Abstract
Substituierte Pyrroloanthracene und -dione der allgemeinen Formel (I), in welcher E und L gemeinsam einen heterocyclischen Rest der Formel (II) bilden, werden hergestellt, indem man entweder Furanoanthracene oder N-unsubstituierte Pyrroloanthracen mit entsprechend substituierten Alkylaminen umsetzt. Die Verbindungen können als Wirkstoffe in Arzneimitteln, insbesondere als Immunmodulatoren, eingesetzt werden.
Description
SUBSTITUIERTE PYRROLOANTHRACENE UND -DIONE ALS IMMUNMODULATOREN
Die vorliegende Erfindung betrifft substituierte Pyrroloanthracene und -dione, Verfahren zu ihrer Herstellung und ihre Verwendung in Arzneimitteln.
Aus der Publikation US 48 04-751 A sind polycyclische Dicarbonsäureimide mit einer antipsychotischen und anxiolytischen Wirkung bekannt. In den meisten Theorien über Autoimmunerkrankungen werden T-Lymphocyten als Initiatoren vieler zellulärer Prozesse benannt, die zu Gewebezerstörung und zu Symptomen führen, die mit spezifischen Autoimmunerkrankungen verbunden sind [Paul, E.W. 1984, Fundamental Immunology, Ravens Press, New York). Die rheumatoide Arthritis ist ein Beispiel für solche Erkrankungen und die Adjuvans-induzierte Arthritis in der Ratte wird als ein repräsentatives Tiermodell dafür angesehen [Lombardino, J.G. 1985, Nonsteroidal Antiinflammatory Drugs, John Wiley & Sons, New York]. Für das Ratten Adjuvans Arthritis Modell wurde anhand einer Vielzahl von Studien gezeigt, daß T-Zellen am Fortschreiten des Krankheitsverlaufs beteiligt sind. Außerdem übertragen T-Zellen von Ratten mit Adjuvans Erkrankung, diese Erkrankung auf gesunde Tiere in Abwesenheit von i ndeiner Quelle von Antigen oder anderen entzündlichen Faktoren.
F ich sollte die Inhibition der T-Zellen-Funktion oder der T-Zellen Aktivierung so hl die Adjuvans Arthritis in der Ratte als auch den Krankeitsverlauf verschiedener menschlicher Autoimmunerkrankungen positiv beeinflussen.
Kürzlich wurde ein Serotonintyp-Rezeptor auf Jurkart Zellen identifiziert [Aune, T., M. Kelley, U.A. Ranges, G.E. Bombera, M.P. 1990, J. Immunol. 145, 1826] von
dem angenommen wird, daß er die T-Zellen-Funktion reguliert. Deshalb wird angenommen, daß selektive Antagonisten dieses Rezeptors die T-Zell-Proliferation inhibieren.
Die vorliegende Erfindung betrifft substituierte Pyrroloanthracene und -dione der allgemeinen Formel (I)
in welcher
A und D gleich oder verschieden sind und für Wasserstoff, Hydroxy, Halogen, Cyano, Carboxy, Nitro, Trifluormethyl, Trifluormethoxy stehen, oder für geradkettiges oder verzweigtes Alkyl oder Alkoxy mit jeweils bis zu 8 Kohlenstoffatomen stehen,
R1 und R2 gleich oder verschieden sind und für Wasserstoff, Halogen, Cyano, Formyl, Phenyl oder Hydroxy stehen, oder
für geradkettiges oder verzweigtes Alkoxy mit bis zu 8 Kohlenstoffatomen stehen, oder
für geradkettiges oder verzweigtes Alkyl oder Alkenyl mit jeweils bis zu 8 Kohlenstoffatomen stehen, die gegebenenfalls bis zu 2-fach gleich oder verschieden durch Hydroxy, Nitro, Phenyl, Halogen, durch geradkettiges oder verzweigtes Alkoxy mit bis zu 6 Kohlenstoffatomen oder durch eine Gruppe der Formel -NR3R4 substituiert sind, worin

und deren Salze.
