Verfahren zur Darstellung von aliphatisch und fettaromatisch substituierten
Pyridiniumäthanolen Durch eine vor kurzem erfolgte Veröffentlichung (Berichte der
Deutschen Chemischen Gesellschaft 66 [i933], S.607) wurde das Entstehen von den
Pyridinrest enthaltenden Äthanolen wahrscheinlich gemacht, wenn Benzaldehyd oder
m-Nitrobenzaldehyd mit Phenacylpyridiniumbromidoder Pyridiniumbrommethylat in Gegenwart'van
Alkali-kondensiert werden. -Es-wurde nun gefunden, daß man zu ähnlichen Kondensationsprodukten
gelangt, wenn man aliphatische oder auch solche Aldehyde von aliphatisclherii Charakter,
die mit aromatischen Resten substituiert sind, in -Gegenwart von Alkali auf Salze
von Pyridiniumalkylaten oder Acylverbindungen von ihnen zur Einwirkung bringt. Die
dabei entstehenden, bisher unbekannten Äthanole besitzen sehr wertvolle therapeutische
Eigenschaften und zeichnen sich durch ihre geringere Giftigkeit und besonders durch
ihre mehr auswählende Wirkung vorteilhaft vor den beiden bekannten Kondensationsverbindungen

Die Verbindung bildet . aus Alkohol lange Nadeln und zersetzt sich bei 22o bis 22z°,

in ii ccm go%igem Alkohol werden mit o,4 ccm ionormaler Natronlauge nach
24 Stunden bei o bis 20° durch Aufarbeiten wie, bei Beispiel i, wobei zum Schluß
im Vakuum eingeengt wird, 7001, der Theorie an N-(2-Oxy-3, 3, 4-tricblorp,entyl)
-pyridiniumbromid
erhalten; schmale Prismen aus 7 Teilen absolutem.Alkohol; F.219°.
Beispiel
3 -Zur Gewinnung des i-n-Hexyl-2-pyridiziumbromidäthanols-(i)
läßt man zog önanthol in iioccm goo/oigemAlkohol mit io g Phenacylpyridiniumbromid
mit 4 ccm i o normaler Natronlauge bei o bis 2o° go Stunden stehen. Dann werden
überschüssiges Önanthol, Benzoesäureester und Nebenprodukte nach Verwässern fortgeäthert,
die Lösung mit Bromwasserstoffsäure angesäuert, abermals zur Beseitigung von Önanthsäure,
Benzoesäure usw. ausgeäthert, heiß mit Tierkohle filtriert und dann im Vakuum, zunächst
bei geringem Unterdruck, verdampft. Der harzige Rückstand wird in 45 ccm Aceton
aufgenommen, mit Tierkohle filtriert und vorsichtig mit Äther gefällt; es kommen
49g schimmernde Blättchen des Hydrats; F. (nach dem Trocknen) 223° unter Zersetzung.
Beispiel 4 o,6g Hydrozimtaldehydin ii ccm 9o%igem Alkohol gelöst blieb mit i g Ph:enacylpyridiniumbromid
und 0,36 ccm io normaler Natronlauge 3 Tage stehen. Die Aufarbeitung geschah
wie bei Beispiel i. Die heiße, auf io ccm eingeengte Restlösung schied auf Zusatz
von Perchlorsäure ein Öl ab, das mit etwas Alkohol bei 4o° in Lösung gebracht und
mit Tierkohle filtriert wurde. Es schieden sich schmale Prismen des i-(ß-Phenyläthyl)-2-pyridiniumperchloratäthanol-(i
)
ab; F. (aus io Teilen heißem Wasser) 1q.5°. Beispiel 5 Bei der Einwirkung von i
g Phenylacetaldehyd in i i ccm go % igem Alkohol auf i g Phenacylpyridiniumbromid
unter dem Einfluß von o,:25 ccm io normaler Natronlauge bei o bis 20° wurde nach
Aufarbeiten @,d,@i.e bei Beispiel i, zum Schluß durch Eingen im Vakuum, das Bis-(i@oxy-2-phenyläxhyl)-methylpyridiniumbromid
der Formel
erhalten; 6seitige Blättchen aus io Teilen heißem Wasser, F. 2o6 bis 207°. In ihm
haben also 2 Molekülre Aldehyd mit i Molekül Phenacylpyridiniumsalz (unter Abspaltung
von i Molekül Benzoesäure) reagiert. Die erste Mutterlauge schied durch Einengen
und Abkühlen ein Öl aus, das mit wässeriger Pikrinsäure ein Kristallgemisch lieferte.
Trennung mit heißem Wasser führte zum Pikrat eines Diastereomeren obiger Kondensationsverbindung,
F. i73,5°, gelbe Nadelbüschel aus 4oo Teilen Wasser, und zu einem etwas leichter
löslichen Pikrat von F. i62°, zentimeterlange Nadeln aus Wasser; in ihm liegt das
Pikrat der Kondensationsverbindung aus i Molekül Aldehyd mit i Molekül Pyridinsalz
der Formel
