Description
Verfahren zur Herstellung blutgerinnungshemmender Stoffe aus phosphorylierten Pektinen Nachdem experimentielle Untersuchungen von B e s t, C ow an und Mc. Le a n (Science, Jahrgang 1937, S. 85, 337) gezeigt hatten, daB Heparin, welches ein Polysaccharnd, das Glucuronsäure und Glucosamin enthält und reich an osterartig gebundener Schwefelsäure ist, dem Entstehen von Thrombosen entgegenwirkt, ist, im Hinblick auf die Erkenntnis, daB auch verschiedene Polysulfonsäuren, wie Kongorot, Chlorazol und Chikagoblau (die sich wegen Giftwirkungen nicht für die Humanmedizin eignen), ebenfalls kaagulationshemmend wirken, in den letzten Jahren mehr oder weniger erfolgreich versucht worden, klinisch brauchbare Heparinoide durch Veresterung von Polysacchariden mit Hilfe von Chlorsulfonsäure künstlich herzustellen. So sind blutgerinnungshemmende Stoffe dadurch erhalten worden, daB man Cellulose, Pektin, Alginsäure, Dextran, Xylan und Hya;luronsäure mit Chliorsulfonsäure in Pyri-din in Schwefelsäurcester übergeführt hat.Process for the production of anti-coagulant substances from phosphorylated pectins After experimental investigations by B est, C ow an and Mc. Aus Pektinen mit derart geringem Gehalt an Esterphosphorsäure wurden im Zuge der Arbeiten, die zu der vorliegenden Erfindung führten, nach der eingangs angegebenen Sulfonierungsbehandlung nur solche hochwirksamen Anticoagulantien erhalten, die ausschließlich bei intravenöser Anwendung den angestrebten Effekt herbeiführten. Letzteres ist beispielsweise dann der Fall, wenn der Aufschluß des Protopektins in den Ausgangsmaterialien der Pektingewinnung in Gegenwart von Phosphorsäure (Patent 730 898) erfolgt oder die Ausgangsstoffe der Pektingewinnung vor der hydrolytischen Aufschlußbehandlung mit Phosphorsäure behandelt (Patent 75916'I) worden sind. Daß man selbstverständlich auch anderweitig hergestellte und nachträglich aufphosphorylierte (Henglein, Krässig und Steimig, a. a.
