Description
Method for preparing bones suitable for transplantation It it is known to obtain glue or gelatine from bones, but this is already the case with this method is avoided from the use of harsh bleaches, including hydrogen peroxide advised against. In a known method of this type, the bone material is destroyed. It is also known to use trypsin as an additive to soaking agents. the. All these procedures lie because of the other purpose and also because of the principle other effect apart from the present invention, the purpose of which is to provide a method to create for the treatment of bones so that these bones with success Living beings, especially humans, can be transplanted. In addition, there is compliance various biologically important factors are essential for pure bleaching of bones do not play a role and have not been taken into account so far. - It It is important to treat the bones in such a way that they allow for growth are not destroyed and, from a purely biological point of view, the agents used are not harmful Leaving influences in this direction. That would be the case, for example, if from sodium compounds or other alkali compounds dung residues remain, which are toxic. After the procedure. the registration will be on the one hand preserve the biological properties of the bone that are important for transplantation, on the other hand, sterility is ensured.
It is also already known, bone preparations from a living being to overplant on the other, e.g. B. are bones of corpses or of living Donors used surgically for transplantation after a limited storage time.
Bone and Joint Surgery, 36A, S. 72I bis 73I (I954)].The most common method is to store them in freezers. Another method is to keep the bone in or in blood a chemical solution (e.g. in a mercury compound). Finally there is still different procedures, bones (outside and inside. also in the small inner cavities) rid of all soft tissues, the maceration. Those so far in this field Practiced procedures change either the biological or the mechanical properties of the bone so strong that the macerated bone is less suitable for grafting was. For the expediency of the selection of the bones stands at the moment as the most sensitive Method of the »Spongiosatest« (Maatz) available [Journ. Bone and Joint Surgery, 36A, pp. 72I to 73I (I954)].
According to the invention, this object is achieved by a method for the preparation of bones suitable for transplantation with removal of the soft tissue the same inside and outside, the fatty substances and the blood pigment, by the Bones with protein removers, e.g. B. at least 10% hydrogen peroxide solution or trypsin solution are treated, whereby temperatures are maintained that rule out a change in the biological material properties due to temperature. According to the invention, bones of very young individuals, in particular, are treated Bones of calves. Any materials, containers or tools that serve this process at a low temperature of at least 340 C aftertreated with fat-dissolving, low-boiling agents. Furthermore, the Procedure while keeping away the influences of damaging metals and / or their connection z. B. carried out in glass or porcelain containers. One gets in this way animal or human bones, stripped of soft tissues without those used for transplantation important biological or mechanical properties of the bones influence. The method even improves those favorable for transplantation biological properties of the bone.
The bone prepared in this way can be kept practically indefinitely and can, factory-made and fused in glass ampoules, on the market to be brought.
First of all, the starting material is important for the process. It is coming especially the use of human or animal bones more youthful Individuals in question because it has been found that their chemical and mechanical Structure compared to bones of older individuals is usually particularly suitable.
The maceration consists of a deproteinization, some of it even the hard bone substance grasps, a degreasing and a detoxification and finally in a sterilization, with a temperature during the individual work steps is paused, in which the risk of heat denaturation is safely avoided.
The deproteinization is caused by hydrogen peroxide, this has the following advantages in maceration according to the present process: It leaves behind no toxic degradation products, but can be used as water after the process has ended and rinse out oxygen without leaving any residues, causes sterilization in the same operation and bleaching of the preparations, with mechanical properties and biological Quality can at least be maintained and a complete removal of the loosened soft tissue is achieved.
The repeated sterilization and degreasing is based on the fact that the Preparations (or objects of apparatus) are treated with fat dissolving agents, dissolve the lipoid substances of pathogenic germs from their structure. The for the Transplantation of important e biologically favorable structure of the preparation remains unaffected, Adhering dirt particles and toxic substances are released or rinsed, the fatty substances are radically removed, the preparation is sterilized and the solvent particles are removed by evaporation without leaving any residue.
All operations are preferably carried out in glass or porcelain vessels made so that the bones in no way with damaging, from the metal or substances derived from its compounds can be contaminated.
The invention is carried out as follows, for example: starting material. Calf bones (up to 3 months old); in exceptional cases for particularly stressed Share ox bones.
Procedure I. Soak in flowing water for 2 to 3 days at room temperature (around 180) (washout of blood pigment). The even flushing of the preparations is guaranteed by storage on a z. B. from the bottom flushed rust, the contains the bones, with the lighter parts rising up in the solvent, the heavier ones sink through the grate.
Arbeitsgang 2. Step 2. Deproteinization according to Paragraph 5, treatment with over Io ° / oiger H2 02 solution in standing liquid, after every 24 hours of renewal, two to three times, So 2 to 3 days duration, with constant (thermostat) about 370 C for the solution maintained will. Instead of H2 O2, trypsin (I OI crystalline Trypsin in Ringer's solution, which is made alkaline by adding Na2CO3 to a pH of 8.5) are used. By treating in more concentrated H2 O2 a sterilization has already been carried out (with the bones attached to the Rising up are prevented from the liquid), so that as part of the overall treatment Sterilization process according to step 5 is planned to kill the germs, who may have infected the preparations during work steps 3 and 4 could.
Step 3. Soak in cold running water for 10 to 12 hours to wash off and removal of the loosened soft tissue.
Arbeitsgang 4. Step 4. Open drying at a maximum of 400 C (I2 up to 24 hours under protection against coarse contamination).
Arbeitsgang 5. Operation 5. Degreasing and sterilization in pure ether. the Bone parts lie on a metal-free grate that penetrates the vaporized ether the bone, is knocked down on top of a cooling coil and drips again and again as practically completely pure ether down on the bones, the fat and dirt parts rinsing and taking away. Recommended temperature below 500 C. Duration up to 24 hours. One can possibly kill particularly resistant pathogens, under pressure, increase above 500 C.
Then sterile removal and sterile storage until operative Use, if necessary melting in glass ampoules.
A suitable apparatus for operation 5 is a water bath I, which is kept at 600 C; in the same the porcelain containing the ether is immersed or a glass vessel 2, which is a glass grate 3 above the ethereal mirror for inclusion of the material to be degreased and sterilized. The end cover the extraction chamber carries a cooling coil 4 with cooling water 5 and drain 6, a funnel 7 with a tap for filling in the ether, a stopper 8 for a thermometer 9 and a discharge line I0 through which at the end of the distillation the ether is discharged into a condenser II after cooling on the cooling coil 4 is disabled by shutting off the cooling water.
After completion of operation 5, the bones become sterile the extraction came. mer and can be melted in sterile glass ampoules will.
In the method described, the sterilization is at least 340 C with fat-dissolving, low-boiling agents such as ether, alcohol, light petrol etc. in the agent itself or its vapors in general for all substances and containers as well as devices and instruments that serve this process, insofar as they are supported by the Agents are not attacked, applicable, especially for knives, scissors or Instruments with cutting surfaces, the cutting surfaces of which are at higher temperatures To suffer.