Im Rahmen der vorliegenden Erfindung werden physiologisch unbedenkliche Salze bevorzugt. Physiologisch unbedenkliche Salze der substituierten Pyrroloanthracene und -dione können Salze der erfindungsgemäßen Stoffe mit Mineralsäuren, Carbonsäuren oder Sulfonsäuren sein. Besonders bevorzugt sind z.B. Salze mit Chlorwasserstoffsäure, Bromwasserstoffsäure, Schwefelsäure, Phosphorsäure, Methansulfonsäure, Ethansulfonsäure, Toluolsulfonsäure, Benzolsulfonsäure, Naphthalindisulfonsäure, Essigsäure, Propionsäure, Milchsäure, Weinsäure, Zitronensäure, Fumarsäure, Maleinsäure oder Benzoesäure.
Salze im Rahmen der vorliegenden Erfindung sind außerdem Salze der einwertigen Metalle wie Alkalimetalle und die Ammoniumsalze. Bevorzugt werden Natrium-, Kalium- und Ammoniumsalze.
Die erfindungsgemäßen Verbindungen existieren in stereoisomeren Formen, die sich entweder wie Bild und Spiegelbild (Enantiomere), oder die sich nicht wie Bild und
Spiegelbild (Diastereomere) verhalten. Die Erfindung betrifft sowohl die Antipoden als auch die Racemformen sowie die Diastereomerengemische. Die Racemformen lassen sich ebenso wie die Diastereomeren in bekannter Weise in die stereoisomer einheitlichen Bestandteile trennen.

Außerdem wurden Verfahren zur Herstellung der erfindungsgemäßen Verbindungen der allgemeinen Formel (I) gefunden, dadurch gekennzeichnet, daß man [A] Verbindungen der allgemeinen Formel (II)
in welcher
A, D, R1, R2, R5 und R6 die oben angegebene Bedeutung haben, mit Verbindungen der allgemeinen Formel (III)

Die erfindungsgemäßen Verfahren können durch folgende Formelschema beispielhaft erläutert werden:
Als Lösemittel für die Verfahren eignen sich übliche organische Lösemittel, die sich unter den Reaktionsbedingungen nicht verändern. Hierzu gehören bevorzugt Ether wie Diethylether, Dioxan, Tetrahydrofuran, Glykoldimethylether, 1,2-Dimethoxyethan oder Kohlenwasserstoffe wie Benzol, Toluol, Xylol, Hexan, Cyclohexan oder Erdölfrakionen, oder Halogenkohlenwasserstoffe wie Dichlormethan, Trichlormethan, Tetrachlormethan, Dichlorethylen, Trichlorethylen oder Chlorbenzol, oder Essigester, Triethylamin, Pyridin, Dimethylsulfoxid, Dimethylformamid, Hexamethylphosphorsäuretriamid, Acetonitril, Aceton oder Nitromethan. Ebenso ist es möglich, Gemische der genannten Lösemittel zu verwenden. Bevorzugt ist Xylol.
Als Basen für die erfindungsgemäßen Verfahren können im allgemeinen anorganische oder organische Basen eingesetzt werden. Hierzu gehören vorzugsweise Alkalihydroxide oder wie zum Beispiel Natriumhydroxid, Kaliumhydroxid oder Lithiumhydroxid, Bariumhydroxid, Alkali- oder Erdalkalicarbonate wie Natriumcarbonat, Kaliumcarbonat, Calciumcarbonat oder Caesiumcarbonat, oder Alkalioder Erdalkalialkoholate wie Natrium- oder Kaliummethanolat oder Kalium-tert.-butylat, oder Lithiumdiisopropylamid (LDA), oder organische Amine (Trialkyl-(C1-C6)amine) wie Triethylamin, oder Heterocyclen wie 1,4-Diazabicyclo[2.2.2]-octan (DABCO), 1,8-Diazabicyclo[5.4.0]undec-7-en (DBU), Pyridin, Diaminopyridin, Methylpiperidin oder Morpholin. Es ist auch möglich als Basen Alkalimetalle, wie Natrium oder deren Hydride wie Natriumhydrid einzusetzen. Bevorzugt sind Kaliumcarbonat, Natriumhydrid, Kalium-tert.-butylat und Caesiumcarbonat.
Im allgemeinen setzt man die Base in einer Menge von 0,05 mol bis 10 mol, bevorzugt von 1 mol bis 2 mol, jeweils bezogen auf 1 mol der Verbindungen der allgemeinen Formeln (III), (V) und (VII) ein.
Als Hilfsstoffe zur Aktivierung der Carbonsäurefunktion in den Verbindungen der allgemeinen Formel (V) und für die Diels-Alder-Reaktion eignen sich Lewis-Säuren wie beispielsweise Zinkchlorid, Titantetrachlorid, Bortribromid, Aluminiumchlorid oder Lithiumperchlorat oder Thionylchlorid, Phosphortrichlorid, Phosphorpentachlorid, Phosphortribromid oder Oxalylchlorid. Außerdem eignen sich Carbonyl-
Verbindungen wie Carbonyldiimidazol oder 1,2-Oxazoliumverbindungen wie 2-Ethyl-5-phenyl-1,2-oxazolium-3-sulfonat oder Propanphosphorsäureanhydrid oder Isobutylchloroformat oder Benzotriazolyloxy-tris-(dimethylamino)phosphonium-hexylfluorophosphat oder Phosphonsäurediphenylesteramid oder Methansulfonsäurechlorid, gegebenenfalls in Anwesenheit von Basen wie Triethylamin oder N-Ethylmorpholin oder N-Methylpiperidin oder Dicyclohexylcarbodiimid und N-Hydroxysuccinimid.
Die erfindungsgemäßen Verfahren werden im allgemeinen in einem Temperaturbereich von 0°C bis +180°C, bevorzugt von +20°C bis +150°C durchgeführt.
Das erfindungsgemäße Verfahren wird im allgemeinen bei Normaldruck durchgeführt. Es ist aber auch möglich, das Verfahren bei Überdruck oder bei Unterdruck durchzuführen (z.B. in einem Bereich von 0,5 bis 5 bar). Die Reduktion von Carbonylfunktionen erfolgt im allgemeinen mit komplexen Hydriden, wie Lithiumaluminiumhydrid oder Natriumborhydrid, bevorzugt mit Lithiumaluminiumhydrid in inerten Lösemitteln wie Ethern oder Kohlenwasserstoffen oder deren Gemischen, bevorzugt in Ethern wie beispielsweise Diethylether, Tetrahydrofuran oder Dioxan, in einem Temperaturbereich von 0°C bis + 150°C, bevorzugt von +20°C bis + 100°C, bei Normaldruck.
Die oben aufgeführten Derivatisierungen der Substituenten R1, R2, R5, R6, R7, R8, R9, R10 und R11 erfolgen im allgemeinen nach literaturbekannten Methoden, wobei beispielhaft die Reduktion von Aldehyden oder Alkoxycarbonylverbindungen zu Alkoholen (a), die Reduktion von Doppelbindungen (b) und die Alkylierung (c) mit Folgendem erläutert werden sollen: a) Die Reduktion von Carbonylverbindungen zu den entsprechenden Alkoholen erfolgt im allgemeinen mit Hydriden, wie Lithiumaluminiumhydrid oder Natriumborhydrid, bevorzugt im Fall der Alkoxycarbonylverbindungen mit Lithiumaluminiumhydrid und im Fall der Aldehyde bevorzugt mit Natriumborhydrid in inerten Lösemitteln wie Ethern, Kohlenwasserstoffen
oder Alkoholen oder deren Gemischen, bevorzugt in Ethern wie beispiels- weise Diethylether, Tetrahydrofuran oder Dioxan, oder Alkoholen wie Ethanol, im Fall der Aldehyde bevorzugt mit Natriumborhydrid in Ethanol, in einem Temperaturbereich von 0°C bis +150°C, bevorzugt von +20°C bis +100°C, bei Normaldruck.
Die Reduktion einer Doppelbindung erfolgt im allgemeinen durch Hydrierung mit Wasserstoff in Anwesenheit eines Katalysators wie beispielsweise Platin oder Platinoxide, Rhodium, Ruthenium, Chlorotris(triphenylphosphin)rhodium, oder Palladium auf Tierkohle, bevorzugt mit Palladium auf Tierkohle in einem Temperaturbereich von 0°C bis +150°C, bevorzugt von +25°C bis +100°C. b) Als Lösemittel für die Hydrierung eignen sich protische Lösemittel wie beispielsweise Methanol, Ethanol und/oder aprotische Lösemittel wie beispielsweise Tetrahydrofuran, Toluol, Dirne thylformamid, Methylenchlorid,
Dioxan oder Essigester.
