Description

Die Verbindung 1-[(1,1-Dimethylethyl)amino]-3-[(4- morpholinyl-1,2,5-thiadiazol-3-yl)oxy]-2-propanol- maleat ist als Beta-Rezeptoren-Blocker bekannt und wird in handelsüblichen Arzneimitteln, beispielsweise unter den Bezeichnungen Betim, Blocadren, Temserin, Timacor, Timolate usw. aufgrund dieser Wirksamkeit angewendet, beispielsweise bei Angina Pectoris, Herzrhythmusstörun­ gen und Hypertonie. Bekannte Nebenwirkungen sind Kon­ traktion der Bronchialmuskulatur, also Verengung der Atemwege, Kontraktion der Uterusmuskulatur. Ferner wurde über eine Antiglaukomwirksamkeit berichtet.
Nunmehr wurde überraschenderweise gefunden, daß diese unter dem Freinamen Timololmaleat bekannte Substanz ein wirksames Mittel in der Behandlung der Hyperplasien der Prostata darstellt, insbesondere der benignen Prostata­ hyperplasie (BPH).
Die Ursache der BPH ist unbekannt, jedoch werden alters­ bedingte Veränderungen des hormonellen Gleichgewichtes vermutet. Die typischen Symptome sind Harndrang, Pollaki­ surie, Nykturie, Harnverhaltung und Verminderung der Stärke des Harnstrahls, Inkontinenz sowie Harntröpfeln und das subjektive Gefühl von unvollständiger Blasenent­ leerung. Die unvollständige Blasenentleerung wiederum verursacht Stase und Infektion mit sekundär entzündli­ chen Veränderungen in Blase und oberen Harntrakt. Länger andauernde Obstruktion kann zur Hydronephrose und Einschränkung der Nierenfunktion führen. Außerdem besteht die Gefahr der Steinbildung.
Die radikale Behandlung besteht in der chirurgischen Resektion.
An einer größeren Zahl von Patienten durchgeführte Behandlungen lieferten den Nachweis, daß Timololmaleat die oben erwähnten Symptome wesentlich vermindert und erleichtert.
Als Dosierung erwiesen sich 5 bis 20 mg/Tag zweckmäßig bei einer Verabreichung, die 1 bis 2mal täglich erfolgt. Da Timololmaleat zu einer Herabsetzung des Blutdrucks führt, soll dieser beachtet und die Dosierung darauf eingestellt werden.
Das Timololmaleat kann für die Herstellung eines Arznei­ mittels zur Behandlung der Prostatahyperplasien in den üblichen pharmazeutischen Darreichungsformen und Zuberei­ tungen hergestellt werden. Typische Verabreichungsformen in diesem Sinne sind Lösungen, Tabletten, Kapseln, Suppositorien, oral verabreichbare oder injizierbare Lösungen und Retardpräparate. Eine geeignete Zusammen­ setzung besteht beispielsweise aus halbierbaren Tablet­ ten mit 10 mg Timololmaleat, Rest übliche Tablettier­ hilfsstoffe und Geschmackszusätze.

Claims (3)

1. Verwendung von 1-[(1,1-Dimethylethyl)amino]-3-[(4- morpholinyl-1,2,5-thiadiazol-3-yl)oxy]-2-propanol- maleat bei der Behandlung von Hyperplasien der Prostata.
2. Verwendung der Substanz von Anspruch 1 für den Zweck von Anspruch 1 in einer Menge von 5 bis 20 mg/Tag und Person.
3. Verwendung von 1-[(1,1-Dimethylethyl)amino]-3-[(4- morpholinyl-1,2,5-thiadiazol-3-yl)oxy]-2-propanol- maleat zur Herstellung eines Arzneimittels zur Bekämpfung von Hyperplasien der Prostata.
