Verfahren zur Herstellung von 4-Monoalkylaminosalicylsäuren und 4-Monoalkyloxyalkylaminosalicylsäuren
4-Monoalkylaminosalicylsäuren und 4-Mono-(alkyloxyalkyl)-aminosalicylsäuren der
allgemeinen Formel
in der R Alkyl- oder Alkyloxyalkylgruppen bedeutet, welche 2 oder mehr C-Atome in
der am N-Atom gebundenen Alkylgruppe tragen, sind noch nicht beschrieben worden.
Man kann zu diesen alkylierten bzw. alkyloxyalkylierten Aminosalicylsäuren gelangen,
indem man ein am N-Atom alkyliertes oder alkyloxyalkyliertes m-Aminophenol, welches
2 oder mehr C-Atome in der am NAtom gebundenen Allzylgruppe besitzt, mit Kohlensäure
bei normaler oder erhöhter Temperatur und gegebenenfalls unter Anwendung von Druck
in Gegenwart von Wasser und bzw. oder einem anderen geeigneten Lösungsmittel zur
Umsetzung bringt. An Stelle von Kohlensäure bzw. außer Kohlensäure können auch Kohlendioxyd
abspaltende Verbindungen, wie die Carbonate oder Bicarbonate der Alkalimetalle,
verwendet werden. Zu dieser Umsetzung können an Stelle der freien, am N-Atom substituierten
m-Aminophenole

Beispiel i 4-n-Butylaminosalicylsäure 45 g N-(n-Butyl)-m-aminophenol
werden zu einer Lösung von 16 g K O H in Zoo ccm Wasser gegeben. Zu dieser Lösung
gibt man 50 g K H C 03 und setzt das Gemisch in einen kupfernen Rührautoklav
bei i2o bis 13o° unter einem CO,-Druck von 30 at mehrere Stunden um. Man
versetzt das Reaktionsprodukt mit Wasser und extrahiert mit Essigester, um nicht
umgesetztes N-(n-Butyl)-m-aminophenol zu entfernen, filtriert die wäBrige Lösung
und säuert zum pH-Wert 4 an. Die ausfallende 4-n-Butylaminosalicylsäure wird nach
dem Trocknen aus Benzol umkristallisiert. Fp. 134° (Zersetzung). Beispiel 2 4-Isopropylaminosalicylsäure
30 g N-Isopropyl-m-aminophenol werden mit ioo g KHC03 in 5o ccm 6o°/oigem
Alkohol unter Rühren in einem Autoklav bei i2o bis 13o° unter einem C02 Druck von
5o at mehrere Stunden zur Reaktion gebracht. Man nimmt mit Wasser auf und isoliert
nicht umgesetztes N-Isopropyl-m-aminophenol durch Ausziehen mit Essigester. Beim
Ansäuern der wäBrigen Lösung bis zum pH-Wert von 4 fällt die 4-Isopropylaminosalicylsäure
aus. Sie wird aus Benzol umkristallisiert und bildet Blättchen vom Schmelzpunkt
i2o° (Zersetzung). Beispiel 3 4-ß-Methoxyäthylaminosalicylsäure ioo g K H
CO, und eine Lösung von 25 g N-ß-Methoxyäthyl-m-aminophenol und 2o g K 0
H in ioo ccm I320 werden unter einem C02 Druck von 50 at in einem kupfernen
Rollautoklav 8 Stunden auf i2o bis 13o° erhitzt. Nach dem Erkalten löst man das

4-ß-Methoxyäthylaminosalicylsäure wird nach dem Absaugen und Trocknen mehrmals aus
Essigester umkristallisiert.