Die Hydrierung wird bei einem Druck von 1 bis 300 arm, vorzugsweise bei 1 bis 20 atm, durchgeführt. c) Die Alkylierung erfolgt im allgemeinen in einem der oben aufgeführten Lösemitteln mit Alkylierungsmitteln wie beispielsweise (C1-C8)-Alkylhalogenide, Sulfonsäureester oder substituierte oder unsubstituierte (C1-C8)-Dialkyl- oder (C1-C8)-Diarylsulfate, vorzugsweise Methyljodid, p-Toluolsulfonsäureester oder Dimethylsulfat.
Die Verbindungen der allgemeinen Formel (II) sind teilweise bekannt oder neu und können dann beispielsweise hergestellt werden, indem man Anthrazen oder dessen substituierte Derivate mit Maleinsäureanhydrid oder dessen substituierte Derivate, wie beispielsweise Citraconsäureanhydrid, in einem der oben aufgeführten Lösemitteln, vorzugsweise Toluol oder Xylol, in einem Temperaturbereich von 100 - 150°C, vorzugsweise bei 140°C umsetzt, oder in einem aprotischen Lösemittel,
wie beispielsweise Methylenchlorid, in Anwesenheit einer der oben aufgeführten Lewis-Säuren, beispielsweise Aluminiumchlorid, in einem Temperaturbereich von 0°C bis 80°C, vorzugsweise bei Raumtemperatur bis +40°C umsetzt.
Die Verbindungen der allgemeinen Formeln (III), (V), (VII), (VIII) und (X) sind an sich bekannt oder nach üblichen Methoden herstellbar.
Die Verbindungen der allgemeinen Formel (IX) sind teilweise bekannt oder neu und können dann beschrieben wie oben beschrieben hergestellt werden. Die Verbindungen der allgemeinen Formel (IN) sind ebenfalls teilweise bekannt oder neu und können dann beispielsweise, wie oben für die Verbindungen der allgemeinen Formel (II) beschrieben, durch Umsetzung von Anthrazen oder dessen substituierten Derivaten mit substituierten oder unsubstituierten Succinimiden hergestellt werden.
Die Verbindungen der allgemeinen Formel (VI) sind neu und können beispielsweise wie oben beschrieben hergestellt werden.
Die erfindungsgemäßen Verbindungen der allgemeinen Formel (I) zeigen überraschenderweise eine immunmodulierende Wirkung.
Sie sind somit geeignet zur Behandlung von rheumatischer Arthritis, Multipler Sklerose, Muskelschwäche, Lupus erythematodes disseminatus, Basedow-Krankheit, Psoriasis, Transplantatabstoßung, Hashimoto's, Schilddrüsenentzündung, entzündlichen Darmerkrankungen, Entzündungen des Gastrointestinaltraktes, primäre Leberzirrhose und autoimmuner hämolytischer Anämie.
Adjuvans Arthritis Modell in der Ratte Freundsches Adjuvans (FA) wird durch Gabe von 10 mg/ml von in der Hitze abgetötetem Mycobacterium butyricum in extra schwerem Mineralöl hergestellt. Lewis Ratten erhalten eine 0,1 ml Injektion von FA (1 mg/ Tier) subcutan in die
rechte Hinterpfote. Dort findet eine akute Entzündung statt, die sich durch Rötung, Ödeme und eine vorherrschende neutrophile Zellinfiltration auszeichnet (Primär-Reaktion). Anschwellende Gebiete in der behandelten Pfote am Tag 5 werden zwischen dem Tag 10 - 12 gefolgt von einer Zunahme der Schwellung (chronische Entzündungsreaktion) und dem Auftreten einer Schwellung in der gegenüberliegenden unbehandelten Pfote (Sekundäre Immunantwort).
