Description
Process for the production of a healing and protective serum against distemper For a number of years attempts have been made to find an effective To produce protective and healing serum against those diseases of animals, which from Veterinarian is referred to as distemper.
The relevant scientific literature is very extensive, especially with regard to aetiology, there is still no agreement between the Meimingen has been achieved.
While the research of the English L a i d 1 a w and D u n k i n those already set up by the French C 4, r r e at the time Confirm the theory of the presence of an ultravisible, filterable virus appeared, has the failure of Walter S c h u 1 z on this basis Protective and healing sera produced, in turn, placed the view in the foreground, that bacteria are to be regarded as causative agents of distemper. This view eats apparently also gave rise to the fact that using bacteria a A number of sera have been brought onto the market without, however, up to now any of them could have withstood an impeccable check.
A large number of in-depth studies have shown that the distemper actually bacteria must be viewed as pathogens and that bacteria in particular play a role, which; the so-called Kolityphus group belong. These bacteria are closely related, but by cultural-biochemical Methods distinguishable from one another.
The bacteria of the coli group, the paratyphoid groups, belong to this large group A and B (S .c h o t t m ü 11 e r), -the bacteria of the enteritis group, like the bacteria Enteritidis Gärtner and E, nteritidis Breslau, the Kiel tribe and the human enpatho, genes Bacterium typhi abdominalis, also also those that have become known in recent times Intermedius stems (S t a n d f u ß).
The bacteria of the paratyphoid enteritis group are culturally blochemical characterized by the fact that they first produce reddening in litmus whey, which.- but later turns blue and persists that they also ferment mannitol, Do not attack sucrose and lactose, and do not recognize any indole reaction permit. The bacteria of the Koli group constantly redden litmus whey, ferment glucose, Milk sugar and mannitol show a positive indole reaction. The bacteria of the intermedius group show -n in litmus whey the same behavior as that of the paratyphoid enteritis group (first reddening, then blueing), they also ferment mannitol; regarding the other However, types of sugar and in relation to the indole reaction exist in the individual Intermiediustypes great diversity.
Based on the result of the investigations made, it is "succeeded in obtaining a serum by the method explained below," with which a curative effect could be achieved in diseases of distemper and which is also able to develop a protective effect that prevents the disease. It is : but it is necessary to obtain the serum, d. H. for the pre-treatment of the the serum-producing animals to use bacteria, some of which belong to the Paratyphusenteriti.sgruppe, belong partly to the Koligruppe and partly to the Intermediusgroup.
Man beginnt z. The inventive method is accordingly that several Strains of bacteria, some of which belong to the paratyphoid enteritis group and some of the coli group and part of the Intermedius group, incorporated into animals in the form of a vaccine from whose blood the serum is obtained in the usual way. Goes in detail the production z. B. in front of you: For the preparation of the healing and protective serum killed bacterial bodies are used, which in physiological saline solution are bloated. These vaccines are small at first, then larger and larger Amounts incorporated subcutaneously in the animals. You start z. B. with eyelet (normal eyelet), gives 2 noses at the next injection, then increases to about 4 oil, 8 noses, 12 loosening, etc. The immunization process depends on what increases the injection doses as far as the reaction is concerned, the incorporation of the antigen (here the bacterial body) triggers in the vaccinated animal. It generally takes care of the injection site, which is expediently carried out in the shoulder area to allow an infiltrate to occur, which in different animals, also according to the dose, to different degrees is. Individual sensitivity plays a certain role here. Aside from that Occurrence of the infiltrate can also lead to temperature increases, which also tend to show fluctuations in their height. Finally the: injection hand in hand with weight loss, which is also different. To at the Not having any success in the immunization process is to be recorded as an important principle, that the next higher dose may only be administered after the Animal has at least regained the weight it had immediately before the previous injection. It is of great benefit in manufacturing of the serum littermates. You then only inject one of the first Animals, observes its reaction and then directs itself in treating the other Animals according to the experiences made with the first animal. You are in this process Able to try larger doses with the first injection. Tolerates the first animal the dose is good, so you can safely use the same dose with the Apply to siblings. In general, the rule that applies to very young people. Animals should be handled with greater caution than with older animals.
Man fängt . z. B. mit 0,5 ccm an, gibt dann steigend o,8, dann i,oo, 1,5, 2,00-CCM usw. In place of the bacteria, the toxins formed by such bacteria can also be used as a vaccine, as the following example is intended to illustrate: Live bacteria from the groups listed are added to meat broth (bouillon) and the culture is kept in the incubator at 37 ° C. After a certain time, the culture is filtered and the sterile filtrate is used for the injections. Here, too, you first take small amounts and then increase the doses. One catches. z. B. with 0.5 ccm, then gives increasing o, 8, then i, oo, 1.5, 2.00-CCM etc. Here too, as in the first example, one has to look at the body weight of the animals for the injection sequence; Take care.
It is advantageous to use a nutrient broth that has a specific Alkaline. Has acidity. Ordinary bouillon is used in its manufacture, as it reacts acidic, neutralized with alkali. The one for the extraction of toxins Only one third of the amount of alkali should be added to the bouillon to be prepared - n, which would be required to obtain a neutral broth. The Bo.uillon Cultures three guards must be kept in the incubator before they are filtered. the The new serum can also be produced in such a way that the bacteria or the toxins formed by them, separated by groups, used as a vaccine and different animals and ie from the blood of these animals in the usual way Sera obtained in this way are mixed.
