Claims (20)
1. Isoleret hybridpolypeptid med cellobiohydrolaseaktivitet, hvilket hybridpolypeptid omfatter: (a) et første polypeptidfragment, som omfatter et katalytisk i cellobiohydrolasedomæne, i den N-terminale ende af hybridpolypeptidet valgt fra gruppen bestående af (i) et polypeptidfragment, der udviser mindst 96% sekvensidentitet med aminosyrerne 19 til 455 ifølge SEQ ID NO: 2; (ii) et polypeptidfragment, som kodes for af et polynukleotid, der udviser mindst 96% sekvensidentitet med nukleotiderne 55 til 1485 ifølge SEQ ID NO: 1 eller cDNA-sekvensen deraf; og (iii) et polypeptidfragment, som omfatter eller består af aminosyrerne 19 til 455 ifølge SEQ ID NO: 2 eller et fragment deraf med cellobiohydrolaseaktivitet; og (b) et andet polypeptidfragment, som omfatter et carbohydratbindingsdomæne i den C-terminale ende af det første polypeptidfragment valgt fra gruppen bestående af (i) et polypeptidfragment, der udviser mindst 96% sekvensidentitet med aminosyrerne 485 til 529 ifølge SEQ ID NO: 4; (ii) et polypeptidfragment, som kodes for i af et polynukleotid, der udviser mindst 96% sekvensidentitet med nukleotiderne 1453 til 1587 ifølge SEQ ID NO: 3; og (iii) et polypeptidfragment, som omfatter eller består af aminosyrerne 485 til 529 ifølge SEQ ID NO: 4 eller et fragment deraf med carbohydratbindingsaktivitet.
2. Hybridpolypeptid ifølge krav 1, hvor det første polypeptidfragment omfatter eller består af aminosyrerne 19 til 455 ifølge SEQ ID NO: 2 eller et fragment deraf med cellobiohydrolaseaktivitet.
3. Hybridpolypeptid ifølge krav 1, hvor det andet polypeptidfragment omfatter eller består af aminosyrerne 485 til 529 ifølge SEQ ID NO: 4 eller et fragment deraf med carbohydratbindingsaktivitet.
4. Hybridpolypeptid ifølge krav 1, som omfatter eller består af aminosyrerne 19 til 455 ifølge SEQ ID NO: 2 som det første polypeptidfragment i den N-terminale ende af hybridpolypeptidet og aminosyrerne 485 til 529 ifølge SEQ ID NO: 4 som det andet polypeptidfragment i den C-terminale ende af det første polypeptidfragment.
5. Hybridpolypeptid ifølge krav 1, som omfatter eller består af det modne polypeptid ifølge SEQ ID NO: 6 eller et fragment deraf med cellobiohydrolaseaktivitet og carbohydratbindingsaktivitet.
6. Hybridpolypeptid ifølge et hvilket som helst af kravene 1-5, som endvidere omfatter et signalpeptid i den N-terminale ende af det første polypeptidfragment.
7. Hybridpolypeptid ifølge krav 6, hvor signalpeptidet er signalpeptidet ifølge SEQ ID NO: 2.
8. Hybridpolypeptid ifølge krav 7, hvor signalpeptidet er aminosyrerne 1 til 21 ifølge SEQ ID NO: 2.
9. Isoleret polynukleotid, som koder for hybridpolypeptidet ifølge et hvilket som helst af kravene 1-8.
10. Værtscelle, som omfatter polynukleotidet ifølge krav 9.
11. Fremgangsmåde til fremstilling af et hybridpolypeptid med cellobiohydrolaseaktivitet, hvilken fremgangsmåde omfatter: (a) dyrkning af værtscellen ifølge krav 10 under betingelser, som er egnede til ekspression af hybridpolypeptidet; og eventuelt (b) indvinding af hybridpolypeptidet.
12. Transgen plante, plantedel eller plantecelle, som er transformeret med polynukleotidet ifølge krav 9, der koder for et hybridpolypeptid.
13. Fremgangsmåde til fremstilling af hybridpolypeptidet ifølge et hvilket som helst af kravene 1-8, hvilken fremgangsmåde omfatter: (a) dyrkning af en transgen plante eller en plantecelle, som omfatter et polynukleotid, hvilket polynukleotid koder for hybridpolypeptidet, under betingelser, der er befordrende for fremstilling af hybridpolypeptidet; og eventuelt (b) indvinding af hybridpolypeptidet.
14. Fremgangsmåde til nedbrydning eller omdannelse af et celluloseholdigt materiale, hvilken fremgangsmåde omfatter: behandling af det celluloseholdige materiale med en enzymsammensætning i nærværelse af hybridpolypeptidet ifølge et hvilket som helst af kravene 1-8.
15. Fremgangsmåde ifølge krav 14, som endvidere omfatter indvinding af det nedbrudte celluloseholdige materiale.
16. Fremgangsmåde til fremstilling af et fermenteringsprodukt, hvilken fremgangsmåde omfatter: (a) forsukring af et celluloseholdigt materiale med en enzymsammensætning i nærværelse af hybridpolypeptidet ifølge et hvilket som helst af kravene 1-8; (b) fermentering af det forsukrede celluloseholdige materiale med en eller flere fermenterende mikroorganismer for at fremstille fermenteringsproduktet; og (c) indvinding af fermenteringsproduktet fra fermenteringen.
17. Fremgangsmåde til fermentering af et celluloseholdigt materiale, hvilken fremgangsmåde omfatter: fermentering af det celluloseholdige materiale med en eller flere fermenterende mikroorganismer, hvor det celluloseholdige materiale er forsukret med en enzymsammensætning i nærværelse af hybridpolypeptidet ifølge et hvilket som helst af kravene 1-8.
18. Fremgangsmåde ifølge krav 17, hvor fermenteringen af det celluloseholdige materiale frembringer et fermenteringsprodukt.
19. Fremgangsmåde ifølge krav 18, som endvidere omfatter indvinding af i fermenteringsproduktet fra fermenteringen.
20. Totalmedieformulering eller celledyrkningssammensætning, som omfatter hybridpolypeptidet ifølge et hvilket som helst af kravene 1-8.