Die Zellinfiltration der chronischen Entzündung und die zweite Immunantwort sind überwiegend mononuclear, was die Anwesenheit von zellgesteuerter Immunisierung anzeigt. Die Tiere werden täglich beobachtet, und die Schwellung wird an den Tagen 12 und 16 in Millimetern mit einem per Hand einstellbaren Mikrometer gemessen. Die Schwellung erreicht ihren Höhepunkt am 16. Tag, an dem die Tiere getötet werden und Gewebeproben für die histologische Auswertung entnommen werden. Das Ausmaß der Schwellung wird durch Berechnung der größten Differenz zwischen dem 16. und dem 0. Tag des Fußknöcheldurchmessers ermittelt.
Die Tiere erhalten die erfindungsgemäßen Verbindungen in einer Suspension von 5% Polyethylenglykol und 0,5% Tween 80 in Phosphat-Pufferlösung p.o. oder i.p. an den Tagen 0, 1, 2, 5, 7, 9, 12 und 14 [vgl. hierzu L. Sokoloff, 1984, Int. Rev. Exp. Pathol. 26, 107; M.E.J. Bittingham et al., 1989, J. Exp. Med. 171, 339; K.M. Connolly et al., 1989, Agents and Actions 27, 328].
Die in vitro Aktivität der erfindungsgemäßen Verbindungen ergibt sich aus ihrer Fähigkeit, die T-Zellen Proliferation zu inhibieren, die vorher durch Serotonin stimuliert wird.
Inhibition der 5-HT abhängigen T-Zellen Proliferation Periphäre mononukläre Blutzellen (PBMC) werden aus der Speckschicht (butty coat) aus dem Blut von gesunden Probanden erhalten. Nach Isolierung mittels Isolymphgradienten (Pharmacia), werden die PBMC zweimal gewaschen und direkt
verwendet. Gereinigte T-Zellen werden durch E-Rossettenbildung mit Schaferythrozyten [vgl. Blue, M.-L., J..F. Daley, H. Levine, K.A. Craig and S.F. Schlossman, 1986, Biosynthesis and surface expression of T8 by peripheral blood T4+ cells in vitro, J. Immunol. 137, 1202] erhalten. Monocyten werden teilweise durch Adhäsion an Gewebeplättchenkulturen entfernt. PBMC, T-Zellen oder gereinigte T-Zellen (von Monocyten verarmt) mit 5 × 105/ml, 100 μl/Loch werden in RPMI-1640 Medium mit 10% fetalem Kälberserum (GIBCO) und L-Glutamin in 96-Loch-Mikrokulturplatten (Becton-Dickinson) unter einer 5% CO2 Atmosphäre bei 37°C für 7 Tage kultiviert. Die Kulturen werden am 7.Tag für 6 h mit 1 μCi 3H-Thymidin versetzt, auf Filterpapier gesammelt, und die inkorporierte Radioaktivität mittels Flüssigkeitsszintillationszählung bestimmt.
Die Testsubstanzen werden in 10 mM HCl gelöst, um jeweils eine Endkonzentration von 1 mM zu erreichen. Die Verbindungen werden serienmäßig 3 × mit Medium versetzt, um eine Endkonzentration zwischen 33 μM - 0,1 μM zu erhalten. Monocytenfreien menschlichen T-Zellen wird PWM (1:200, Poke Weed Mitogen) und 5-HT (100 μM, Serotonin), die die Testsubstanzen enthalten, zugefügt. Die positive Kontrolle (maximale Proliferation) erhält man aus Kulturen mit T-Zellen, PWM und 5-HT. Die negative Kontrolle (minimale Proliferation) erhält man aus Kulturen mit T-Zellen und PWM ohne 5-HT. Die inhibitorische Aktivität der Testsubstanzen werden als IC50 in μM ausgedrückt,

Die neuen Wirkstoffe können in bekannter Weise in die üblichen Formulierungen überführt werden, wie Tabletten, Dragees, Pillen, Granulate, Aerosole, Sirupe, Emulsionen, Suspensionen und Lösungen, unter Verwendung inerter, nicht-toxischer, pharmazeutisch geeigneter Trägerstoffe oder Lösemittel. Hierbei soll die therapeutisch wirksame Verbindung jeweils in einer Konzentration von etwa 0,5 bis 90-Gew.-% der Gesamtmischung vorhanden sein, d.h. in Mengen, die ausreichend sind, um den angegebenen Dosierungsspielraum zu erreichen.
Die Formulierungen werden beispielsweise hergestellt durch Verstrecken der Wirkstoffe mit Lösemitteln und/oder Trägerstoffen, gegebenenfalls unter Verwendung von Emulgiermitteln und/oder Dispergiermitteln, wobei z.B. im Fall der Benutzung von Wasser als Verdünnungsmittel gegebenenfalls organische Lösemittel als Hilfslösemittel verwendet werden können. Die Applikation erfolgt in üblicher Weise, vorzugsweise oral oder parenteral, insbesondere perlingual oder intravenös.
Für den Fall der parenteralen Anwendung können Lösungen des Wirkstoffs unter Verwendung geeigneter flüssiger Trägermaterialien eingesetzt werden.
Im allgemeinen hat es sich als vorteilhaft erwiesen, bei intravenöser Applikation Mengen von etwa 0,001 bis 1 mg/kg, vorzugsweise etwa 0,01 bis 0,5 mg/kg Körpergewicht zur Erzielung wirksamer Ergebnisse zu verabreichen, und bei oraler Applikation beträgt die Dosierung etwa 0,01 bis 20 mg/kg, vorzugsweise 0,1 bis 10 mg/kg Körpergewicht.
Trotzdem kann es gegebenenfalls erforderlich sein, von den genannten Mengen abzuweichen, und zwar in Abhängigkeit vom Körpergewicht bzw. der Art des Applikationsweges, vom individuellen Verhalten gegenüber dem Medikament, der Art von dessen Formulierung und dem Zeitpunkt bzw. Intervall, zu welchem die Verabreichung erfolgt. So kann es in einigen Fällen ausreichend sein, mit weniger als der vorgenannten Mindestmenge auszukommen, während in anderen Fällen die genannte obere Grenze überschritten werden muß. Im Falle der Applikation größerer Mengen kann es empfehlenswert sein, diese in mehreren Einzelgaben über den Tag zu verteilen.
Ausgangsverbindungen
Beispiel I
1,3-Dioxo-4,6-etheno-perhydro-cycloprop[f]isoindol
Zu einer Lösung von 15 g (0,163 mol) Cycloheptatrien in 80 ml Dichlorbenzol werden portionsweise 15,8 g (0,163 mol) Maleinsäureimid gegeben. Die Suspension geht beim Aufheizen auf 160-170°C in Lösung und fällt nach ca 10 min erneut aus. Nach 6 h Rühren bei 160°C wird die Suspension auf Raumtemperatur abgekühlt und die cremefarbenen Kristalle mit Petrolether abgesaugt und gewaschen.
Schmelzpunkt: 231-233°C
Ausbeute: 25,7 g (83,5% d.Th.)
Beispiel II 4,6-Etheno-perhydro-cycloprop[f]isoindol
Zu einer auf 90°C heißen, kräftig gerührten Suspension von 6 g (0,16 mol) Lithiumaluminiumhydrid gibt man unter Argon innerhalb von 20 Minuten portionsweise 10 g (0,053 mol) der Verbindung aus Beispiel I. Die Suspension wird nach 6 h Rückfluß (100°C) auf Raumtemperatur abgekühlt, tropfenweise mit 6 ml H2O und anschließend mit 18 ml verd. Kalilauge versetzt. Die Suspension wird 10 min bei Rückflußtemperatur gekocht, heiß abgesaugt und mit Dioxan nachge
waschen. Der Niederschlag wird verworfen, die Mutterlauge auf Raumtemperatur abgekühlt und am Rotationsverdampfer bei 50°C / 20 mbar eingeengt. Der ölige Rückstand wird mit Methylenchlorid extrahiert, die organische Phase 2 mal mit Wasser gewaschen und über Natriumsulfat getrocknet. Die organische Phase wird nun am Rotationsverdampfer bei ca. 35°C / 20 mbar eingeengt. Das zurückbleibende Öl wird bei 140- 150°C / 0, 1 mbar destilliert.
