Description
Die vorliegende Erfindung betrifft neue Diphosphonsäure
derivate, Verfahren zu deren Herstellung sowie Arzneimittel,
die diese Substanzen enthalten.
In der DE-PS 18 13 659 sind Diphosphonsäurederivate beschrieben,
von denen die 1-Hydroxyethan-1,1-diphosphonsäure
als Mittel zur Behandlung von Morbus Paget Bedeutung
erlangt hat.
In der DE-PS 23 43 196 sind Azacycloalkan-2,2-diphosphon
säuren beschrieben, die als Calcium-Komplexbildner zur
Reduktion der Calcifizierung der Rattenaorta bzw. -niere
(vgl. Atherosclerosis 30, 313 (1978)) und als Zusatz in
Zahnpasta eingesetzt werden.
In der DE-PS 25 41 981 ist u. a. die Darstellung von
Azacycloheptan-2,2-diphosphonsäure beschrieben.
In der DE-PS 24 56 808 ist die CaHPO₄-Stabilisierung
gegenüber Fluorid von Azacycloheptan- und Azacyclopentan-
2,2-diphosphonsäure aufgezeigt.
Es wurde nun gefunden, daß analoge Derivate dieser Verbindungen
mit mehr als einem Heteroatom im Ring ebenfalls
gute Calcium-Komplexbildner sind, daneben aber auch eine
ausgeprägte Wirkung auf den Calciumstoffwechsel zeigen und
damit zur breiten Behandlung von Calciumstoffwechsel
störungen geeignet sind. Sie lassen sich vor allem sehr
gut dort einsetzen, wo der Knochenauf- und -abbau gestört
ist, d. h. sie sind geeignet zur Behandlung von
Erkrankungen des Skelettsystems wie z. B. Osteoporose,
Morbus Paget, Morbus Bechterew u. a.
Aufgrund dieser Eigenschaften finden sie aber auch Verwendung
in der Therapie von Knochenmetastasen, der Urolithiasis
und zur Verhinderung heterotoper Ossifikationen. Durch
ihre Beeinflussung des Calciumstoffwechsels bilden sie
weiterhin eine Grundlage für die Behandlung der rheumatoiden
Arthritis, der Osteoarthritis und der degenerativen
Arthrose.
Gegenstand der vorliegenden Erfindung sind demnach Diphosphonate
der allgemeinen Formel I
in der
X =O, NR′, S, SO, SO₂,
Y =X oder Valenzstrich,
A =Alkylenkette mit 1-5 Kohlenstoffatomen, die gegebenen
falls ein- oder mehrfach durch niederes
Alkyl substituiert sein kann, wobei diese
Alkylgruppen gegebenenfalls durch Phenyl, Phenoxy,
Alkoxy bzw. Alkylthio substituiert sein können
oder zwei Alkylgruppen zusammen einen Ring bilden
können, der durch Sauerstoff unterbrochen sein kann,
B = D = Aoder Valenzstrich,
R = R′= Hoder niederes Alkyl bedeuten, mit der Maßgabe,
daß für den Fall, daß B Valenzstrich bedeutet,
Y Valenzstrich sein muß,
bedeuten,
sowie deren pharmakologisch unbedenkliche Salze.
Niederes Alkyl soll eine Kohlenwasserstoffkette mit 1-5
Kohlenstoffatomen, vorzugsweise die Methyl-, Ethyl-, Isopropyl-
und n-Butylgruppe darstellen.
Unter Alkoxy bzw. Alkylthio versteht man bevorzugt die
Methoxy- bzw. Methylthiogruppe.
Substituierte Alkylreste sind bevorzugt die Methyl- und
Ethylgruppe.
Falls zwei Alkylgruppen als Substituenten von A, B oder D
zusammen einen Ring bilden, sind dies vor allem Fünf- und
Sechsringe, die höchstens ein Heteroatom der Bedeutung X
oder Y enthalten.
Beispielhaft seien einige Strukturtypen aufgeführt:
P = P(O) (OR)₂
Die Verbindungen der allgemeinen Formel I sind auf mono
cyclische und bicyclische Systeme beschränkt.
