Verfahren zur Reindarstellung von Cozymase Hinsichtlich der Gewinnung
von Cozymase (Diphosphoryridinnucleotid-DPN) sind in neuerer Zeit verschiedene Vorschläge
zur Verbesserung der bis dahin üblichen Verfahren (vgl. »Biochemical Preparations«
Vol. I, S. 28, 1949) gemacht worden. Durch Anwendung moderner Methoden, wie die
der Gegenstromverteilung und des Ionenaustausches, ist es gelungen, wesentliche
Vereinfachungen bezüglich der technischen Darstellung dieses immer wichtiger werdenden
enzymatischen Wirkstoffes zu erzielen (zum heutigen Stand dieses Arbeitsgebietes
vgl. u. a. N e i 1 a n d s und Akeson in »The Journal of Biological Chemistrya,
188, 307, 1951). Die von den genannten schwedischen Forschern angewandte
Technik besteht im wesentlichen in der kombinierten Anwendung von Aktivkohle und
Anionenaustauschern, wodurch sich in nicht unerheblicher Verbesserung der bislang
erhaltenen Resultate DPN-Präparate mit einem etwa 801111 oigen Reinheitsgrad gewinnen
lassen. Allerdings erfährt die technische Bedeutung dieser Methode eine Beeinträchtigung
u. a. dadurch, daß recht erhebliche Flüssigkeitsmengen verarbeitet werden müssen.Process for the pure preparation of Cozymase With regard to the production of Cozymase (diphosphoryridine nucleotide-DPN), various proposals have recently been made to improve the previously customary processes (cf. By using modern methods, such as countercurrent distribution and ion exchange, it has been possible to achieve significant simplifications with regard to the technical presentation of this enzymatic active ingredient, which is becoming increasingly important (for the current status of this field of work see, inter alia, Nei 1 ands and Akeson in “The Journal of Biological Chemistrya, 188, 307, 1951). The technique used by the Swedish researchers mentioned consists essentially in the combined use of activated carbon and anion exchangers, which means that DPN preparations with a degree of purity of about 801111 can be obtained in a not inconsiderable improvement on the results obtained so far. However, the technical importance of this method is adversely affected by the fact that considerable amounts of liquid have to be processed.
Gegenstand der Erfindung ist nun ein Verfahren, mit dessen Hilfe es
gelingt, erstmalig in technisch brauchbarer Weise völlig reine Cozymase (DPN) darzustellen.
Es besteht darin, daß man DPN-haltige Rohkonzentrate, wie sie sich nach den verschiedenen
in der Literatur beschriebenen Verfahren aus Hefe oder anderen für die Gewinnung
dieses Enzyms in Betracht kommenden natürlichen Ausgangsmaterialien gewinnen lassen,
unter Anwendung von Kationen- und Anionenaustauschern selektiven Adsorptions- und
EluierungsmaBnahmen unterwirft. Zunächst läBt man das Konzentrat die Säule eines
starken Kationenaustauschers in der Säureform durchlaufen; hierbei ist darauf zu
achten, daB das p$ der
ausfließenden Lösung so niedrig bleibt, daß
keine intramolekulare Salzbildung der Cozymase eintritt. Hierauf wird die Lösung,
welche neben dem DPN andere Nucleotide und freie Phosphorsäure enthält, durch einen
mit Acetationen beladenen schwachen Anionenaustauscher gegeben, wodurch ein Austausch
u. a. der Gesamtmenge des primär vorhandenen DPN gegen Acetat erfolgt. Wird nun
das Adsorbat mit verdünnter Ameisensäure eluiert, wobei die Eluate zweckmäßig fraktioniert
aufgefangen werden, so wird selektiv das gesamte DPN ausgewaschen, während unter
diesen Umständen die anderen sauren Bestandteile des Konzentrates in überwiegendem
Maße adsorbiert bleiben. Die Eluate werden, gegebenenfalls nach Einengung, z. B.
auf einen DPN-Gehalt von mindestens i 0j0, in der Kälte mit ausreichenden Mengen
solcher mit Wasser mischbarer organischer Lösungsmittel versetzt, in denen, wie
z. B. in Aceton, das DPN bekanntermaßen unlöslich bzw. schwerlöslich ist, worauf

Man erhält so, je nach Güte des dem Verfahren der vorliegenden Erfindung
unterworfenen Konzentrats, in den meisten Fällen zunächst noch schwach verunreinigte
Präparate. Schon nach einmaliger Wiederholung der erfindungsgemäßen Methode indessen
werden Produkte erhalten, die sich nach der enzymatischen und chemischen Analyse
als ioo%ig rein erweisen; die erzielten Ausbeuten entsprechen praktisch dem DPN-Gehalt
der eingesetzten Konzentrate. Beispiel In 851 Wasser, welches mit Dampf zum Sieden
erhitzt ist, werden unter ständigem Einleiten von Dampf 105 kg Hefe in einer solchen
Geschwindigkeit eingefüllt, daß die Temperatur zwischen 8o bis 82° C liegt. Man
läßt 5 Minuten bei dieser Temperatur stehen und kühlt sodann möglichst schnell auf
Zimmertemperatur ab, worauf filtriert wird. Man erhält 140 1 eines klaren Filtrats,
welches mit 14 1 Bleiessig (Plumbum subaceticum DAB 6) versetzt wird. Man filtriert
aufs neue und stellt das pH des Filtrates auf 6,3 ein. Hierauf wird mit 3,i51 25%iger
Silbernitratlösung gefällt. Der Niederschlag wird abzentrifugiert und sorgfältig
mit Wasser ausgewaschen. Die möglichst konzentrierte Suspension des Niederschlags
wird mit Schwefelwasserstoff zerlegt, das ausgeschiedene Silbersulfid abzentrifugiert
und der Niederschlag mehrmals gut mit Wasser ausgewaschen.Depending on the quality of the process of the present invention, this is obtained

Ionenaustauscher gegen Acetat ausgetauscht wird. Das DPN läßt sich nun quantitativ
mit 8,5%iger Ameisensäure aus dem Austauscher eluieren, wobei das Eluat in Fraktionen
von jeweils ioo ccm getrennt aufgefangen wird. Die Lösungen, die mehr als i % DPN
enthalten, werden direkt bei o° mit 5 Teilen kaltem Aceton versetzt, wobei das gesamte
DPN ausfällt. Fraktionen niederer Konzentration werden im Vakuum eingeengt und anschließend
ebenfalls mit Aceton versetzt. Das gefällte Produkt enthält nach dem Trocknen etwa

ein Präparat, das nach der enzymatischen und chemischen Analyse zu ioo % aus DPN
besteht. Die Kopf- und Schwanzfraktionen der Elution ergeben 75- bis go0,ioige Präparate,
welche erneut in den Reinigungsprozeß gebracht und ebenfalls in ioo%iges DPN umgewandelt