Ausbeute: 5,4 g (63,5% d.Th.)
Beispiel III 3-(4,6-Etheno-perhydro-cycloprop[f]isoindol-2-yl)-propionitril
3 g (0,018 mol) der Verbindung aus Beispiel II werden in 10 ml Toluol gelöst. Unter Rühren und Eiskühlung werden innerhalb von 5 Minuten 1,73 g (0,033 mol) Acrylnitril so zugetropft, daß die Temperatur unter 30°C gehalten wird. Die Lösung wird 24 h bei Raumtemperatur gerührt und am Rotationsverdampfer bei 35°C / 20 mbar eingeengt. Das zurückbleibende Öl wird mittels Säulenchromatographie mit Methylenchlorid / Methanol 50: 1 gereinigt.
Ausbeute: 3,6 g (91,4%)
Beispiel IV 3-(4,6-Etheno-perhydro-cycloprop[f]isoindol-2-yl)-propylamin
Die Hydrierung von 3,6 g (0,01696 mol) der Verbindung aus Beispiel III erfolgt im Stahlautoklaven in 50 m Methanol und 225 ml flüssigem Ammoniak unter Zusatz von 2 g Raney-Nickel bei 100 -120 bar und 80°C in ca. 4 Stunden. Nach Absaugen des Katalysators wird mit Methanol nachgewaschen und die Mutterlauge am Rotationsverdampfer bei ca. 35°C / 20 mbar eingeengt. Man erhält 3,5 g eines klaren Öls.
Ausbeute: 3,5 g (94,6% d.Th.)
Beispiel V 9,10-Dihydro-9,10[3',4']-furanoanthracen-12,14(11H,15H)-dion
180 g (1,01 mol) Anthrazen werden zusammen mit 100 g (1,02 mol) Maleinsäureanhydrid in 1000 ml Xylol suspendiert. Die Suspension geht beim Aufheizen auf 140°C in Lösung und fällt nach 10 min Kochen unter Rückfluß erneut aus. Nach Abkühlen auf Raumtemperatur werden die ausgefallenen cremefarbenen Kristalle mit Xylol und Ether gewaschen und getrocknet.
Schmp.: 261-263°C
Ausbeute: 259,9 g (92,8%)
Herstellungsbeispiele
Beispiel 1
13-[3-(4,6-Etheno-perhydro-cycloprop[f]isoindol-2-yl)-propyl]-9,10-dihydro-9,10-[3 ',4']-pyrroloanthracen-12,14[11H,15H)-dion
1,5 g (0,0069 mol) der Verbindung aus Beispiel IV werden zusammen mit 1,90 g (0,0069 mol) der Verbindung aus Beispiel V in 50 ml Xylol suspendiert und 1,5 Stunden am Wasserabscheider gekocht. Die Lösung wird auf Raumtemperatur abgekühlt und am Rotationsverdampfer bei ca. 50°C / 20 mbar eingeengt. Das zurückbleibende Öl wird mit Ether / Petrolether verrieben, der kristalline Niederschlag abgesaugt und mittels Säulenchromatographie an Kieselgel mit Methylenchlorid / Methanol im Verhältnis 3:1 als Laufmittel gereinigt.
Schmelzpunkt: 134-136°C
Ausbeute: 2,14 g (65,3% d.Th.)
In Analogie zur Vorschrift des Beispiels 1 werden die in den Tabellen 1 und 2 aufgeführten Verbindungen hergestellt:
Beispiel 8 13-[3-(4,6-Etheno-perhydro-cycloprop[f]isoindol-2-yl)-propyl]-9,10,11,12,14,15- hexahydro-9,10[3',4']pyrroloanthracen
In einem ausgeheizten Kolben werden unter Argon 100 ml absolutes Dioxan vorgelegt und unter Rühren 0,29 g (0,0075 mol) Lithiumaluminiumhydrid
suspendiert. Nach Aufheizen auf 90°C werden innerhalb von 10 Minuten 1,2 g (0,0025 mol) der Verbindung aus Beispiel 1, gelöst in 20 ml Dioxan, zugetropft. Anschließend wird die Suspension 6 h unter Rückfluß gekocht. Nach Abkühlen auf Raumtemperatur werden nacheinander unter kräftigem Rühren 0,3 ml Wasser und 0,9 ml verdünnte Kalilauge zugetropft, 10 Minuten unter Rückfluß gekocht, der voluminöse Niederschlag heiß abgesaugt und mit Dioxan nachgewaschen. Die Mutterlauge wird auf Raumtemperatur abgekühlt und am Rotationsverdampfer bei ca. 40°C / 20 mbar eingegengt. Der ölige Rückstand wird mit Methylenchlorid /Wasser versetzt, 2 × mit Methylenchlorid extrahiert, die organische Phase wird 2 × mit Wasser gewaschen und über Natriumsulfat getrocknet und am Rotationsverdampfer bei 35°C / 20 mbar eingeengt Der zurückbleibende Rückstand wird mit Methanol / verdünnte Ammoniaklösung (90:10) als Laufmittel an Kieselgel gereinigt.
