Для лечения воспалительных процессов и изменений дистальных отделов толстой кишки, например язвенного колита, необходима лекарственная форма, которая после ее перорального приема обеспечивает локальную доступность действующего вещества, которым является будесонид, в достаточной высокой концентрации в месте воспаления. Такая концепция, называемая также нацеливанием на толстую кишку, нереализуема с использованием традиционной фармацевтической композиции для перорального приема, поскольку существует высокий риск того, что невозможно обеспечить присутствие действующего вещества в эффективных концентрациях в месте его действия, т.е. в патологически измененных отделах толстой кишки. Поэтому проблема нацеливания на толстую кишку преимущественно должна решаться путем использования композиции с модифицированным высвобождением действующего вещества, которое максимально полно высвобождается из нее в толстой кишке и может оказывать в ней свое действие. The present invention is particularly suitable for the preparation of a dosage form that is simple and reliable in administration and satisfies these requirements.

Наряду с предлагаемой в изобретении композицией испытывали другие опытные образцы содержащей будесонид лекарственной композиции. Схематично показанная на фиг. 2 композиция с условным обозначением BUX-PVII позволяет оптимально реализовать принцип нацеливания на толстую кишку. Достигаемый профиль высвобождения действующего вещества in vivo должен после фазы задержки обеспечивать его высвобождение равномерно на протяжении более продолжительного периода времени при одновременно низком уровне в сыворотке и плазме. Средние концентрации будесонида в плазме измеряли после однократного приема содержащих его пеллет во времени. На фиг. 6 в графическом виде представлены результаты для двух контрольных композиций (BUX-E и контроль) и для предлагаемой в изобретении композиции (BUX-PVII).The results obtained in this example are presented graphically in Fig. 6. In fig. 6 graphically presents the results for two control compositions (BUX-E and control) and for the composition proposed in the invention (BUX-PVII).
