einer Lösung
von 7-(4-Fluorbenzyloxy)-2,3-dimethyl-1-[(1S,2S)-2-methylcyclopropylmethyl]pyrrolo[2,3-d]pyridazin
(67,9 g, 200 mMol) in Essigsäure
(800 ml) wurde Ammoniumcer(IV)-nitrat (329 g, 600 mMol) bei Raumtemperatur
gegeben. Die Mischung wurde für
8 Stunden bei 55°C
gerührt,
in Wasser gegossen und mit Ethylacetat extrahiert. Der Extrakt wurde
mit einer wässrigen
gesättigten
Natriumchlorid-Lösung

Zu dem Rückstand
wurde Methanol (500 ml) und eine wässrige 2N Lithiumhydroxid-Lösung (160
ml) gegeben und die Mischung bei Raumtemperatur für 40 Minuten
gerührt.
Das Reaktionsgemisch wurde mit 1N Salzsäure neutralisiert und das Methanol
im Vakuum abgedampft. Die resultierende Mischung wurde mit Chloroform
extrahiert. Der Extrakt wurde mit einer wässrigen gesättigten Natriumchlorid-Lösung gewaschen, über wasserfreiem
Magnesiumsulfat getrocknet und im Vakuum eingeengt. Der Rückstand
wurde auf einer Silicagel-Säule
unter Verwendung von Ethylacetat und Ethylacetat/Methanol = 9/1
als das Eluierungsmittel chromatographiert, um Kristalle zu ergeben,

Kalium-tert-butoxid

Als ein Protonen-Kalium-Adenosintriphosphatase-Präparat wurde eine mikrosomale Fraktion eingesetzt, die nach der Methode von Sachs et al. [J. Bio. Vorkultur eingesetzt, wurde Helicobacter pylori für 3 Tage
bei 37°C
unter mikroaerophilen Bedingungen in Kultur genommen. Nach Beendigung
der Kultivierung wurde visuell das Wachstum der Bakterien in dem
inokulierten Teil eingeschätzt.
Die kleinste Konzentration einer Verbindung der vorliegenden Erfindung,
bei der kein bakterielles Wachstum beobachtet wurde, wurde als MIC-Wert
(μg/ml)
bezeichnet. Die Verbindung vom Beispiel 2 zeigte eine hervorragende
antibakterielle Wirksamkeit, d.h. einen MIC-Wert kleiner als 12,5 μg/ml. FORMULIERUNGSBEISPIEL
