So beschreibt Yamamoto in Tetrahedron Lett.
10 mol-% Jodwasserstoff. Man erhält bei 90°C und 9 MPa Kohlenmonoxid-Druck in

This is how Yamamoto describes Lett in Tetrahedron. 1997, Vol. 38, 3747 to 3750 a synthesis of isochroman-3-one by reacting 1,2-bis-hydroxymethyl-benzene and carbon monoxide in the presence of 1 mol% of a palladium catalyst and 10 mol% hydrogen iodide. Isochroman-3-one is obtained in isolated form at 90 ° C. and 9 MPa carbon monoxide pressure in acetone water as solvent after a reaction time of 42 hours in 56% yield.
The disadvantages of this method are the presence of the very corrosive To mention hydrogen iodide and the rather long reaction time.
Stille beschreibt in J. Am. Chem. Chem. Soc. 1980, vol. 102, 4193 to 4198 the synthesis of isochroman-3-one by reacting ortho-bromomethylbenzyl alcohol, Carbon monoxide and potassium carbonate in the presence of 1.6 mol% of one Palladium catalyst and a drop of hydrazine in tetrahydrofuran as Solvent. One obtains after 24 hours at 25 ° C and 0.1 MPa carbon monoxide pressure Isochroman-3-one in isolated form with a yield of 71%.
The disadvantage is that the ortho-bromomethylbenzyl alcohol required as the starting material is not easily accessible. In addition, the use of potassium carbonate makes it difficult to carry out the process easily (release of CO 2 ). In addition, a comparatively long response time has to be accepted.
der allgemeinen Formel (A)
Eisenkatalysatoren in Betracht. Als Wasserstoffhalogenidaufnahmemittel kommen
Basen, insbesondere anorganische Basen, beispielsweise Calciumhydroxid zum
Einsatz.
Isochroman-3-on herbeizuführen. Die maximale TOF beträgt 153 x h-1; TON = 153;
Ausbeute 76,7 % (vgl.
a,a'-ortho-Xylylendichlorid (1,2-Bis-chlormethylbenzol) in nicht weniger als 100 g
gewonnen (TON = 87;
Due to the use of bases in the first step of the process and the Acidification in the second step produces no less than 3 equivalents of monovalent Salt per equivalent of isochroman-3-one. On the one hand, this method is disadvantageous the use of large amounts of solvents and the formation of large amounts of salt and on the other hand, the two-stage process and the numerous cleaning and Extraction steps as well as the repeated use of ether as Extractant.
In view of the disadvantages of the methods described above, the The present invention has for its object a new method of manufacture of isochroman-3-ones to provide, on the one hand, with comparative can perform little effort, on the other hand, the disadvantages described above avoids the prior art processes and the desired product good yield and high purity.
der Formel (I)
The reaction of the 1,2-bis-halomethylbenzene of the formula (II) can be demonstrated using a 1,2-bis-chloromethylbenzene as an example of a compound of the formula (II) and using tert-butanol as an example of a compound of the formula (III) - described in simplified form by the following equation.
Wasser.
As can be seen from this equation, which serves as an illustrative example, the corresponding isochroman-3-one of the formula (I), tert-butyl chloride and water is formed.
The process according to the invention makes it possible to implement the 1,2-bis-halomethylbenzene of the formula (II) in concentrations which are significantly higher than in the process according to WO 97/00850 A1. As a result, the space / time yield is advantageously increased and technical implementation is favored to a corresponding extent.
Another advantage is that compared to the method of WO 97/00850 A1 does not form the salt of formula (B) and also does not form the salt is obtained by the reaction of the hydrogen halide (base) with Hydrogen halide is formed. The method according to the invention thus runs in Absence of such a hydrogen halide absorbent and it can also advantageously to the second reaction step with the addition of Acid are dispensed with.

The radicals R 1 , R 2 , R 3 and R 4 are, in particular independently of one another, hydrogen, fluorine, C 1 -C 4 -alkyl or C 1 -C 4 -alkoxy or two of the radicals R 1 , R 2 , R 3 and R 4 are linked together and form an aliphatic or aromatic ring with 5 to 10 carbon atoms. R 1 , R 2 , R 3 and R 4 are preferably independently of one another hydrogen, fluorine or C 1 -C 4 -alkyl or C 1 -C 4 -alkoxy, in particular hydrogen, fluorine or C 1 -C 4 -alkyl.

