Peptide anti-microbien, dénommé myticine, caractérisé en ce qu'il est susceptible d'être obtenu à partir d'un mollusque bivalve, et en ce que sa masse moléculaire est d'environ 4,5 kDa; son pI est d'environ 8,7; il comprend 8 résidus cystéine, son procédé de préparation et ses applications. Acide nucléique codant pour ledit peptide.
Description
PEPTIDES ANTI-MICROBIENS DE MOLLUSQUES.
L' Invention est relative à de nouveaux peptides anti-microbiens produits par des mollusques.
Des polypeptides dotés de propriétés anti- microbiennes sont produits par une grande variété d'espèces (animales ou végétales), chez lesquelles ils participent à des mécanismes non-spécifiques de défense contre les infections.
Dans le cas des mollusques bivalves, on a jusqu'à présent identifié chez Mytil us galloprovincialis, un peptide dénommé MGD-1, apparenté aux défensines d'insectes [HUBERT et al., Eur. J. Bioche . , 240, 302- 306, (1996)] ; des peptides de la famille des défensines ont également été mis en évidence chez Mytilus edulis, ainsi que des peptides dénommés « mytilines » [CHARLET et al., J. Biol. Chem.,271, 21808-21813, (1996) ;.
Les Inventeurs ont maintenant mis en évidence de nouveaux peptides anti-microbiens produits par Mytilus galloprovincialis, qui sont différents des défensines MGD1, et des Mytilines précédemment connues.
La présente invention a pour objet des peptides anti-microbiens, dénommés ci-après : « myticines » qui possèdent les caractéristiques suivantes : - leur masse moléculaire est d'environ 4,5 kDa ;
- leur pi est d'environ 8,7 ;
- ils comprennent 8 résidus cystéine.
Selon un mode de réalisation préféré d'un peptide anti-microbien conforme à l'invention, il comprend la séquence (I) suivante (code 1 lettre) :

Les séquences (la) et (Ib) représentent les formes matures, isolées à partir de l' hémolymphe de Mytilus galloprovincialis, de 2 Myticines dénommées Myticine a et Myticine b, dont les ADNc ont également été clones par les Inventeurs. A titre d'illustration de l'objet de la présente invention, les caractéristiques des Myticines a et b sont plus spécifiquement indiquées ci-après .
La séquence d'ADNc et la séquence polypeptidique de la Myticine a sont représentées dans la liste de séquences en annexe sous les numéros SEQ ID NO: 1 et SEQ ID NO: 2. La séquence d'ADNc et la séquence polypeptidique de la Myticine b, sont représentées dans la liste de séquences en annexe sous les numéros SEQ ID NO: 3 et SEQ ID NO: 4.
Le peptide actif de 40 acides aminés, correspondant à la séquence (I), et plus particulièrement à l'une des séquences (la) et (Ib) est flanqué d'une séquence signal de 20 acides aminés, et d'un peptide C- terminal de 36 acides aminés. La séquence signal permettrait l'adressage du produit de traduction vers le réticulum endoplasmique. Le peptide C-terminal permettrait ensuite l'adressage vers les granules cytoplasmiques dans lesquels les Myticines sont stockées sous forme mature et/ou la protection de la cellule contre une éventuelle activité cytolytique du peptide mature.
La masse moléculaire de la forme mature de la Myticine a est de 4438 Da ; la masse moléculaire de la forme mature de la Myticine b est de 4562 Da.
Les Myticines ne présentent aucune homologie significative avec les peptides anti-microbiens connus dans l'art antérieur, et définissent un nouveau groupe de peptides anti-microbiens. Les Myticines peuvent être obtenues par extraction à partir des mollusques qui les produisent, ou bien par synthèse peptidique, ou, avantageusement, par génie génétique, en exprimant au moins une séquence d'acide nucléique codant une Myticine, dans une cellule hôte appropriée.
La présente Invention englobe également des acides nucléiques comprenant une séquence codant une Myticine, tel que définie ci-dessus.
