Появление селективных агонистов I1 имидазолиновых рецепторов привело ко второму рождению класса антигипертензивных препаратов центрального действия (или симпатолитиков) в лечении артериальной гипертонии (АГ). При этом официально зафиксирован факт возврата симпатолитиков в кардиологическую практику после долгого перерыва. Препараты этой группы были одними из первых антигипертензивных препаратов, которые начали применяться в клинической практике около 40 лет назад, поскольку в патогенезе АГ симпатической нервной системе придавалось важное значение еще со времен нейрогенной теории Г.Ф. Ланга. Актуальность создания таких препаратов еще более возросла, когда выяснилось, что активация симпатической нервной системы приводит к повышению артериального давления (АД) и играет роль в возникновении ряда других негативных эффектов, в том числе метаболических нарушений, которые значительно увеличивают риск развития осложнений у лиц с АГ. Среди этих эффектов достаточно назвать гипертрофию миокарда, дисфункцию эндотелия, активацию тромбоцитов, инсулинорезистентность и дислипидемию. Таким образом, одними из важнейших направлений фармакотерапии АГ являются одновременное снижение активности симпатоадреналовой системы, коррекция метаболических нарушений и органопротекция. С открытием имидазолиновых рецепторов и созданием его селективных агонистов появление новых эффективных и безопасных симпатолитиков стало реальностью. Французскими учеными было установлено, что имидазолиновые рецепторы находятся в двух важнейших органах регуляции АД - головном мозге и почках. Они расположены в боковых ретикулярных ядрах рострального отдела продолговатого мозга и в проксимальных канальцах почек. Оказалось, что указанные структуры не реагируют на катехоламины, а реагируют на химические соединения, сходные с имидазолином. Именно поэтому эти рецепторы были названы имидазолиновыми. It turned out that these structures do not react to catecholamines, but react to chemical compounds similar to imidazoline. That is why these receptors were called imidazoline. В исследование, в котором изучалась эффективность моксонидина, был включен 41 пациент с АГ 1-3 степени (III степень риска). С целью оценки активности всем больным до и после лечения симпатоадреналовой системы дополнительно к стандартному исследованию биохимических показателей крови (уровень гликемии, HbA1c, липидный спектр крови) проводили двойной динамический тест (ДДТ) на катехоламины (дофамин, норадреналин, адреналин). Также у всех больных исследовали уровень лептина. После 8-недельного лечения моксонидином достижение целевого уровня АД сопровождалось достоверным снижением уровня гормонов стресса, индекса массы тела, инсулинорезистентности и концентрации лептина. Следует отметить, что на фоне лечения было зафиксировано смещение липидного спектра крови в сторону антиатерогенности достоверное снижение триглицеридов и увеличение холестерина липопротеинов высокой плотности(ЛПВП). Of particular interest is the analysis of data on the metabolic effects of AIR. The most convincing results were obtained in studies by Haenni A. et al. using the euglycemic clamp test method. Moxonidine has been found to reduce insulin resistance. A Russian study conducted at the State Scientific Research Center for Medical Sciences, which included patients with mild and moderate hypertension and compensated type 2 diabetes, also found a positive effect of moxonidine on insulin resistance. After 3 months of treatment with moxonidine, there was a significant decrease in insulin and blood glucose levels measured 2 hours after a standard breakfast (equivalent to a glucose tolerance test). These results indicate an improvement in tissue sensitivity to insulin, since less insulin is required to maintain lower glucose levels than before treatment after treatment with moxonidine. Проведенный генетический анализ показал, что частота и динамика возникновения РДЛ в линии ВЭС полностью определяется генотипом самки. During the selection process, a series of lines differing in the selected traits (BA-, HA-, HA+) were repeatedly obtained using reciprocal selection. This selection led to the acquisition by low-active lines of a complex of genetically controlled changes, the most important of which affected the neuroendocrine system of flies, which became the subject of special research). In the lines established from the natural population of LR, selection was carried out for embryonic mortality, accompanied by close inbreeding. As a result, contrasting inbred lines were obtained: with high (VES line) and low embryonic mortality (NES line). In parallel, a sample of flies from the natural population of the Republic of Latvia was maintained without selection in mass cultures. It was found that the WES and LR lines are characterized by threefold differences in the number of viable offspring, without differing in fertility, determined by the number of eggs laid per unit of time. In the VES line, 81% of eggs stop developing already at the early stages of ontogenesis, that is, this line is characterized by a high frequency of early dominant lethalities (EDL), and it increases from 65% on the first day of oviposition to 95% on the fourth. There were no differences between the studied lines in terms of late dominant lethality. The genetic analysis showed that the frequency and dynamics of the occurrence of RDF in the VES line is completely determined by the genotype of the female. In addition, it was discovered that in the VES line, by the 86th generation of directed selection, a system of balanced lethal mutations had arisen, creating permanent heterozygosity in a small region of the second chromosome.

Наиболее эффективными соединениями-геропротекторами следует выделить вещества: IIIh; IXa; IXb; XIa; XIIIh; XVa; XVg; XVh; XVi, которые продлевали жизнь дрозофил до 50 и более дней, тогда как в контроле этот показатель был 21-26 дней при ожидаемом значении 30 дней.
