Den foreliggende opfindelse angår fremstilling af naturligt gpl60 af human immunde-5 fektvirus (HIV), et etiologiske middel i erhvervet immundefektsyndrom (AIDS).
Human immundefektvirus (HIV) er nu fastslået som det etiologiske middel i erhvervet immundefektsyndrom (AIDS). Virusen er tropisk for celler, som basrer CD4 antigenet og er stærkt cytopatisk for hjælper-inducer (T4) celler. Indhylningsgenproduktet af 10 HIV syntetiseres som et gpl60 forstadiemolekyle, som derefter forarbejdes til det ydre ! indhylningsprotein gpl20 og transmembranproteinet gp41. Forstadium/produktslægt- skabsforholdet mellem gpl60 og de mindre proteiner gpl 20 og gp41 er nu blevet godt i ! dokumenteret, og det samme er aminosyresekvenseme af alle tre [Allan med flere, Sci- j ence, 228:1+1094 (1985) og Veronese med flere, Science, 229:1402-1405 (1985)]. Det 15 ydre glycoprotein gpl20 bindes til CD4-molekylet på følsomme celler i indledningsfasen af viral cellefusion og dannelse af kæmpecelle induceret af virusen [Dalgleish med flere, Nature, 312: 723-766].
Foruden deres rolle i celleoverfladereceptorgenkendelse og cellefusion er HIV gpl20 20 og gp41 de vigtigste mål for immungenkendelse i individer inficeret med HIV. Disse proteiner har derfor fået særlig opmærksomhed ved undersøgelser af virusneutralisation og vaccineudvikling. Det er blevet iagttaget, at store segmenter af gpl20 udtrykt ved 1 rekombineret DNA teknik eller naturlig gpl20 renset af HIV inficerede celler udløser for det meste typespecifikke neutraliserende antistoffer i dyr. HIV-indhylningsforstadi-25 umproteinet gpl 60 udtrykt i insektceller med baculovirusvektorer frembragte desuden en stærkt typespecifik immunreaktion i geder [Rusche med flere, PNAS, USA 84:6924-6928(1987)].
Evnen til at inficere visse cellelinier med HIV og til at etablere de inficerede celler i en 30 kontinuerlig producent af intakt virus er blevet beskrevet i US patent nr. 4.652.599.
Selv evnen til at inficere cellelinien og HIV varianten ifølge den foreliggende opfindel- I DK 175733 B1 I se er blevet beskrevet tidligere [Getchell med flere, J. Clin. Microbiol. 23:737-742 I (1986)].
I Ingen af disse begivenheder alene muliggjorde dog skabelse af en fremgangsmåde, der I 5 er i stand til at producere HIV glycoproteinet gpl60 i dets naturlige form. Normalt ned- I brydes naturligt gpl60 til gpl20 og gp41. Indhylningsproteinet, der fås af cellekultur- I medier eller af lyset virus, er derfor gpl 20 og gp41. Det er derfor højst overraskende, at I gpl60 kan fås i sin naturlige form.
10 Glycoprotein gpl60 er kun blevet fremstillet ad rekombineret vej. Rekombineret gpl60 I i er imidlertid forskelligt fra det naturlige gpl60, specielt med hensyn til glycosylering.
I I Disse forskelle bliver afgørende i eftersøgningen efter en HIV vaccine, især fordi ind- I hylningsglycoproteineme af HIV bestemmer viral tropisme og huser epitoper, der er væsentlige for udviklingen af neutraliserende antistoffer mod virussen.

Endnu et formål med opfindelsen er at angive intakt HIV gpl60 i dets naturlige form.
Disse og andre formål og fordele ved opfindelsen opnås ved at dyrke inficeret cellelinie 6D5451 i et serumfrit medium og isolere det naturlige gpl 60 frigjort af cellerne i mediet.

Cellerne blev fotograferet efter 48 timer. For at undersøge virkningerne af de virale glycoproteiner blev CEM celler forinkuberet i 1 time med proteinerne før cokul-tiveringen. Figur 4 viser bindingen af gpl20 og gpl60 med CD4. Konditioneret medium fra en 35S-methioninmærket 6D545, kultur blev klaret ved centrifugering ved 2000 x g efter-20 fulgt at filtrering gennem et 0,45 μ filter. En halv milliliter af mediet blev inkuberet med CEM50 celler i et samlet rumfang på 2 ml som beskrevet nedenfor. De bundne proteiner blev immunudfældet med OKT4 antistof. Spor 1: 0,5 x 106 celler. Spor 2: 1 x 106 celler. Spor 3: 2 x 106 celler. Spor 4: 5 x 106 celler. Spor 5: 10 x 106 celler. Spor 6: 20 x 106 celler.

