Abstract

L'invention porte sur une protéine membranaire associée à la préséniline (PAMP), et sur des acides nucléiques codant cette protéine. PAMP et les acides nucléiques de PAMP sont des outils diagnostiques et thérapeutiques permettant d'évaluer et de traiter ou de prévenir les maladies dégénératives. Selon une réalisation spécifique, des mutations dans PAMP sont diagnostiquées pour la maladie d'Alzheimer ou le spina bifida. Cette invention porte également sur la recherche automatique, notamment à l'aide de cribles à haut rendement et de modèles d'animaux transgéniques, de composés qui modulent l'activité de PAMP et des présénilines. Ces composés, ou thérapie génique utilisant PAMP, peuvent être utilisés dans le traitement des maladies dégénératives telles que la maladie d'Alzheimer. Cette invention porte en outre sur des mutants de PAMP, sur des acides nucléiques codant ces mutants, et sur des animaux transgéniques exprimant les mutants de PAMP qui, selon une variante préférée, entraînent des modifications biochimiques similaires à celles induites par des mutations dans βAPP, PS1, ou PS2 associées à la maladie d'Alzheimer.