Die Verbindungen können als Stereoisomerengemisch oder als
reine cis- bzw. trans-Isomere vorliegen.
Asymetrische Kohlenstoffatome können die R- bzw. S-Konfiguration
besitzen, und die Verbindungen können optisch
aktiv oder als Racemate vorliegen.
Bevorzugt sind Verbindungen der Formel I, in den X =
Sauerstoff, Y = Sauerstoff oder Valenzstrich bedeuten.
Die Alkylketten A, B und D können insgesamt 1-15 Kohlenstoff
atome enthalten.
R und R′ sollen bevorzugt Wasserstoff darstellen.
Bei monocyclischen Verbindungen der Formel I handelt es
sich bevorzugt um 6-8 und 13-18-gliedrige Ringsysteme, wie
z. B. den Morpholin-, Oxa-azacyclooctan- sowie den Dioxa-aza-
cyclooctadecanring.
Bei den bicyclischen Verbindungen der Formel I handelt es
sich bevorzugt um den Oxa-aza-bicyclo[4,4,0]-, Dioxy-aza-
bicyclo[4,4,0]- und den Oxa-aza-spiro[5,5]undecanring.
Die Verbindungen der allg. Formel I werden nach an sich
bekannten Verfahren hergestellt, vorzugsweise indem man
ein Lactam der allgemeinen Formel II
in der A, B, D, X, Y und R′ die oben angegebene Bedeutung
haben, mit einem Gemisch aus phosphoriger Säure oder Phos
phorsäure und einem Phosphorhalogenid oder Phosphorylhalogenid
umsetzt, wobei man das Phosphorhalogenid auch allein
oder in Gegenwart von Wasser zur Reaktion bringen kann,
und anschließend zur freien Diphosphonsäure hydrolysiert,
oder gewünschtenfalls die isolierten Verbindungen der allgemeinen
Formel I in deren Ester bzw. in pharmakologisch
unbedenkliche Salze überführt.
Die bei dem Herstellungsverfahren eingesetzten Lactame der
allgem. Formel II werden mit 1-5, vorzugsweise 2-3 mol phosphoriger
Säure oder Phosphorsäure und 1-5, vorzugsweise 2-3
mol Phosphorylhalogenid, Phosphortrihalogenid oder Phosphor
pentahalogenid versetzt und bei 80-130°C, vorzugsweise
100°C zur Reaktion gebracht. Man kann die Reaktion auch in
Gegenwart von Verdünnungsmitteln wie Halogenkohlenwasser
stoffen, insbesondere Chlorbenzol, Tetrachlorethan oder
auch Dioxan, gegebenenfalls unter Zusatz von Wasser,
durchführen. Die anschließende Hydrolyse erfolgt durch
Erhitzen mit Wasser, zweckmäßiger jedoch mit halbkonzentrierter
Salz- oder Bromwasserstoffsäure.
Die freien Diphosphonsäuren der allgemeinen Formel I
können durch Erhitzen mit Orthoameisensäurealkylestern in
die entsprechenden Tetraalkylester überführt werden und zu
Diestern oder wieder zu den freien Tetrasäuren verseift
werden. Die Verseifung zu Diestern geschieht in der Regel
dadurch, daß man den Tetraalkylester mit einem Alkalihalogenid,
vorzugsweise Natriumjodid in einem geeigneten
Lösungsmittel wie z. B. Aceton bei Zimmertemperatur
behandelt.
Hierbei entsteht das symmetrische Diester/Dinatriumsalz,
das gegebenenfalls durch einen sauren Ionenaustauscher in
die Diester/Disäure umgewandelt werden kann. Die Verseifung
der Ester zu freien Diphosphonsäuren geschieht in
der Regel durch Kochen mit Salz- oder Bromwasserstoffsäure.
Man kann jedoch auch eine Spaltung mit Trimethyl
silylhalogenid, vorzugsweise dem Bromid oder Jodid
vornehmen.
