Description
Process for the preparation of 1Vlonoacylderivaten cyclischer Aminothiophenolcarbon- and sulfonic acids The patent specification 487 104 relates to the representation of derivatives cyclic Aminometal mercapto compounds. These derivatives, which are highly therapeutic Have value are obtained by relying on cyclic aminometallic mercapto compounds or the salts thereof, acylating agents, or the aminomercapto compounds first acylated and then introduces the desired metal into the sulfhydryl group.
It has now been found that the monoacyl derivatives are also cyclic Aminothiophenolcarbonünd sulfonic acids, i.e. non-metal-containing compounds Surprisingly distinguished by a high chemotherapeutic 'value and can serve especially to combat spirillosis.
Monoacylierte Aminothiophenole sind zwar schon bekannt (vgl. Ber. d. deutsch. chem. Ber. d. deutsch. chem. Ges. 46 [igi3] S.778). Man erhält hiernach jedoch eine Diacetylverbindung.Monoacylated aminothiophenols are already known (cf. Ber. D. German. Chem. Ges. 39 [igo6] pp. 243o and 2434). The preparation was carried out by acylation of the dithio compounds and subsequent reduction of the acylated dithio compounds to the mercap-.tanes. However, this process is not economical because the yields are very low. Acylation makes the SS bond stronger than the non-acylated bodies and less vulnerable to attack by reducing agents. The difficulty of reduction can be circumvented and the monoacyl compounds mentioned can be obtained in excellent yield (96 to 100%) if one starts with the aminothiophenol carbonic or sulfonic acids or their salts and treats them with acylating agents. - Aminomercaptan has also already been acylated (cf. Ber. D. Deutsch. Chem. Ges. 46 [igi3] p.778). However, a diacetyl compound is then obtained.
Surprisingly, the acylation of aminothiophenol carbon- or sulfonic acids that always only one acyl group enters the molecule, namely on nitrogen, if the acylation is carried out in aqueous media or that treated reaction product formed in an anhydrous medium with water. The sulfhydryl group is not acylated, which is achieved by the introduction of a heavy metal, e.g. B. Gold, proven will.
The compounds are expediently used in the form of their alkali salts, which are neutrally soluble in water.
Example i 9 g of 4-amino-2-thiophenol-i-carboxylic acid are heated with 5 g of anhydrous sodium acetate and 30 cc of acetic anhydride for 3 hours under the reflux condenser with the addition of a drop of concentrated sulfuric acid. When the reaction mixture is poured into water, the 4-acetylamino-2-thiophenol-i-carboxylic acid is obtained as a yellowish-white powder. It dissolves in alkalis, hot alcohol and methyl alcohol, more difficult in acetone. It is insoluble in other common organic solvents and in water and does not melt until 270 °.
Example.?, If one proceeds as in example i using 4-amino-2-thiophenol-i-sulfonic acid (reports 32, p.1152), one arrives at 4-acetylamino-2-thiophenol-i-sulfonic acid. This compound, which decomposes above 250 °, is easily soluble in sodium carbonate and caustic alkalis, insoluble in the common organic solvents and crystallizes from hot water in colorless flakes. EXAMPLE 3 10 g of 4-amino-2-thiophenol-i-carboxylic acid are treated with phosgene in sodium carbonate solution at 50 to 80 ° until no more diazo reaction can be detected, the liquid being stirred well to ensure good agitation. After the reaction has ended, the symmetrical di- (2-thiophenol-i-carboxylic acid) q.-urea is obtained by acidification as a gelatinous precipitate which, after drying, forms an amorphous, yellowish-white powder that is easily soluble in alkalis.
Beispiel q.Example q.
4.-m-Nitrobenzoylamino-2-thiophenol-i-carboxylic acid is obtained, from the sodium salt of which, by reduction with zinc dust, the [m-aminobenzoylamino-2-thiophenol-carboxylic acid is obtained as a yellowish white compound in fine needles. The compound is soluble in alkalis, ethyl and methyl alcohol and insoluble in water, ether, petroleum ether, benzene, chloroform and carbon disulfide. If this acid is dissolved in sodium carbonionate and phgsgene is passed in at 5o to 8o 'with stirring until no more diazo reaction can be detected, the symmetrical di (4.-m-aminobenzoylamino-2-thiophenol-i-carboxylic acid is obtained after acidification of the reaction mixture -) urea of the formula as a yellowish amorphous compound that is insoluble in water and in common organic solvents, soluble in alkalis.
