Verfahren zur Herstellung von oxyalkylierten Anminocarbonsäurenitrilen
Es wurde gefunden, daß man am Stickstoff oxy alkylierte Aminocarbonsäurenitrile
erhalten kann, wenn. man auf Produkte, die durch Umsetzung von primären oder sekundären
Oxyalkylaminen mit Aldehyden oder Ketonen erhältlich sind, Cyanwasserstoffsäure
oder diese liefernde Verbindungen einwirken läßt. Die bei der Einwirkung von Aldehyden
bzw. Ketonen erhältlichen Produkte stellen Oxazolidine oder am Stickstoff oxyalkylierte
Amine dar, die durch Behandlung mit Cyanwasserstoffsäure Nitrile liefern. Die Einwirkung
der Cyanwasserstoffsäure kann z. B. in der Weise erfolgen, daß man die Säure in
gasförmigem Zustand in Reaktion bringt oder sie im Reaktionsgemisch aus ihren Salzen
durch Zusatz von Säuren, z. B. durch Einleiten von Kohlendioxyd, in Freiheit setzt.
In vielen Fällen ist es nicht erfor derlich, die Kondensationsprodukte aus den Oxalkylaminen
und Aldehyden oder Ketonen abzutrennen; man kann vielmehr die Cyanwasserstoffsäure
unmittelbar auf das Reaktionsgemisch einwirken lassen. Die so erhältlichen Nitrile
sind infolge der Anwesenheit von reaktionsfähigen Gruppen weiterer Umsetzungen fähig;
sie lassen sich z. B. durch Verseifung in Carbonsäuren überführen, ferner zu Aminen
reduzieren usw. Sie sind Wertvolle Zwischenprodukte für die Herstellung von pharmazeutischen
Präparaten, Farbstoffen, Kunststoffen u. dgl. Beispiel i 6i Teile Äthanolamin werden
bei normaler Temperatur mit 75 Teilen 40 °/aiger Formaldehydlösung versetzt, wobei
das entsprechende Oxazolidin entsteht. Dem so erhältlichen Reaktionsgemisch werden
56,6 Teile 9o °/oiges Natriumcyanid zugegeben, worauf man bei 4.o bis 50° unter
Schütteln so lange gasförmiges Kohlendioxyd einleitet, bis dieses nicht mehr aufgenommen
wird. Die anfangs leicht bewegliche Flüssigkeit wird im Laufe der Umsetzung zähflüssig
und stellt nach deren Beendigung eine schwer bewegliche, wasserlösliche schwach
alkalisch reagierende Flüssigkeit dar. Das auf diese Weise erhältliche Nitril des
N-Oxyäthyiglykokolls wird mit 25 °/o seines Gewichts an 2o °/oiger Natronlauge 2
Stunden lang am Rückflußkühler gekocht. Es entwickelt sich dabei Ammoniak, und man
erhält das in Wasser leicht lösliche Natriumsalz des N - Oxyäthylglykokolls. Beispiel
e i Mol Formaldehydcyanhydrin läßt man unter Kühlung in i Mol Äthanolamin langsam
eintropfen. Die auf diese Weise erhaltene zähflüssige alkalisch reagierende Flüssigkeit
wird unter Zusatz von Natronlauge durch 2stündiges Kochen verseift. Man erhält ein
Produkt, das mit dem gemäß Beispiel i erhältlichen Nitril identisch ist.
Man
kann das gleiche Produkt auch erhalten, wenn man in eine wäßrige Lösung aus 53,3
Teilen 92 o/oigem Natriumcyanid bei 5 bis i o° Innentemperatur 104 Teile 35 o/oige
Salzsäure eintropfen läßt, hierauf 61 Teile Monoäthanolamin zufügt und anschließend
bei io bis i5° 75 Teile 40 ofoigen Formaldehyd langsam einfließen läßt. Nach beendetem
Eintragen wird das Reaktionsgemisch i Stunde lang auf 5o° erwärmt und das erhaltene

Beispiel s io5 Teile Diäthanolamin werden mit ioo Teilen 30 o/oiger Formaldehydlösung
versetzt und '/.Stunde lang auf 50° erwärmt. Hierbei bildet sich das 0xyäthyloxazolidin.
Man läßt hierauf in das Reaktionsgemisch bei _i o bis i5° 27 Teile wasserfreie Cyanwasserstoffsäure
eintropfen und erwärmt anschließend zwecks vollständiger Umsetzung das Gemisch etwa
2 Stunden lang auf 6o°. Die Bildung des N-Dioxyäthylglykokollnitrils ist beendet,
sobald kein Cyanwasserstoff mehr nachzuweisen; ist. DurchDestillation untervermindertem
Druck wird das Nitrit vom Wasser befreit; es stellt eine zähflüssige, schwach alkalische,

In eine Lösung von 77 Teilen ß-Oxypropylamin in. 5o Teilen Wasser
werden bei Temperaturen unterhalb 20° 72 Teile n-Butylaldehyd unter gutem Rühren
eingetragen. Es bildet sich sofort das ß-Methyl-d-n--propyloxazolidin von nachstehender
Konstitution:
Hierauf wird in die wässerige Suspension des Oxazolidins gasförmige Cyanwasserstoffsäure
eingeleitet. Die Temperatur hält man während des Einleitens zweckmäßig bei 18 bis
ao°. Anschließend rührt man das Reaktionsgemisch bei dieser Temperatur noch etwa
i Stunde lang und erwärmt hierauf zur völligen Umsetzung noch kurze Zeit auf 5o°.
Das erhaltene N-ß-oxypropylvaleriansäurenitril scheidet sich in der Hauptmenge als
ölige Flüssigkeit ab und wird von dem wässerigen Anteil getrennt. Der noch im Wasser

Teile wasserfreie Cyanwasserstoffsäure eintropfen, erwärmt dann auf 6o° und hält
diese Temperatur etwa i Stunde lang. Hierauf entfernt man das Wasser aus dem Reaktionsprodukt
durch Destillation unter vermindertem Druck und erhält so ein Nitrol von folgender
Zusammensetzung:
in Form eines dickflüssigen Öles.
An Stelle des Oxyäthylanilins
können 143 Teile Cyclohexyläthanolamin angewandt werden. Man erhält dabei als Endprodukt
ein Nitril von nachstehender Konstitution:
Beispiel 8 In eine Lösung von 75 Teilen ß-Oxypropylamin in 4o Teilen Wasser werden
bei 30° 86 Teile Methylisopropylketon eingetragen. Es bildet sich sofort ein Ox-azoli.din
von folgender Konstitution:
Zu dem erhaltenen Reaktionsgemisch fügt man dann 155 Teile einer 3a°/oigen Natriumcyanidlösung
und leitet bei etwa 5o° unter Rühren in das jetzt verschlossene Gefäß so lange Kohlendioxyd
ein, bis Sättigung erfolgt ist. Man erhält eine zähflüssige Lösung des Nitrils,
das nachstehende Konstitution aufweist:
Verwendet man an Stelle des Methylisopropylketons 58 Teile Aceton, so erhält man
zunächst das entsprechende Oxazolidin:
das durch Anlagerung von HCN in ein Nitril von nachstehender Zusammensetzung übergeht:
