Verfahren zur Herstellung einer Verbindung aus Cotarnin und einer
Barbitursäure Gegenstand der Erfindung ist ein Verfahren zur Herstellung einer Verbindung
aus Cotarnin und einer Barbitursäure. Es ist an sich bereits bekannt, Verbindungen
des Cotarnins mit Säureamiden u. dgl. in der Weise herzustellen, daß man die betreffenden
Komponenten in -Gegenwart geeigneter Lösungsmittel aufeinander einwirken läßt (Patentschrift
23.2 785). Beschrieben ist insbesondere die Gewinnung von Verbindungen aus
Cotarnin und Bromisovalerylharnstoff bzw. Diäthylbarbitursäure,wobei als Lösungsmittel

Während nun auf diesem Wege die Verbindung mit der Diäthylbarbitursäure
ziemlich leicht hergestellt werden kann, hat sich herausgestellt, daß man beim Behandeln
des Cotarnins mit Dipropylbarbitursäure in Gegenwart von Benzol oder Alkohol nicht
zu der gewünschten Doppelverbindung gelangt. Man erhält bestenfalls ein rotgelbes

Doppelverbindung nach dem Abkühlen z. B. mit Wasser ausfällt, abtrennt und gegebenenfalls

Die Untersuchung hat zweifelsfrei ergeben, daß durch dieses Verfahren
eine Verbindung und nicht etwa nur ein Gemenge von Cotarnin und Dipropylbarbitursäure
gewonnen wird. Sie hat einen Schmelzpunkt von z15°, während der Schmelzpunkt der
Dipxopylbarbitursäure bei 14.5° und der des Cotarnins bei 132° liegt. Die Verbindung
ist schwerlöslich in Wasser, dagegen löslich in Äther, Trichloräthylen und Benzöl
und leichtlöslich in Aceton, Chloroform und Alkohol. Außerdem ist das durch Röntgendiagramm
ermittelte Kristallgitter der neuen Verbindung vollkommen verschieden von dem der
beiden Bestandteile. Im einzelnen- verfährt man bei der Herstellung in der Weise,
daß das Cotarninsalz und das Alkalisalz der betreffenden Barbitursäure in flüssiger
Phase einige Zeit am Rückflußkühler gekocht werden, wobei als Lösungsmittel Aceton
oder auch Alkohole verwendet werden. Allerdings ist die Ausbeute bei Verwendung
von Äthylalkohol etwas geringer als bei Aceton.The investigation has unequivocally shown that through this procedure

Die gewonnene Verbindung zeichnet sich durch vorteilhafte Eigenschaften,
insbesondere überraschende therapeutische Wirkungen, aus. In der Uterusbehandlung
zeigte bisher das Hydrastinin gegenüber anderen Mitteln insofern überlegene Wirkungen,
als es den isolierten Uterus in atonischem Zustande stillstellte; jedoch ist das
I3:ydrastinin verhältnismäßig schwer zu gewinnen und seine Anwendung daher sehr
kostspielig. Man hat sich infolgedessen in den meisten Fällen mit dem billigeren,
auch auf synthetischem Wege gewinnbaren Cotarnin beholfen, das
jedoch
den isolierten Uterus in der Kontraktur stillstellt. Damit sind gewisse Nachteile
verbunden. Bei der Prüfung der Dipropylbarbitursäureverbindung am isolierten, kontrahierten
Uterus ergab sich überraschender-a weise eine atonische Stillsetzung. Ein der-:.
artiges Ergebnis war nicht zu erwarten, da das freie Cotarnin eine gegensätzliche
Wirkung hat, Hydrastinin aber, das den Uterus ebenfalls in der Erschlaffung stillsetzt,
eine andere chemische Zusammensetzung besitzt und aus anderer Quelle gewonnen wird.
Am isolierten Uterus ausgewertet, erweist sich die neue Verbindung 3,7mal wirksamer
als Hydrastinin. Ausführungsbeispiel Cotarninchlorid und Natriumsalz der Dipropylbarbitursäurewerden
im Verhältnis i : i in Aceton unter Zusatz von etwas Wasser gelöst und i Stunde
am Rückflußkühler gekocht. Beim Abkühlen und Verdünnen mit Wasser fallen schwachgelbe
Kristalle aus. Ihce 'Zusammensetzung entspricht nach der #a e-,taranalyse der Formel
C=- H29 N3 01-g-:-dem Umkristallisieren aus Äther erhält 21.. 1

Kristalle vom Schmelzpunkt i5°, während der Schmelzpunkt der Dipropylbarbitursäure