En enkelt celleklon af HUT78 celler er blevet inficeret med human immundefekt virus type 1 (HIV-1), hvorved den inficerede cellelinie blev en kontinuerlig producent af virus. Klon 6D5 er følsom for kronisk infektion med HIV-1, som beskrevet i Getchell med flere, J. Clin. Microbiol., 23:7^7-742 (1986). Klon 6D5 inficeres med en specifik 30 stamme af HIV-1, HTLV-III^, for at fremstille den inficerede cellelinie 6D5451. Den inficerede cellelinie dyrkes så i serumfrit medium ved at pelletere 6D5451 celler og gen-

suspendere dem i serumfrit medium (såsom HB101 medium, der fås i handelen fra Du I
Pont). Serumfrit medium HB104, der også kan fås fra Du Pont, kan også anvendes til I
udførelse af opfindelsen. I

I den foretrukne udførelsesform indeholder HB101 mediet også vækstsupplementer, I
10 såsom transferrin, insulin og okseserumalbumin. For at understøtte væksten af celler, I
blev cellerne subkultiveret hver 4. dag. 6D5451 cellerne blev dyrket i 2 til 3 generatio- I
ner. Mængden af HIV proteiner frigjort i medierne målt ved ekstracellulær omvendt I
transcriptaseaktivitet var næsten 5 gange større i serumfrit medium end i serumholdigt I
medium. Omvendt transcriptase (RT) i kulturmediet af de inficerede celler blev analy- I
15 seret med (dT) ~5 (A)„ som igangsættende skabelon, som beskrevet i Poiesz med flere, I
PNAS, USA, 77:7415-7419 (1980). I
Det cellefrie medium blev anvendt som kilden til glycoproteinet. Mediet blev indstillet I
til 20 mM med natriumphosphat, pH 7,5, 0,5% Triton X-100, 0,1 mM phenylmethyl- I
20 sulfonylfluorid og 400 mM natriumchlorid. Efter inkubation ved stuetemperatur i 1 I
time, blev mediet koncentreret 30 gange med et Pellicon kassettesystem, der fås i han- H
delen fra Millipore. Fremmede proteiner, der stammede fra mediesupplementet, blev
fjernet fra koncentratet ved immunaffinitetsabsorption (natten over) med et Sepharose- I
bundet gede antistof udviklet mod proteinerne i vækstsupplementet i det serumfri me- H
25 dium. Proteiner, som blev bundet til gedeantistoffer blev fjernet og det ubundne mate- I
riale blev så ført gennem en lectinaffrnitetssøjle, fortrinsvis en lectin-Sepharosesøjle I
(Pharmacia). Selv om brug af en linselectinsøjle foretrækkes, kan der også anvendes I
andre lectiner, som vil genkende mannose, såsom concanavalin-A. Efter vask med salt- I
vand med phosphatstødpude (PBS) blev søjlen elueret med 400 mM -methylmannosid I
30 for at udvinde det viral glycoprotein. Selv om brug af methylmannosidet til at eluere I
søjlen foretrækkes, kan der anvendes enhver mannose, pyranosid eller saccharid, som I
5 DK 175733 B1 konkurrerer med lectinen i affinitetssøjlen. Fig. 1 (spor 1) viser SDS-PAGE profilen af glycoproteinet elueret fra linselectin-Sepharosen. De iøjnefaldende glycoproteiner i prøverne var 120 og 160 kd proteinerne. Disse proteiner reagerede også stærkt i im-munaftrykninger med HIV-1 antistofpositiv human serum. Immunaftrykningsanalyse 5 af HTLV- III451 glycoprotein udføres ved en velkendt fremgangsmåde såsom beskrevet i Samgadharan med flere, Science 224:506-508 (1984). I det væsentlige føres proteinerne på 7% SDS-polyacrylamidgeler og overføres til nitrocellulosestrimler (der fås i handelen). Nitrocellulosestrimleme behandles så med de ønskede antistoffer og af-trykningeme udvikles med peroxidasekonjugerede sekundære antistoffer. Båndene gø-10 res synlige ved reaktion af strimlerne af diaminobenzidin.
gpl60 blev renset af blandingen af glycoproteiner elueret fra linselectin Sepharosesøj-len ved immunaffinitetskromatografi under anvendelse af et monoklonalt antistof til HIV-1 gp41 protein. Det monoklonale antistof blev udviklet under anvendelse af delvis 15 rensede HTLV-III45i glycoproteiner ved standardteknik. Immunoglobulinffaktionen af antistoffet blev koblet til Sepharose efter metoden beskrevet af fabrikanten (Pharmacia). Eluatet fra linselectin Sepharosesøjlen blev bragt i ligevægt ved 4°C med an-ti-HIV-1 gp41 Sepharose i 20 mM tris-HCl, pH 8,5 indeholdende 0,5% Triton X-100, 1M kaliumchlorid og 0,1 mM PMSF. Sepharosen blev så pakket i en søjle, vasket med 20 PBS og det bundne protein blev elueret med 100 mM natriumbicarbonat. HTLV-IH^i gpl60 eluerede fra søjlen i næsten homogen tilstand. Figur 1 (spor 2) viser det rensede gpl60 adskilt ved SDS-PAGE og farvet med coomassie blåt.
Glycoprotein gpl60 og dets derivater fremstillet ifølge opfindelsen kan anvendes på 25 sædvanlig måde i immunterapeutiske og/eller immundiagnostiske metoder og midler. Sådanne behandlingsmetoder og anvendte mængder er velkendt og kan vælges af fagfolk ud fra til rådighed stående metoder og teknikker. F.eks. gpl60 fremstillet ifølge den foreliggende opfindelsen kombineres med et farmaceutisk acceptabelt tilsætningsstof i en mængde, der er effektiv til at give diagnostisk anvendelighed ved en ELISA 30 prøve.

