Hierbei wird Pantethin mit einem Phosphorsäure-halogenid-diaralkylester bei Abwesenheit von Wasser und in Gegenwart einer tertiärenIt is the aim of the present invention to provide a manufacturing method for a product create at least one of the compounds pantethine-4'-pho'Sphat or pantethein - ^ - phosphate contains. Here pantethine is mixed with a phosphoric acid halide diaralkyl ester in the absence of water and in the presence of a tertiary
ίο organischen Base umgesetzt. That way Pantethine-4'-phosphorsäu.rediaralkylester formed is with an aralkyl group splitting off Treated agent and, if necessary, with an agent, which at least a partial reduction the disulfide group into the mercapto group. The phosphorylation of pantethine is preferred in the presence of pyridine or lutidine or • another, tertiary base of the pyridine series accomplished. Phosphoric acid halide diaralkyl esters, such as Phosphoric acid chloride dibenzyl ester
(C6H5CH2O)2(PO)Cl,(C 6 H 5 CH 2 O) 2 (PO) Cl,
verwendet. The phosphorylation can be carried out within a temperature range from -40 to 0 ° will. When a mixture of. Pantethine-4'-phosphate and pantethine-4'-phosphate desired is, the phosphorylation product, the pantethine-4'-phosphoric acid di-alkyl ester, can be easily Way by the same reactant dearalkylated and partially to the mercapto compound be reduced. This means can be a hydrogen ply acting compound, i. H. one Substance that splits an organic chain with simultaneous addition of hydrogen to both parts, preferably an alkali metal in liquid ammonia. If the product is made entirely from pantethein-4'-phosphate should consist of a mixture of pantethine-4'-phosphate and pantethine-4'-phosphate, as just described, prepared and then subjected to a further reduction until no Pantethine-4'-phosphate remains.
If one only strives for pantethine ^ '- phosphate, said mixture must be oxidized again by z. B. air or oxygen through a aqueous solution of the mixture is blown until a sample of this solution shows no positive mercaptan reaction results in more. The relative proportions of the two products in the mixture produced depend on the working speed after the reduction stage, i. H. of the extent to which the mixture is treated with air or oxygen. One leads therefore to the treatment with alkali metal "in liquid ammonia subsequent stages very quickly, is obtained as End product almost exclusively pantethein-4'-phosphate.
The product can by subsequent barium salt precipitation, silver salt precipitation and repeated Barium salt precipitation are purified.
The pantethine used in the process of the present invention is normally obtained by oxidation of the Pantetheine. A preferred oxidation process is based on that oxygen is obtained by a pantethein solution in a basic solvent can flow. The basic solvent should at least partially consist of a tertiary base which is a member of the pyridine series, for example pyridine itself or Lutidine. Preferred basic solvents are also the alkylenediamines, e.g. B. ethylenediamine in the presence of copper hydroxide.
If one wishes that the product made by the method of the invention more the D - (- | -) - form as the racemic mixture (D, L-form), the starting material, pantethine 1> ζλν. Phosphoric acid dibenzyl ester chloride (from 3.6 g of dibenzyl, 1 mol) was added to the obtained solution under 1 and the resulting mixture was 15 minutes at a temperature of - 40 held 0th After being kept at room temperature for 3-4 hours, the pyridine was removed by distillation under reduced pressure and the residue was dissolved in chloroform. The chloroform solution was washed with normal sulfuric acid, water and sodium bicarbonate solution with sequential addition, dried with anhydrous magnesium sulfate and evaporated. The residual syrup was washed with 3 parts of benzene. During this washing process, it turned into a hard resin. The resin was dissolved in alcohol, a small amount of solid separated by centrifugation, and most of the alcohol removed by evaporation.
Dann wurden die Bariumsalze desThe rather viscous solution was poured into liquid ammonia (about 50 com) and small pieces of sodium added until a passing blue color spreading over the whole solution was observed. Ammonia was evaporated, the residue dissolved in cold water (20 ecm) and passed through a column of an ion exchange resin of the alkylsulphonic acid type. The eluted solution was adjusted to the p n value of 8 with barium hydroxide solution, the barium phosphate was removed by centrifugation and the clear solution was evaporated to a small volume. Then the barium salts of

3. Reinigung durch Silbersalze3. Purification by silver salts
Silver nitrate solution was added in excess to a solution of the barium salts (1.2 g) in water (50 ecm). Then, the pH value was adjusted to 7 with dilute sodium hydroxide solution. The precipitated silver salts were collected by centrifugation and rinsed with water until the laundry was free of barium ions, which required about four rinses. The silver salts were then suspended in water and reacted with hydrogen sulfide. Excess hydrogen sulfide was removed by means of aeration and the solution adjusted with Bariumhydroxydlösung on the pH value. 8 silver sulfide was removed by centrifugation and the clear supernatant liquid was evaporated under reduced pressure to a very small volume. the barium salts "(0.3 g) were precipitated by addition of acetone and ether . Another amount (0.6 g) was extracted from the silver sulfide precipitate by slurrying it in water and bubbling hydrogen sulfide to the point of saturation. During this treatment, the precipitate decreased in mass considerably and turned dark. It was removed by centrifugation and the supernatant liquid evaporated to a small volume from which the barium salts were isolated as described above.
4. Herstellung von Pantethin-4'-pho'sphat oder
Pantethein-4'-pho'Sphat4. Production of pantethine-4'-phosphate or
Pantethein-4'-pho'Sphat
One proceeds first according to paragraph 2 and changes this process for the production of the pure pantethein-4'-phosphate to the effect that the flow of the solution through an ion exchange column proceed as quickly as possible after reduction with alkali metal in liquid ammonia goes. In order to obtain a pure product with absolute certainty, you can use a further reduction Perform alkali metal in liquid ammonia. On the other hand, if you want pure pantethine-4'-phosphate win, the aqueous solution is after the reduction stage an air or Exposed to oxygen flow until, all pantethein-4'-phosphate is converted into pa.ntethine-4'-phosphate. This can be done in a simple way. Way through a Detect negative sodium nitroprusside reaction. If necessary, you can use the Isolate barium salts by precipitation with acetone.

Manufacture of pantethine (different process)
Pantethein (5 g) was dissolved in ethyl alcohol (10 ecm) and water (10 ecm) dissolved. The solution was Ethylenediamine (0.5 ecm) and aqueous copper hydroxide given, which is fresh by precipitation made from copper chloride (20 mg). The solution was in an oxygen atmosphere shaken until there was a negative nitroprusside reaction to mercapto groups. Basic ingredients were removed by passing the solution through a column of ion exchange resin of the type of alkyl sulfonic acids in the hydrogen form. The eluted liquid was evaporated under reduced pressure to give pantethine essentially as calculated Yield.
The pantethine obtained in this way is then phosphorylated and, as described in the Sections 1 and 2 of Example I covered.
