(54) Lagerbestandigt cholesterinoxidase= præparat.
Den foreliggende opfindelse angår et lagerbestandigt, vandigt chole= sterinoxidasepræparat med et indhold af stødpude og stabiliseringsmiddel.
Den enzymatiske aktivitet af et protein bestemmes af den rumlige anordning af aminosyreresterne i dets polypeptidkæder. Denne "kon~ formation" påvirkes i vandig opløsning af tilstedeværelsen af elek-ω trisk ladede eller uladede uorganiske samt organiske molekyler. Som -j- ^ følge heraf er enzymer i reglen meget følsomme i vandig opløsning UD og lider let et aktivitetstab. Vandige enzympræparater må derfor sta-

fe* J[· 4 %," Med hensyn til virkningen af lavmolekulære uorganiske salte er det kendt, at deres anioniske og kationiske komponenter enten kan indvirke mere stabiliserende eller mere denaturerende på enzymets kon-formation. Sådanne anioner og kationer er med hensyn til deres virkning klassificeret i den såkaldte Hoffmeister-række ("Structure and Stability of biological Macromolecules" af Timasheff og Fasman, forlag Mercel Dekker, Inc., New York, 1969, side 427) . Ifølge Hoffmeisterrækken er anioner og kationer hver især uafhængig af hinanden enten mere stabiliserende eller mere denaturerende. I spidsen af stabiliseringsvirkningen står sulfatanionen og kvaternære ammoniumioner, fulgt af ammoniumionen. Det er derfor let forståeligt, at det overvejende antal af de i handelen værende enzympræparater indeholder ammoniumsulfat som stabiliseringsmiddel alene eller eventuelt med yderligere specifikke stabiliseringsmidler. Også cholesterinoxidase er i de i handelen værende sædvanlige vandige opløsninger hidtil blevet stabiliseret med ammoniumsulfat.
Det har dog vist sig, at stabiliteten af cholesterinoxidase i ammo= niumsulfatopløsning gav anledning til betænkeligheder. Den bedste stabiliseringsvirkning blev opnået med ca. 1 M ammoniumsulfat og aftager ved højere ammoniumsulfatkoncentration. Allerede ved en koncentration på 1 M optræder der dog lejlighedsvis grovkornede fedtede udfældninger af enzymet, som ganske vist stadig er enzymatisk aktive, men som skader doseringen ved aftapningen.
Overraskende har det nu vist sig, at denne vanskelighed kan overvindes, og at der dels helt kan undgås udfældninger og dels opnås en overlegen stabilisering, hvis man som stabiliseringsmiddel anvender natri umchlorid eller kaliumchlorid. i en koncentration mellem 2,0 og 3.5 M, og præparatet har en pH-værdi mellem 4,5 og 8,5.
Opfindelsen angår derfor et lagerbestandigt, vandigt cholesterinoxi= dasepræparat med et indhold af stødpude og stabiliseringsmiddel, som er ejendommeligt ved, at det pr. liter indeholder 2 til 3,5 mol NaCl eller KC1 som stabiliseringsmiddel og har en pH-værdi mellem 4,5 og 8,5. Fortrinsvis indeholder det NaCl i en koncentration mellem 2.5 og 3,2 M.
Den overlegne stabiliseringsvirkning af de nævnte salte kunne ikke 145614 3 forventes, da kalium og natrium ifølge Hoffmeister-rækken stabilise*· rer betydeligt mindre, dvs. inaktiverer stærkere end ammonium, og da også chlorid inaktiverer betydeligt stærkere end sulfat. Hertil kommer, at flavinenzymer, hvortil også cholesterinoxidase hører, som bekendt inakti veres af alkaline talchloride.r yed højere koncentration (Theo= rell, i Acta Chem. Scand. 3, 1954, side 1649).
Fortrinsvis har cholesterinoxidasepræparatet ifølge opfindelsen en pH-værdi mellem 5,0 og 6,5. Stødpudemolariteten ligger i almindelighed mellem 0,01 M og 0,2 M,og fortrinsvis i området 0,01 M til 0,1 M.
Som stødpudestoffer kan anvendes alle salte med puffervirkning inden for det anførte område, f.eks. kaliumphosphatstødpude, natrium= phosphatstødpude, kaliumphosphat/citronsyre-stødpude, natriumacetat= stødpude osv..
Man har ganske vist i enkelte tilfælde lagret enzymer i alkalichlorid= opløsninger. Til dette formål var dog fremgangsmådegrunde afgørende, f.eks. den bedre krystalliseringsevne af papain i natriumchlorid, sammenlignet med ammoniumchlorid eller rensningen af lectiner ved kromatografi i natriumchloridopløsninger, således at man måtte tage en forringet stabilitet med i købet. Til flavinenzymer var en sådan lagringsform dog ikke kendt, da den fører til fraspaltning af den pro-stetiske gruppe ved sure pH-værdier.
Den overlegne lagringsstabilitet af cholesterinoxidasepræparaterne ifølge opfindelsen sammenlignet med cholesterinoxidasepræparater stabiliseret med ammoniumsulfat fremgår af følgende forsøg:
Et parti cholesterinoxidase med et indhold af 450 U/ml enzym i 0,05 M kaliumphosphatstødpude, pH 6, blev indstillet på den i følgende tabel anførte saltkoncentration og holdt i 4 uger ved 4°C henholdsvis +35°C. Den efter forløbet af denne tid konstaterede restaktivitet viser følgende tabel (2-ugersværdi i parentes).

Den overlegne stabilitet af præparatet ifølge opfindelsen gpnås uafhængigt af enzymkoncentrationen af enzympartiet (enkelte partier adskiller sig ofte med hensyn til stabilitet) og er endnu mere udpræget, når der ikke sammenlignes med 1 M ammoniumsulfat, men med 3 M ammoniumsulfat-opløsning.
To forskellige enzympartier blev indstillet på en koncentration på 300 U/ml,.henholdsvis 30 U/ml med den ovenfor anførte stødpude og derefter holdt som ovenfor beskrevet i 4 uger på 35°C. Følgende tabel angiver den efter 4 uger målte restaktivitet. I parentes er anført 2-ugersværdierne.

For at vise, at den overlegne stabilitet af praparatet ifølge opfindelsen er begrænset til et bestemt koncentrationsområde, blev den ovenfor beskrevne ældningsprøve ved 4°C henholdsvis 35°C med et enzymparti ved en koncentration på 30 U/ml udført i 4 uger (2 uger) med to forskellige ammoniumsulfat- og fire forskellige natriumchloridkoncen-trationer. Resultaterne er vist i tabel III.