Als pharmakologisch verträgliche Salze werden vor allem
Mono- bzw. Dialkali- oder Ammoniumsalze verwendet, die in
üblicher Weise z. B. durch Titration der Verbindungen mit
anorganischen oder organischen Basen wie z. B. Natrium-
oder Kaliumhydrogencarbonat, Natronlauge, Kalilauge,
wäßrigem Ammoniak oder Aminen wie z. B. Trimethyl- oder
Triethylamin hergestellt werden.
Die Alkalisalze werden in der Regel durch Umfällen aus
Wasser/Methanol oder Wasser/Aceton gereinigt.
Die erfindungsgemäßen neuen Substanzen der Formel I und
ihre Salze können in flüssiger oder fester Form enteral
und parenteral appliziert werden. Hierbei kommen alle
üblichen Applikationsformen infrage, beispielsweise
Tabletten, Kapseln, Dragees, Sirupe, Lösungen, Suspensionen
etc. Als Injektionsmedium kommt vorzugsweise Wasser
zur Anwendung, welches die bei Injektionslösungen üblichen
Zusätze wie Stabilisierungsmittel, Lösungsvermittler und
Puffer enthält.
Derartige Zusätze sind z. B. Tartrat- und Citrat-Puffer,
Ethanol, Komplexbildner (wie Ethylendiamintetraessigsäure
und deren nichttoxische Salze), hochmolekulare Polymere
(wie flüssiges Polyethylenoxid) zur Viskositätsregelung.
Flüssige Trägerstoffe für Injektionslösungen müssen steril
sein und werden vorzugsweise in Ampullen abgefüllt. Feste
Trägerstoffe sind z. B. Stärke, Lactose, Mannit, Methyl
cellulose, Talkum, hochdisperse Kieselsäuren, höhermolekulare
Fettsäuren (wie Stearinsäure), Gelatine, Agar-Agar,
Calciumphosphat, Magnesiumstearat, tierische und pflanzliche
Fette, feste hochmolekulare Polymere (wie Polyethylenglykole);
für orale Applikation geeignete Zubereitungen
können gewünschtenfalls Geschmacks- und Süßstoffe
enthalten.
Die Dosierung kann von verschiedenen Faktoren, wie Appli
kationsweise, Spezies, Alter und/oder individuellem
Zustand abhängen. Die täglich zu verabreichenden Dosen
liegen bei etwa 1-1000 mg/Mensch, vorzugsweise bei 0-200
mg/Mensch und können auf einmal oder mehrere Male verteilt
eingenommen werden.
Bevorzugt im Sinne der vorliegenden Erfindung sind außer
den in den Beispielen genannten Verbindungen und durch
Kombination aller in den Ansprüchen genannten Bedeutungen
der Substituenten ableitbaren Verbindungen die folgenden

6-Oxa-1-aza-cyclooctan-2,2-diphosphonsäure 7-Oxa-2-aza- bicyclo[6,4,0]dodecan-3,3-diphosphonsäure.
Die nachfolgenden Beispiele zeigen einige derVerfahrens
varianten, die zur Synthese der erfindungsgemäßen Verbindungen
verwendet werden können. Sie sollen jedoch nicht
eine Einschränkung des Erfindungsgegenstandes darstellen.
Die Verbindungen fallen in der Regel als hochschmelzende
Festprodukte an (Mono- oder Dinatriumsalz), deren Struktur
durch H-, P- und gegebenenfalls durch ¹³C NMR-Spektroskopie
gesichert wurde. Die Reinheit der Substanzen wurde
mittels C,H,N,P,S, Na-Analyse sowie durch Dünnschicht
elektrophorese (Cellulose, Oxalat-Puffer von pH = 4,0)
bestimmt. Zur Charakterisierung der einzelnen Verbindungen
werden die M rel -Werte (= relative Mobilität) bezogen auf
Pyrophosphat (M rel = 1) angegeben.