Des acides nucléiques conformes à l'invention, peuvent être obtenus par criblage de banques d'acide nucléique à l'aide d'oligonucléotides dérivés des séquences SEQ ID NO: 1 ou SEQ ID NO: 3, ou de leurs séquences complémentaires. Les oligonucléotides utilisables dans ce but font également partie de l'objet de la présente invention ; avantageusement, ces oligonucléotides comprennent au moins 15 pb, et de préférence au moins 20 pb, de la portion codante de l'une des séquences SEQ ID NO: 1 ou SEQ ID NO: 3, ou de son complémentaire . Les acides nucléiques conformes à l'invention englobent également les cassettes d'expression, comprenant au moins une séquence d'acide nucléique codant une Myticine, placée sous contrôle transcriptionnel d'un promoteur approprié. Par « promoteur approprié » on entend tout promoteur fonctionnel dans la cellule-hôte destinée à héberger la cassette d'expression. Il peut s'agir d'un promoteur constitutif ou d'un promoteur inductible ; il peut également s'agir, dans le cas où la cassette est destinée à l'expression d'une Myticine chez un animal ou une plante, d'un promoteur tissu-spécifique.
Une cassette d'expression conforme à 1 ' invention peut comprendre également au moins une séquence codant une séquence d'adressage appropriée ; ladite séquence d'adressage peut être choisie parmi celles qui sont naturellement associées aux Myticines, telles que les séquences signal et/ou les séquences C- terminales associées aux isoformes Myticine a et Myticine b décrites ci-dessus ; il est également possible de choisir une ou plusieurs séquences d'adressage hétérologues, fonctionnelles dans une cellule-hôte donnée : il peut s'agir notamment de séquences permettant l'adressage d'une Myticine vers un compartiment cellulaire déterminé, ou sa sécrétion dans le milieu de culture. L'Invention a également pour objet :
- des vecteurs recombinants caractérisés en ce qu'ils comprennent au moins une séquence d'acide nucléique conforme à l'invention, codant une Myticine, et en particulier des vecteurs comprenant une cassette d'expression telle que définie ci-dessus. des cellules procaryotes ou eucaryotes transformées par au moins une séquence d'acide nucléique conforme à l'invention. Il peut s'agir de cellules en culture, ou de cellules faisant partie d'un organisme pluricellulaire, animal ou plante. La séquence d'acide nucléique conforme à l'invention présente dans une cellule transformée peut être soit incorporée dans l'ADN chromosomique de ladite cellule, soit être portée par un vecteur extra-chromosomique. L'invention a également pour objet un procédé de production d'une Myticine, caractérisé en ce qu'il comprend l'expression de ladite Myticine, dans au moins une cellule transformée conforme à l'invention.
Les Myticines conformes à l'invention peuvent être exprimées dans des cultures de cellules transformées en utilisant des techniques similaires à celles utilisées
pour des peptides antimicrobiens de l'art antérieur, par exemple dans des cellules d'insecte, comme décrit par HELLERS et al. [Eur. J. Bioche . 199, pp. 435-439, (1991)] pour les cécropines, ou dans la levure, comme décrit par REICHHART et al. [Invertebrate Reproduction and Development, 21, pp. 15-24, (1992)].
Elles peuvent également être exprimées dans des animaux ou des plantes transgéniques, pour augmenter la résistance de ceux-ci aux infections, comme décrit par exemple par JAYNES et al., [Plant Science, 89, pp. 43-53
(1993)] dans le cas de peptides analogues de la cécropine
B, exprimés dans des plants de tabac transgénique, ou par
NORELLI et al. [Euphytica, 77, pp. 123-128 (1994)] pour des plants de pommiers transgéniques exprimant le gène de l' attacine-E.
Les Myticines sont utilisables en particulier pour l'obtention de produits et en particulier de médicaments anti-infectieux, par exemple anti-bactériens ou fongicides. De tels produits trouvent leur application pour la prévention et le traitement de différentes maladies microbiennes, dans des secteurs très variés, en particulier dans les domaines de la santé et de l'agriculture, et dans celui de l'aquaculture, pour limiter le développement de maladies infectieuses dans les élevages.
La présente invention sera mieux comprise à l'aide du complément de description qui va suivre, qui se réfère à des exemples de purification et de caractérisation des Myticines.
EXEMPLE 1 ISOLATION DE PEPTIDES ANTI-MICROBIENS A PARTIR DE L'HEMOLYMPHE DE MYTILUS GALLOPROVINCIALIS.
Préparation des fractions de l' émolymphe
Une réaction immunitaire est induite chez des moules (Mytilus galloprovincialis) adultes selon le protocole suivant : la coquille est vidée de son liquide,
et 100 μl d'une suspension de bactéries (109 bactéries/ml) ou de champignons (suspension d'hyphes à 1 DO à 600nm) , préalablement tués par la chaleur sont injectés dans le muscle adducteur. L' hémolymphe (approximativement 0,5 ml/animal) est prélevée à partir du muscle postérieur adducteur à l'aide d'une seringue en présence de 1 volume de tampon anti-agrégant MAS (Modified Alsevier Solution) et centrifugée immédiatement à 800g pendant 15 min à 4°C. Le surnageant, correspondant à la fraction plasmatique, est additionné d'aprotinine (5μg/ml) et congelé (-80°C) jusqu'à utilisation, et le culot cellulaire est séché et conservé à -80°C jusqu'à utilisation. Purification des Myticines .
Fraction plasmatique : Le plasma est dilué (1:1 v/v) dans de l'eau stérilisée par ultrafiltration
(MilliQ) additionnée de 0,1% d'acide trifluoroacétique.
Le pH est amené à 3,9 par addition d'HCl 1M, sous agitation dans un bain-marie glacé pendant 30 min. Après centrifugation (10000 g, 20 min, 4°C) le surnageant est recueilli et maintenu à 4°C jusqu'à utilisation.
Hémocytes : Après décongélation le culot d'hémocytes est resuspendu dans 5 volumes de tampon Tris 50 mM, pH 8.7, contenant 50mM NaCl, et homogénéisé. Après centrifugation (10000 g, 20 min, 4°C), le culot contenant les organites cellulaires est repris dans 3 volumes d'acide acétique 2 M, et traité par sonication (3 X 30s) dans dans un bain-marie glacé. Après élimination des débris par • centrifugation (10000 g, 20 min, 4°C) l'extrait acide est conservé à 4°C jusqu'à utilisation. Purification HPLC
La fraction plasmatique ou les extraits acides d'hémocytes sont déposés sur des colonnes SEP-PAK C18 VAC
(WATERS ASSOCIATES) pré-équilibrées par de l'eau acidifiée (0,05% acide trifluoroacétique) . Après lavage à l'eau acidifiée, on effectue 2 elutions successives par des solutions d' acétonitrile à 10% et 40% dans de l'eau
acidifiée (0,05% acide trifluoroacétique). Les fractions obtenues sont lyophilisées et reconstituées avec de l'eau ultrafiltrée avant d'être soumises à une chromatographie HPLC en phase inverse. Toutes les étapes de purification HPLC ont été effectuées sur un système HPLC BECKMAN GOLD HPLC équipé d'un détecteur BECKMAN 168. L'élution est suivie par mesure de l'absorption UV à 225 nm.
Etape 1: les fractions éluées sur SEP-PAK à 40% d' acétonitrile sont déposées sur une colonne HPLC phase inverse SEPHASIL C18 (250 mm X 4,1 mm) (PHARMACIA). L'élution est effectuée par un gradient linéaire de 5 à 50% d' acétonitrile dans l'eau acidifiée, pendant 90 min à un débit de 0,9 ml/min. Les fractions correspondant aux pics d'absorbance sont collectées dans des tubes de polypropylène (MICROSORB 75 X 12 mm, NUNC IMMUNOTUBES) , séchées sous vide, et reconstituées avec de l'eau ultrafiltrée, préalablement au test de leur activité antimicrobienne . Etape 2: Les fractions actives récupérées à l'issue de l'étape 1 sont déposées sur une colonne HPLC phase inverse SEPHASIL C8 (250 mm X 4,1 mm) (PHARMACIA). L'élution est effectuée, à un débit de 0,9 ml/min, par un gradient linéaire de 20 à 30% d' acétonitrile dans l'eau acidifiée pendant 40 min.
