Description
Attachment of cover slips to microscope slides with colored tissue pieces for microscopic specimens The invention relates to the preparation of histological specimens Ge "" heal for microscopic examinations and especially the attachment of the Cover slip for the stained piece of tissue on the slide.
When dissecting tissue for making a microscopic Examining the same, it is common to place a coverslip over the stained piece of tissue the slide by connecting the cover glass to the slide by means of a To apply resin. The resin is used in the form of a solution; after evaporation of the solvent, the resin binds the coverslip to the slide. So far for Resins used for these purposes are: Canada balsam, damar resin and synthetic resin, such as B. cycloparaffin or naphthalene polymers. Resins of the type previously used are not flawless for a number of reasons, among which the following should be emphasized are: i. They are not sufficiently transparent, i. i.e., they have a proportionate low refractive index; 2. they darken with age; 3. They develop at the Aging a degree of acidity, which is particularly the case in connection with natural resins is, such as B. with Canada balsam, which forms acid with aging and thereby fades the color or harmful deterioration of the tissue; q .. they are not sufficiently inert to the tissue, or they do not stay during slow to aging; 5. They become brittle on hardening and cause the microscope slide break easier; 6. They tend to form crystallization with formation an unsightly foam rim around the coverslip; 7. they soften under temperature conditions, in which the slides are used, for example in microprojection work, which requires the use of carbon arc lamps to project the slides.
The main object of the present invention is to provide a new Method of attaching the coverslip to the slide using a Resin, which is essentially free of the complaints mentioned, to be provided. Further purposes and advantages emerge from the following detailed description the invention.
According to the invention, the cover slip is colored with the one Microscope slide provided with a tissue sample using a solution of a coumarone-indene resin, which is bright in its color, d. H. is light amber and has a melting point of at least 150 ° C, preferably from about 150 to about 175 ° C; in one aromatic hydrocarbon, the solution being about 5o to 6o% - in weight Contains resin. The resin solution is applied to the slide carrying a piece of tissue applied by means of a suitable device, such as. B. a glass rod for these purposes is used, whereupon the coverslip is brought into place. The resin solution spreads through the piece of tissue between the slide and the cover slip, and that After evaporation of the solvent, resin binds the cover slip to the carrier. Of the thin resin film thus formed on the piece of cloth is substantially colorless.
The resin according to the invention is a coumarone-indene resin which is prepared by polymerizing coumarone and indene components, which are present in a coal tar fraction, coke oven light oil or petroleum fraction, with appropriate purification so that the resulting resin is substantially colorless or is water-clear. It has a melting point of at least 150 ° C, preferably from 150 to about 175 ° C, a refractive index of 1.65 ° at 25 ° C, a molecular weight of 700 to 750 and is neutral with an acid number or value less than i. The resin can be polymerized by heat or catalytically as is known in the art of making coumarone-indene resins. The advantageous example of such a resin is that manufactured and sold by the Neville Company under the tradename Nevindene Ri, which has a color value of 1/2 the Neville Color Rating. This color value of 1/2 corresponds to the color from light amber to water white of the rosin standards, the resins of which are prepared in accordance with the US Department of Agriculture standards. Any aromatic hydrocarbon such as benzene, toluene or xylene can be used as the solvent for the resin. That. a preferred solvent is a petroleum benzene.
It has surprisingly been found that the above-described Resin solutions result in a marked improvement in the finished slide. The resin solution spreads smoothly into a flawless bond with the coverslip. It dries quickly and allows freshly prepared slides to be handled shortly after attaching the cover slip. It hardens quickly with no residual stickiness. The resin is chemically inert; it neither fades nor discolor the cuts, nor it has a harmful effect on the pieces of tissue, even after aging. The resin breaks not, even after aging, and remains clear as a polished lens. Even the resin does not darken after aging. It neither crystallizes nor develops an acid in aging. The slides can be used safely at high temperatures used by light sources such as B. in microprojection, without softening the resin bond between slide and cover slip. The resin has a relative high refractive index compared to the resins used so far with a corresponding Improving the transparency of the finished slide.
Within the scope of the inventive concept described above are of course Various changes are still possible without restricting the subject matter should take place.
