Description

Verfahren zur Herstellung von toxin-antitoxinhaltigen Niederschlägen aus unter den üblichen Bedingungen nicht oder nur sehr spärlich flockenden Toxin-Antitoxin-Gemischen. Durch die Untersuchungen von R a m o n (C. r. Soc. Biol. I922, Bd.86, S.661, 711, 813; 1923, Bd. 88, S. 167) und anderen Forschern ist bekannt, daß beim Zusammenbringen von Diphtherietoxin und -antitoxin in optimalen Mengenverhältnissen eine Präzipitation auftreten kann. Diese bleibt jedoch aus oder ist nur sehr spärlich bei Toxinen, wie die von Scharlach, Ruhr, Tetanus und manchen anderen.Process for the production of toxin-antitoxin-containing precipitates from toxin-antitoxin mixtures which do not flocculate or flocculate only very sparsely under the usual conditions. Durch das nachstehend beschriebene neue Verfahren gelingt es nun, auch aus an und für sich nicht oder nur sehr spärlich flockenden Toxin-Antitoxin-Gemischen so reichliche Mengen der toxin-antitoxinhaltigen Niederschläge zu erhalten, daß eine weitgehende Verwendung dieser Präparate als Impfstoffe ohne weiteres möglich wird, was bisher ganz ausgeschlossen war. Das Verfahren ist gekennzeichnet durch Verwendung hoch konzentrierter Gifte, Arbeiten bei Salzarmut und einer Wasserstoffionen - Konzentration Ptl gleich oder kleiner als 6,8. Beispiel s. 21 durch Züchtung von Scharlachstreptokokken erhaltene Giftbouillon werden nach Filtration mit Ammoniumsulfat oder anderen eiweißfällenden Mitteln zur Ausfällung gebracht. Die Lösung des gefällten Toxins in der gerade ausreichenden Menge Wasser wird durch Dialyse gereinigt. Die so erhaltene salzarme bzw. salzfreie konzentrierte Scharlachtoxinlösung wird durch Ersatz von geringer Menge Säure so angesäuert, daß PH gleich oder kleiner als 6,8 ist, sodann wird mit genau soviel antitoxinhaltigem Serum versetzt, als sich im Tierversuch zur Neutralisation nötig erweist. Beim Stehen des so erhaltenen schwach erwärmten Toxin-Antitoxin-Gemisches scheiden sich dann reichliche Mengen des toxin-antitoxinhaltigen Niederschlages aus, der nach Abzentrifugieren und Waschen mit Wasser zwecks Impfstoffbereitung in isotonischer Kochsalzlösung suspendiert werden kann. Beispiel 2. Durch anaerobes Wachstum von Tetanusbazillen in Bouillon erhaltene Giftbouillon wird nach keimfreier Filtration entweder als solche oder nach Fällung mit Ammoniumsulfat und Dialysierung der den Niederschlag enthaltenen wäßrigen Lösung durch Zusatz von geringer ;Menge Säure so angesäuert, daß PH gleich oder kleiner als 6,8 ist. Diese Giftlösung wird nun mit so viel antitoxinhaltigem Serum versetzt, als sich im Tierversuch zur Neutralisation nötig erweist.