1,05 g (R,S)-6-Methyl-morpholin-2-on (Herstellung: (R,S)-
2-Aminopropanol wird mit Chloracetylchlorid zum (R,S)-2-
Chloracetamido-propanol (Öl) umgesetzt, das mit Natrium
hydrid zum (R,S)-6-Methyl-morpholin-2-on (Fp: 60-62°C)
ringschließt) werden mit 1,55 g phosphoriger Säure 10
Minuten bei 100°C zusammengeschmolzen. Man entfernt das
Ölbad, tropft langsam 1,7 ml Phosphortrichlorid zu und
erhitzt 20 Stunden auf 100°C. Man läßt abkühlen, versetzt
mit 20 ml Wasser und läßt 30 Minuten unter Rückfluß
kochen. Ein gelber Niederschlag wird abgesaugt und das
Filtrat eingeengt. Den Rückstand nimmt man in 15 ml Wasser
auf, stellt mit 2 N Natronlauge auf einen pH = 5 ein, versetzt
die Lösung mit 250 ml Methanol und saugt den Niederschlag
ab. Man erhält so 1,0 g = 32% der gewünschten
Verbindung als Dinatriumsalz mit 2 mol Kristallwasser; Fp.
ca. 270°C Z.; M rel : 0,6.
In analoger Weise erhält man durch Reaktion von phosphoriger
Säure und Phosphortrichlorid mit
- a) 6-Benzyl-morpholin-2-on (Fp: 98-100°C) die 4-Oxa-1- aza-6-benzyl-cyclohexan-2,2-diphosphonsäure als Dinatriumsalz mit 1 mol Kristallwasser in einer Ausbeute von 27%; Fp. ca 260°C Z.; M rel : 0,75.
- b) (S)-6-Methyl-morpholin-2-on (Fp: 61-63°C; [α] = + 14,9°, c = 2,3 in Methanol) die (S)-4-Oxa-1-aza-6- methyl-cyclohexan-2,2-diphosphonsäure, die als freie Säure mit 0,5 mol Kristallwasser isoliert wurde (s. Beispiel 2), in einer Ausbeute von 41%; Fp: 233- 237°C Z.; [α] = +5,2°; c = 1,2 in Wasser bei pH = 5; M rel : 0,6.
- c) (R)-6-Methyl-morpholin-2-on (Fp: 60-62°C; [α] = -15,9°, c = 1,4 in Methanol) die (R)-4-Oxa-1-aza-6- methyl-cyclohexan-2,2-diphosphonsäure, als Dinatriumsalz mit 1 mol Kristallwasser in einer Ausbeute von 12%; Fp: 230-234°C Z.; [α] = -5,0°; c = 1,2 in Wasser bei pH = 5; M rel : 0,6.
- d) 6-Isopropyl-morpholin-2-on (Fp: 65-67°C) die 4-Oxa-1- aza-6-isopropyl-cyclohexan-2,2-diphosphonsäure, als Dinatriumsalz mit 1 mol Kristallwasser in einer Ausbeute von 17%; Fp: <300°C; M rel : 0,65.
- e) 5-Thia-1-aza-cycloheptan-2-on (Fp: 114-117°C) die 5- Thia-1-aza-cycloheptan-2,2-diphosphonsäure als Dinatriumsalz mit 2 mol Kristallwasser in einer Ausbeute von 27%; Fp: ca. 270°C Z.; M rel : 0,55.
- f) 1,5-Diaza-5-ethoxycarbonyl-cycloheptan-2-on (wachs artige Substanz; hergestellt durch Umsetzung von 1,5- Diaza-cycloheptan-2-on mit Chlorameisensäureethylester) die 1,5-Diaza-cycloheptan-2,2-diphosphonsäure als Dinatriumsalz mit 1 mol Kristallwasser; Fp <300°C; M rel : 0,4; die Hydrolyse wurde in diesem Fall mit 6 N Salzsäure in 7 Stunden durchgeführt, um die Carbethoxyschutzgruppe quantitativ abzuspalten.