Etape 3: Les fractions actives récupérées à l'issue de l'étape 2, sont déposées sur une colonne SEPHASIL C18, (250 mm X 4,1 mm) (PHARMACIA) en utilisant le gradient biphasique décrit dans l'étape 2, à un débit de 0, 9 ml/min.
Etape 4: La dernière étape de purification est effectuée sur une colonne phase inverse DELTA PAK HPI C18
(2 X 150 mm) (WATERS ASSOCIATES) en utilisant le gradient biphasique décrit dans l'étape 2, à un débit de 0,3 ml/min.
EXEMPLE 2 : ACTIVITE ANTIMICROBIENNE DES PEPTIDES OBTENUS
Micro-organismes utilisés :
La liste des micro-organismes utilisés pour déterminer les activités anti-microbiennes de la Myticine a et de la Myticine b est indiquée ci-après, dans le Tableau 1
Essais antibactériens et détermination de la CMB:
La concentration minimale bactéricide (CMB) des peptides a été déterminée selon le protocole décrit par HANCOCK et al. [http://www.interchg.ubc.ca/bobh/ methods .htm] .
On effectue une série de dilutions successives au 1/2 des peptides dans une solution aqueuse contenant 0,01% d'acide acétique et 0,2% de sérum albumine bovine (BSA) .
Des aliquotes de 10 μl de chaque dilution sont incubées dans des plaques stériles de microtitation en polypropylène à 96 puits, en présence de 100 μl de suspension bactérienne à une densité optique de départ A600 = 0,001 dans du milieu liquide MUELLER HINTON BROTH. L'incubation est effectuée pendant 18 h à 37 °C sous agitation, sauf dans le cas des bactéries marines, pour lesquelles l'incubation est effectuée à 25°C. La CMB est déterminée en étalant sur milieu solide MUELLER HINTON AGAR le contenu des puits correspondant aux 3 premières dilutions pour lesquelles on n'observe aucune croissance bactérienne, et en incubant à 37 °C pendant 18 heures. La plus faible concentration de peptide qui prévient toute formation résiduelle de colonies correspond à la CMB. Activité antifongique :
L'activité antifongique a été déterminée par calcul de la CMI (concentration minimale inhibitrice) dans un test d'inhibition de croissance de Fusarium oxysporum en phase liquide, selon le protocole décrit par FELHBAUM et al [J. Biol . Chem. , 269:33159-63, (1994)].
On effectue une série de dilutions successives au 1/2 des peptides, comme indiqué ci-dessus pour la détermination de l'activité antibactérienne.
80μl de spores suspendues (concentration finale, 104 spores/ml) dans du milieu « Potato Dextrose
Broth » (DIFCO) sont ajoutés à lOμl de solution de peptide dans des plaques stériles de microtitation en polypropylène à 96 puits. Le volume final est ajusté à lOOμl par addition d'eau. L'inhibition de croissance est déterminée après une incubation de 24 heures à 25°C à l'obscurité, par observation au microscope et mesure de l'augmentation de la D06oo- La valeur de la CMI correspond à un intervalle (a-b) de concentrations en peptide, où
(a) représente la concentration la plus élevée à laquelle on observe une croissance, et (b) représente la concentration la plus faible qui induit 100% d'inhibition de croissance.
Activité anti-protozoaire :
Le protozoaire parasite des huitres, Perkinsus marinus est cultivé en milieu DMEM (GIBCO) , selon le protocole décrit par GAUTHIER et VASTA [J. Invertebr.
Pathol., 66, 156-168, (1995)].
