Beim Arbeiten im Verhältnis i : i kann
Process for the preparation of thiosemicarbazones It was not yet known how the three isomeric phthalaldehydes would behave when reacted with thiosemicarbazide. In particular, it could not be foreseen whether the reaction of 1 mol of phthalaldehyde with z mol of thiosemicarbazide would lead to phthalal monothiosemicarbazones or to a mixture of phthalal monothiosemicarbazone with dithiosemicarbazione. would. The formation of mixtures of mono- and dithiosemicarbazones was also possible in the reaction of 1 mole of phthalaldehyde with a mole of thiosemicarbazone, although it was not known whether the products formed by the concurrence of these two reactions could be easily separated. It has now been found that the three isomeric phthalaldehydes can be reacted with thiosemicarbazide in the molar ratio i: a to give the dithiosemicarbazones and that the phthalaldehydes can also be reacted with thiosemicarbazide in the ratio i: i with the formation of monothiosemicarbazones. When working in the ratio i: i, the formation of dithiosemicarbazones can be suppressed by gradually adding the thiosemicarbazide solution to the phthalaldehyde and avoiding prolonged heating of the mixture. In both cases, the separation of the mono- or dithiosemicarbazones can easily be carried out by recrystallization from suitable solvents. The products produced according to the invention are intended to be used for pharmaceutical purposes or as intermediates for pharmaceuticals.
Beispiel i 27g Terephthalaldehyd werden in 2ooo ccm Wasser heiß gelöst
Example i 27 g of terephthalaldehyde are dissolved in 2,000 cc of hot water
and an equally hot solution of 36 g of thiosemicarbazide in 250 cc of water was added.
A dense precipitation occurs immediately. After the reaction has ended, suction
and drying gives the terephthalaldehyde-di-thiosemicarbazone in good yield
as a pale yellow powder. It has not yet melted in the melting point tube at 34o °.
The product obtained is insoluble in common solvents. It solves
in caustic soda with a yellow color.
in kleinen Portionen zugegeben.
Man saugt heiß vom Ungelösten ab.
The composition is EXAMPLE 2 27 g of terephthalaldehyde are boiled in .75.0 cc of alcohol on a reflux condenser, and a bite solution of g of thiosemicarbazide in 250 cc of water is added in small portions. A separation begins towards the end of the addition. It is advisable to avoid prolonged heating in order to avoid the formation of dithiosemicarbazone due to disproportionation. One sucks hot from the undissolved matter. The undissolved part turned out to be the "dithiosemicarbazone" described in Example i, which is formed here as a by-product.
in guter Ausbeute.
When the filtrate cools, yellow, shiny crystals separate in abundance. They can be recrystallized from dilute alcohol or from butanol. Soluble in caustic soda with a yellow color. Melting point around 25 ° with decomposition. According to the result of the analysis, the monothiosemicarbazone of terephthalaldehyde is present EXAMPLE 3 27 g of isophthalaldehyde are dissolved while hot in 750 cc of alcohol and a hot solution of 3.6 g of thiosemicarbazide in 6a, cc of water and a few drops of glacial acetic acid are added. After a few minutes, an almost colorless deposit sets in, which multiplies rapidly. It is left to stand on the water bath for a short time, then cooled and filtered off with suction. The dithiosemicarbazone of isophthalaldehyde is obtained in good yield. For cleaning, the substance is boiled with alcohol. In the melting point tube against z47 ° decomposition with intense gas evolution.
The monothiosemicarbazone of isophthalaldehyde can be obtained
if one works in a similar way as with terephthalaldehyde (example e). Even
mono- and thiosemicarbazone can be obtained from o-phthalaldehyde in a similar manner
will.