- g) 2,6-Diaza-6-ethoxycarbonyl-bicyclo[5,4,0]undecan-3-on (Fp: 188-190°C; hergestellt durch Umsetzung von 2,6- Diaza-bicyclo[5,4,0]undecan-3-on (Fp: 140-141°C) mit Chlorameisensäureethylester) die 2,6-Diaza-bicyclo- [5,4,0]undecan-3,3-diphosphonsäure als Dinatriumsalz mit 1 mol Kristallwasser; Fp <300°C; M rel : 0,45; die Hydrolyse wurde mit 6 N Salzsäure in 7 Stunden durchgeführt.
1,1 g 4-Thia-1-aza-bicyclo[4,4,0]-decan-2-on (Fp:168-
170°C; durch Umsetzung von 2-Mercapto-cyclohexylamin mit
Bromessigsäureethylester hergestellt) werden mit 1 g phosphoriger
Säure 10 Minuten bei 100°C zusammengeschmolzen.
Man entfernt das Ölbad, tropft langsam 1,7 ml Phosphortrichlorid
zu und erhitzt 24 Stunden auf 100°C. Man läßt abkühlen,
versetzt mit 20 ml Wasser und läßt 2 Stunden unter
Rückfluß kochen. Man saugt ab, engt das Filtrat ein, verrührt
den Rückstand mit Aceton und saugt das abgeschiedene
Produkt ab. Man erhält so 0,7 g = 32% der gewünschten
Verbindung als Säure mit 1 mol Kristallwasser; Fp: 245-
250°C Z.; M rel : 0,5.
Claims (4)
1. Diphosphonate der allgemeinen Formel I
in derX =O, NR′, S, SO, SO₂,
Y =X oder Valenzstrich,
A =Alkylenkette mit 1-5 Kohlenstoffatomen, die gegebenen
falls ein- oder mehrfach durch niederes
Alkyl substituiert sein kann, wobei diese
Alkylgruppen gegebenenfalls durch Phenyl, Phenoxy,
Alkoxy bzw. Alkylthio substituiert sein können
oder zwei Alkylgruppen zusammen einen Ring bilden
können, der durch Sauerstoff unterbrochen sein kann,B = D = Aoder Valenzstrich,
R = R′= Hoder niederes Alkyl bedeuten, mit der Maßgabe,
daß für den Fall, daß B Valenzstrich bedeutet,
Y Valenzstrich sein muß,
bedeuten,sowie deren pharmakologisch unbedenkliche Salze.
2. Verfahren zur Herstellung von Diphosphaten der Formel I
in derX =O, NR′, S, SO, SO₂,
Y =X oder Valenzstrich,
A =Alkylenkette mit 1-5 Kohlenstoffatomen, die gegebenen
falls ein- oder mehrfach durch niederes
Alkyl substituiert sein kann, wobei diese
Alkylgruppen gegebenenfalls durch Phenyl, Phenoxy,
Alkoxy bzw. Alkylthio substituiert sein können
oder zwei Alkylgruppen zusammen einen Ring bilden
können, der durch Sauerstoff unterbrochen sein kann,B = D = Aoder Valenzstrich,
R = R′= Hoder niederes Alkyl bedeuten, mit der Maßgabe,
daß für den Fall, daß B Valenzstrich bedeutet,
Y Valenzstrich sein muß,
bedeuten,sowie deren pharmakologisch unbedenkliche Salze, dadurch
gekennzeichnet, daß man eine Verbindung der Formel II
in der A, B, D, X, Y und R′ die oben angegebene Bedeutung
haben, mit einem Gemisch aus phosphoriger Säure oder Phosphor
säure mit einem Phosphorhalogenid oder Phosphorylhalogenid
umsetzt, wobei man das Phosphorhalogenid auch allein
oder in Gegenwart von Wasser zur Reaktion bringen kann,
und anschließend zur freien Diphosphonsäure hydrolysiert,
oder gewünschtenfalls die isolierten Verbindungen der allgemeinen
Formel I in deren Ester bzw. in pharmakologisch
unbedenkliche Salze überführt.
3. Arzneimittel, enthaltend eine Verbindung gemäß Anspruch 1
neben üblichen Träger- und Hilfsstoffen.
4. Verwendung von Verbindungen gemäß Anspruch 1 zur Behandlung
von Calciumstoffwechselstörungen.
