : Chromatographie N° 1
La fraction 3 ( 91 g) obtenue dans l'exemple 4 est dissoute dans du toluène (400 ml) et la solution obtenue est soumise à un traitement chromatographique sur colonne de silice 60 H (1kg).
La colonne de silice est éluée par fraction de 1 000 ml d'un mélange de toluène-éther (7 : 3). Chaque fraction est évaporée à sec sous vide et pesée.
La fraction 1 (2 L) fournit un résidu de 1,55 g. La fraction 2 (1 L) fournit un résidu de 10,95 g.

Les eaux mères de la fraction 2 de l'exemple 5 sont évaporées à sec, et le résidu obtenu (8,5 g), redissout dans l'heptane est soumis à une chromatographie sur colonne de silice (450 g). L'éluat heptane-acétate d'éthyle ( 4 : 1) est recueilli par fraction de 25 ml dans des tubes.
On évapore les fractions 13-15 à sec. Leur cristallisation dans le mélange de solvants, heptane-acétate d'éthyle (4:l)permet d'obtenir 373 mg ^haperforine A3 qui présente les caractéristiques suivantes : Haperforine A3 : C27H32O10 = 516, F 146-147° (MeOH) ; HR ICSM : MH+

* EXEMPLE 6 :
L'extrait N°l obtenu selon l'exemple 1 (1,48 g ) est dissous dans le chlorure de méthylène (40 ml ) et soumis à un traitement chromatographique sur colonne de silice (75 g).
L'éluat dans un mélange de chlorure de méthylène-acétate d'éthyle (90:10) est collecté par fractions de 8 ml.
On réunit les fractions (62-102), et on les évapore sous vide à sec (833 mg). Le produit obtenu (833 mg) est dissous dans le toluène et chromatographie sur la silice 60 H (30 g).
L'éluat obtenu avec le mélange toluène-éther (50:50) est évaporé à sec sous vide, pour donner 350 mg de résidu.

L'extrait N°l obtenu selon l'exemple 1 (3,24 g) est dissous dans du toluène et soumis à un traitement chromatographique sur colonne de silice (150g). L'éluat constitué par le mélange toluène-éther est récolté par fraction de 8 ml.
On réunit les fractions (161-174) et on les évapore à sec pour obtenir un résidu de 503 mg. L'acétylation de l'haperforine B3 selon le procédé suivant conduit à l'obtention de l'acétyl-haperforine B3 :
Une solution d'haperforine B3 (22 mg) dans l'anhydride acétique (0,5 ml) et la pyridine (0,5 ml) est laissée à la température ambiante pendant 24 h. Après addition d'eau (5 ml), les produits organiques sont extraits par de l'éther (3 fois 3 ml). La phase éthérée lavée, séchée sur MgSO et évaporée sous pression réduite fournit un résidu qui, purifié par chromatographie sur plaque de silice (éluant éther), fournit Tacétylhaperforine B3 (5 mg) dont les caractéristiques physiques ('H RMN, SM, CCM) sont identiques à celles de l'haperforine A.

Eluat 13 (chlorure de méthylène). L'éluat 13 évaporé à sec fournit un résidu de 66mg. Par cristallisation dans le chlorure de méthylène, on obtient ainsi l'haperforine Edéjà décrite dans l'exemple 6.
* EXEMPLE 10 : Traitement de l'haperforine C
A une solution de diisopropylamidure de lithium (0,3 ml d'une solution 0,35M dans le THF, soit 0,10 mM), on ajoute à -78°C une solution d'haperforine C (25 mg, 0,05 mM). Après lh, la réaction est neutralisée par addition d'acide acétique (15 mM). De l'éther est ajouté et la solution organique lavée successivement avec une solution aqueuse de bicarbonate de sodium et de l'eau, est évaporée à sec sous pression réduite. On obtient un mélange 1/1 d'haperforine C et du produit de retroMichaël de l'haperforine C dénommée isohaperforine C. Ce mélange est purifié par chromatographie sur plaque préparative. On obtient ainsi l'isohaperforine C pure qui présente les caractéristiques suivantes:

* EXEMPLE 1 1 : Utilisation des haperforines contre la prolifération cellulaire Le pouvoir anti-turnoral exercé in vitro par une famille de produits contenant une vingtaine d'exemples fut évalué à l'aide du test colorimétrique MTT sur 6 lignées cancéreuses humaines.
Le principe du test MTT est basé sur la réduction réalisée au niveau d'enzymes mitochondriales (de cellules vivantes puisque métaboliquement actives) du produit MTT (3-(4,5-diméthylthiazol-2-yl)-2,5 diphenyl tetrazolium bromide) de couleur jaune en un produit de couleur bleue, le formazan. La quantité de formazan ainsi obtenue est directement proportionnelle à la quantité de cellules vivantes dans le milieu de culture. Cette quantité de formazan est mesurée par spectrophotométrie.
Six lignées cellulaires cancéreuses humaines ont été utilisées, à savoir les modèles Al 72, U373 et U87 (ghoblastomes), A549 (cancer du poumon non-à-petites-cellules), et HCT-15 et LoVo (cancers colorectaux). Ces lignées cellulaires ont été obtenues auprès d'une banque de tissus tumoraux située aux USA, à savoir V American Type Culture Collection (ATCC, Rockville, MD, USA).
Le procédé expérimental consiste à ensemencer 20 000 à 50 000 (selon le type cellulaire utilisé) cellules/ml de milieu de culture dans des plaques multi-puits
de 96 puits contenant chacun 100 μl de milieu de culture avec la concentration cellulaire adéquate. Les cellules sont laissées au repos pendant 24 heures afin de favoriser au maximum leur adhérence sur le support de la plaque, puis on ajoute les différents produits dont on veut évaluer l'activité anti-tumorale. Chaque produit a été testé aux concentrations de 0.001, 0.01, 0.1, 1, 10 et 100 μg/ml, chaque concentration de produit étant étudiée en hexaplicat.
On effectue un contrôle, qui correspond à la condition expérimentale dans laquelle est ajouté le solvant. Dans le cas présent, le solvant est du DMSO utilisé à une concentration finale inférieure à 0.1% par rapport au volume total de culture cellulaire. En effet, on sait que des concentrations de DMSO allant jusqu'à 5% du volume totale de culture cellulaire ne perturbent pas la croissance des lignées utilisées pour le test.
On laisse les cellules en présence des produits pendant 72 heures, puis on ajoute le produit MTT jaune et on laisse ce produit en présence des cellules pendant 3 heures. L'intensité de la coloration bleue résultant de la transformation du produit MTT jaune en formazan bleu par les cellules encore vivantes au terme de l'expérience est quantifiée par spectrophotométrie à l'aide d'un appareil de type DYNA TECH IMMUNOASSA Y SYSTEM.
Un logiciel intégré au spectrophotomètre calcule les valeurs moyennes de densité optique ainsi que les valeurs de déviation standard (Dév. Std.) et d'erreur standard sur la moyenne (ESM). Les évaluations statistiques se font à l'aide de tests paramétriques (analyse de la variance selon le test de Student) ou non paramétriques (test de rang de Mann-Whitney) selon que les distributions de valeurs de densité optique répondent ou non à une distribution normale (testées par un test Chi2) et que les variances sont ou ne sont pas égales (testées par le test de Levene).
Les résultats obtenus figurent dans le Tableau I, qui détaille l'activité observée pour 7 produits à chacune des concentrations testées pour chacune des 6 lignées utilisées dans le test.
On observe donc que les composés selon l'invention possèdent une activité remarquable de cytotoxicité pour les cellules cancéreuses, le produit HBl se révélant le plus intéressant en termes d'inhibition de la croissance tumorale.

Les nombres figurant dans le tableau I représentent la valeur moyenne (calculée sur 6 valeurs pour chacune des conditions expérimentales) du pourcentage de cellules présentes en fin d'expérimentation par rapport à la condition contrôle, cette dernière étant calibrée à 100% de manière arbitraire.

Procédé d'extraction de produits actifs bruts de limonoïdes à partir de végétaux, caractérisé en ce que : a. les feuilles des végétaux sont réduites en poudre, b. on traite les feuilles par une solution hydroalcoolique, pour obtenir la dissolution des principes actifs amers dans ladite solution hydroalcoolique, c. on traite la solution hydroalcoolique de principes amers par une solution aqueuse d'acétate de plomb, d. on sépare le précipité formé par filtration, e. on traite le filtrat obtenu par une solution aqueuse de sulfate de sodium, f. on filtre après décantation pour obtenir un filtrat hydroalcoolique de principes amers,

Procédé selon la revendication 6, caractérisé en ce que l'étape (f) est suivi des étapes suivantes : g. on traite le filtrat hydroalcoolique, sous agitation, par du charbon actif pour absorber les principes amers h. on recueille le charbon par filtration, on le dessèche et le réduit en poudre, i. on extrait les principes amers retenus par la poudre de charbon par du chloroforme, que l'on évapore ultérieurement à sec pour obtenir l'extrait brut de principes actifs.
8. Procédé selon la revendication 6, caractérisé en ce que l'étape (f) est suivie des étapes suivantes : g. on évapore le filtrat hydroalcoolique à sec sous vide h. on extrait l'extrait brut au chloroforme, que l'on évapore ultérieurement à sec pour obtenir l'extrait brut de principes actifs.
. Procédé selon l'une quelconque des revendication 6 à 8, caractérisé en ce que les végétaux sont du genre Harrisonia, en particulier de l'espèce Harrisonia perforata.
10. Extrait brut susceptible d'être obtenu par un procédé selon l'une quelconque des revendications 6 à 9.