I 30 proteinet. Disse resultater viser at 160 kD proteinet har både gpl20 og gp41 områder af I

Eksempel 3
De naturlige 120 og 160 kD glycoproteiner fremstillet ifølge opfindelsen blev yderligere karakteriseret ved deres reaktionsdygtigheder med antistoffer specifikke for HTLV-5 II1B gpl20 og gp41. Til dette formål blev proteinerne adskilt ved SDS-PAGE, overført til nitrocellulosestrimler og behandlet med antistoffer specifikke for HTLV-IIIB gpl20 og gp41. Både 120 og 160 kD proteinerne reagerede med HIV-1 positiv serum og med ged anti-gpl20. Kun gpl60 kD proteinet reagerede med antistofferne til HTLV-IIIB gp41. Af de to anvendte monoklonale antistoffer til gpl20 reagerede kun et med begge, 10 hvilket viser typespecifikke reaktionsdygtigheder af de monoklonale antistoffer med forskellige isolater af HIV.
Eksempel 4 15 Selv om det primære mål for HIV-1 infektion er hjælper/inducer undersættet af T lymfocytter, som bærer CD4 celleoverflademarkører, er man kun begyndt at forstå den faktiske mekanisme, hvorved virusen inficerer følsomme målceller. Monoklonale antistoffer til visse epitoper af CD4 antigener fandtes at blokere viral infektion og kunne immunudfælde komplekser af CD4 og gp!20. Dette kan antyde, at nøglereaktionen mel-20 lem gpl20 og CD4 igangsætter infektionsprocessen af HTV-1. En konsekvens af virusinfektionen er dannelsen af flerkemede kæmpeceller, der er resultat af cellefusioner. En klon af CEM-celler er blevet påvist at udvise hurtig og kvantitativ syncytiadannelse, når de blandes med HIV-1-inficerede cellelinier [Mathews med flere, PNAS, USA 84:54-5428 (1987)] (fig. 3B). Denne type syncytiadannelse tjener ofte som et mål for 25 gp!20-CD4 reaktion under virusinfektion. Evnen hos HTLV-II1451 glycoprotein til at gribe ind i HTLV-IIIB-induceret fusion af CEM celler blev målt ved, at inkubere mål CEM cellerne med et delvis renset præparat af glycoprotein før blanding med Molt-3/-HTLV-IIIg celler i 36 timer. Forinkubation af CEM celler med glycoproteinpræparater fra uinficerede 6D5 celler havde ingen virkning på syncytiadannelsen (fig. 3C). I mod-30 sætning hertil blokerede forbehandling af CEM cellerne med HTLT-IILui glycoproteinpræparater fuldstændigt syncytiadannelsen induceret af HTLV-fflB/Molt-3-celler