10 μM de peptide purifié sont additionnés à
4 x 10^ P. marinus, dans de l'eau de mer (volume final 20 μl) . Le mélange est incubé pendant 1 heure à température de la pièce. La viabilité des parasites est estimée par coloration à l'orange d'acridine et au bromure d'éthidium, comme décrit par MORVAN et al. [J.
Invertebr. Pathol., 69, 177-82, (1997)]. La viabilité maximale est évaluée, à titre de contrôle positif, dans des échantillons dans lesquels le peptide n'a pas été ajouté .
Les résultats des différentes expérimentations effectuées, pour les peptides Myticine a et Myticine b sont illustrées par le Tableau 1 ci-dessous ; les activités biologiques sont exprimées en μM.

Ces résultats montrent que les 2 peptides sont actifs en particulier sur Micrococcus luteus ; le peptide Myticine b apparaît en outre plus actif que le peptide Myticine a sur Micrococcus luteus , Escherichia coli , et Fusari um oxysporum. EXEMPLE 3 : CLONAGE DES ADNC DES PEPTIDES MYTICINE.
Une banque d'ADNc a été construite dans le vecteur ZAP EXPRESS (STRATAGENE) à partir des ARN poly(A)+ totaux d'hémocytes de moules adultes. Une sonde d'ADN représentant 83 pb de l'ADNc de la Myticine a a été construite en utilisant le kit de clonage PCR SCRIPT Amp
(SK+) (STRATAGENE) , et marquée par amorçage aléatoire en utilisant le kit de marquage d'ADN READY-TO-GO
(PHARMACIA), et utilisée pour cribler la banque d'ADN transférée sur membranes HYBOND-N, (AMERSHAM) . Des hybridations à stringence élevée ont été effectuées pendant une nuit à 65°C en solution de Denhardt 5X, 5X SSPE, 0,1 %SDS, 100 μg/ml d'ADN de sperme de saumon. Les
filtres préalablement rincés à 65°C en solution 0,5 X SSC contenant 0,1 % SDS, ont été autoradiographiés . Un criblage secondaire a été effectué pour purifier les clones positifs. Les phagemides ont été obtenus par excision in vivo et leurs 2 brins ont été séquences.
110 clones positifs ont été obtenus. Parmi ces clones, 4 ont été séquences, et correspondent aux peptides Myticine a et Myticine B.
Dans les 2 cas, la séquence en acides aminés déduite du cadre de lecture ouverte commence par un peptide signal de 20 acides aminés ; ce peptide-signal est directement suivi, à son extrémité C-terminale, par un peptide de 40 acides aminés, débutant par un résidu histidine, qui correspond à la forme active du peptide ; ce peptide actif est suivi par une extension C-terminale de 36 acides aminés.
Claims
REVENDICATIONS
1) Peptide anti-microbien, dénommé myticine, caractérisé en ce qu'il est susceptible d'être obtenu à partir d'un mollusque bivalve, et en ce que - sa masse moléculaire est d'environ 4,5 kDa ;
- son pi est d'environ 8,7 ;
- il comprend 8 résidus cystéine.
2) Peptide selon la revendication 1, caractérisé en ce qu'il comprend la séquence (I) suivante :

6) Cassette d'expression, comprenant au moins une séquence d'acide nucléique selon la revendication 4, sous contrôle transcriptionnel d'un promoteur approprié.
7) Vecteur recombinant caractérisé en ce qu'il comprend au moins une séquence d' acide nucléique selon la revendication 4.
8) Cellule procaryote ou eucaryote transformée par une séquence d'acide nucléique selon la revendication 4.
9) Procédé de production d'un peptide selon l'une quelconque des revendications 1 à 3, caractérisé en ce qu'il comprend l'expression d'un acide nucléique selon la revendication 4, dans au moins une cellule transformée selon la revendication 8.
10) Utilisation d'un peptide selon l'une quelconque des revendications 1 à 3 pour l'obtention d'un agent anti-microbien.