Schmelzpunkt: 108-110°C
Ausbeute: 0,412 g (36,7% d.Th.)
In Analogie zur Vorschrift des Beispiels 8 werden die in den Tabellen 3 und 4 aufgeführten Verbindungen hergestellt:

1. Substituierte Pyrroloanthracene und -dione der allgemeinen Formel (I)
in welcher
A und D gleich oder verschieden sind und für Wasserstoff, Hydroxy, Halogen, Cyano, Carboxy, Nitro, Trifluormethyl, Trifluormethoxy stehen, oder
für geradkettiges oder verzweigtes Alkyl oder Alkoxy mit jeweils bis zu 8 Kohlenstoffatomen stehen,
R1 und R2 gleich oder verschieden sind und für Wasserstoff, Halogen, Cyano, Formyl, Phenyl oder Hydroxy stehen, oder
für geradkettiges oder verzweigtes Alkoxy mit bis zu 8 Kohlenstoff- atomen stehen, oder

Ganz besonders bevorzugt sind Verbindungen der allgemeinen Formel (I) (subst. Pyrroloanthracene und -dione) nach Anspruch 1, worin A, D, R1 und R2 für Wasserstoff stehen.
4. Substituierte Pyrroloanthracene und -dione nach Anspruch 1 bis 3 zur therapeutischen Anwendung.
5. Verfahren zur Herstellung von substituierten Pyrroloanthracenen und -dionen nach Anspruch 1 bis 3, dadurch gekennzeichnet, daß man
[A] Verbindungen der allgemeinen Formel (II)
in welcher
A, D, R1, R2, R5 und R6 die oben angegebene Bedeutung haben, mit Verbindungen der allgemeinen Formel (III)
H2N-T-V (III), in welcher T und V die oben angegebene Bedeutung haben, in inerten Lösemitteln, in Anwesenheit einer Base und/oder Hilfsmittels, gegebenenfalls unter Schutzgasatmosphäre umsetzt, oder
[B] Verbindungen der allgemeinen Formel (IV)
in welcher
A, D, R1, R2, R5 und R6 die oben angegebene Bedeutung haben, zunächst durch Umsetzung mit Verbindungen der allgemeinen Formel (V)
X-T-Y (V), in welcher
X für Halogen oder für eine typische Abgangsgruppe, vorzugsweise aber für Brom steht,
Y die oben angegebene Bedeutung von X hat oder für Carboxy oder einen aktivierten carbonylischen Rest steht, und

6. Arzneimittel enthaltend mindestens ein substituiertes Pyrroloanthracen und -dion nach Anspruch 1 bis 3.
7. Arzneimittel nach Anspruch 6 zur Immunmodulation.
8. Verfahren zur Herstellung von Arzneimitteln nach Anspruch 6 und 7, dadurch gekennzeichnet, daß man die substituierten Pyrroloanthracene und -dione gegebenenfalls mit Hilfs- und Trägerstoffen in eine geeignete Applikationsform überführt.
9. Verwendung von substituierten Pyrroloanthracenen und -dionen nach Anspruch 1 bis 3 zur Herstellung von Arzneimitteln.
10. Verwendung von substituierten Pyrroloanthracene und -dionen nach Anspruch 1 bis 3 zur Herstellung von Immunmodulatoren.