A compound of the formula (III) is used with good success, in which the radicals R 5 , R 6 and R 7 are identical or different and are for a C 1 -C 18 -alkyl- or (C 6 -C 18 -aryl) -CH 2 radical, in particular for a C 1 -C 12 alkyl radical, preferably for a C 1 -C 8 alkyl radical. The alkyl radical can be straight-chain or branched and is in particular straight-chain.
Of particular interest are compounds of the formula (III) in which one of the radicals R 5 , R 6 and R 7 is a C 1 -C 12 -alkyl radical, in particular a C 1 -C 8 -alkyl radical and the remaining radicals are an ethyl or methyl radical, in particular a methyl radical.
The process according to the invention is particularly simple by using tert-butanol used as a compound of formula III.

The H 0 value results from the following equation:
The following examples describe the invention without restricting it.
Nach dem Zudosieren läßt man 60 Minuten unter Rühren nachreagieren, kühlt ab und entleert den Autoklaven.299 g of 1,2-bis-chloromethylbenzene and 75 mg of palladium (II) chloride are in 725 g N-methyl-pyrrolidone dissolved and in a 2000 ml autoclave made of HC 4 steel Shielding gas atmosphere (argon) mixed. Then carbon monoxide added to a pressure of 2 MPa and the temperature increased to 150 ° C. At 150 ° C. and 2.5 MPa pressure become 1.7 g of a mixture within 2 minutes from water / N-methylpyrrolidone (weight ratio 1: 1) and then 415 g of a mixture of tert-butanol / N-methylpyrrolidone within 165 minutes (Weight ratio 2: 1) metered. The reaction pressure will kept constant at 4 MPa during metering. After dosing leaves the reaction is continued for 60 minutes with stirring, the mixture is cooled and the autoclave is emptied.
Man erhält 1445 g Reaktionsmischung.
(TON = 2649;
Man beobachtet keine Aufnahme
3-Isochromanon enthält.35.0 g of bis-chloromethylbenzene, 35 mg of palladium (II) chloride and 0.12 g of water are dissolved in 85 ml of tert-butanol and mixed in a 300 ml autoclave made of HC 256 steel in a protective gas atmosphere (argon). Then carbon monoxide is added at a pressure of 1.8 MPa and the temperature is raised to 100 ° C. No carbon monoxide uptake is observed during the temperature increase and at 100 ° C. The temperature is then slowly increased to 130 ° C, and carbon monoxide is not absorbed.
A further 35 mg of palladium (II) chloride dissolved in 5 ml of tert-butanol and 0.12 g of water are metered in at a pressure of 2.3 MPa. No absorption of carbon monoxide is observed. A further dosage of 2 ml water / tert-butanol (1: 1) also does not lead to a reaction.
After a further 2 hours, the autoclave is cooled and let down. Gas chromatographic analysis shows that the reaction mixture contains no 3-isochromanone.
52,5 g 1,2-Bis-chlormethylbenzol, 1,05 g Bis(triphenylphosphin)-palladium(II)-chlorid und 0,87 g Triphenylphosphin werden in 100 ml tert.-Butanol vermischt und in einem 500 ml Glasautoklaven unter Schutzgasatmosphäre (Argon) mit 48,0 g Wasser und 46,8 g Calciumhydroxid suspendiert.52.5 g 1,2-bis-chloromethylbenzene, 1.05 g bis (triphenylphosphine) palladium (II) chloride and 0.87 g triphenylphosphine are mixed in 100 ml of tert-butanol and in one 500 ml glass autoclave under a protective gas atmosphere (argon) with 48.0 g water and 46.8 g of calcium hydroxide suspended.
The temperature is then raised to 70 ° C. under a carbon monoxide atmosphere elevated. The carbon monoxide uptake started with vigorous stirring at 60 ° C is finished after 2 hours and you get a viscous porridge. The addition of a further 1.05 g of bis (triphenylphosphine) palladium (II) chloride and 0.87 g Triphenylphosphine leads to no further carbon monoxide uptake.
extrahiert. Man erhält 383 g Wasserphase und 299 g organische Phase. Die
Wasserphase enthält nach gaschromatographischer Analyse kein Isochroman-3-on,
Isochroman-3-on (Ausbeute = 24 %; TON = 46;
