Description
- Verfahren zur Inhibition von Lipasen
- Die vorliegende Erfindung betrifft ein Verfahren, Triacylglycerol-Lipasen (EC. 3.1.1.3) durch substratanaloge Substanzen zu hemmen. Bei diesen Substanzen handelt es sich um Malonsäurediester und O-Acyl-tartronsäurediester mit mittellangen und langkettigen Alkylresten, so daß die Verbindungen hydrophobe Eigenschaften entsprechend denen des natürlichen Substrats, den Triacylglycerolen, besitzen. Aufgrund ihrer Esterbindungen und der hydrophoben Eigenschaften können sie in Wechselwirkung mit dem aktiven Zentrum des Enzyms treten. Sie werden jedoch von dem Enzym selbst nicht gespalten und hemmen die Enzymaktivität.
- Zweck Die Hemmung von Lipasen kann von großer Bedeutung sein für medizinische Zwecke. Die Fettverdauung erfolgt durch die Pankreaslipase im Darm. Zur Behandlung von Adipositas und Hyperlipämien kann durch Gabe von Lipase-Inhibitoren die Fettverdauung teilweise verhindert werden. Das Nahrungsfett wird ungespalten ebenso wie der ungespaltene Inhibitor nicht resorbiert und wieder ausgeschieden.
- Die Anwendung der fettähnlichen Inhibitoren auf der Haut kann den mikrobiellen Abbau der Hautfette hemmen und die Verwertung der Triacylglycerole im Stoffwechsel der Mikroorganismen der Hautflora verhindern.
- Nahrungsmittel können lurch die Hemmstoffe vor der Hydrolyse durch Lipasen geschützt werden.
- Stand der Wissenschaft Pankreaslipase als bestuntersuchte Lipase spaltet die primären Esterbindungen von Triacylglycerolen. Die Produkte sind freie Fettsäuren und 2-Monoacylglycerole.
- Beide Produkte werden vom Körper resorbiert und verwertet.
- 1 ,213-Tri-O-alkylglycerole werden von Lipasen nicht gespalten (Morgan, R. G. H. and A. F. Hofmann, J. Lipid Res. 11 (1970) 223 - 230) und wurden als unverdauliche Fettersatzstoffe vorgeschlagen (Mangold, H. K., in: Ether Lipids, edit. by F. Snyder, Academic Press, New York-London 1972). Ebenfalls werden 1,3-Di-O-alkyl-2-acylglycerele (mit mittellangen Alkylresten, z. B. mit 12 C-Atomen) von Pankreaslipase nicht gespalten (eigene Untersuchungen). Die Triether- und Dietherverbindungen werden zwar nicht umgesetzt, aber sie beeinflussen auch nicht den Umsatz des natürlichen Substrats bei gleichzeitiger Anwesenheit (eigene Untersuchungen).
- Die Aufgabe, eine Verbindungsklasse zu finden, die Triacylglycerolen in ihren physikalisch-chemischen Eigenschaften möglichst ähnlich ist und an das aktive Zentrum von Lipasen gebunden wird, ohne umgesetzt zu werden, wurde dadurch gelöst, daß Malonsäure und Tartronsäure (Hydroximalonsäure) mit mittellangkettigen Alkoholen und die Hydroxylgruppe der Tartronsäure mit einer Fettsäure verestert wurden. Malonsäure und Tartronsäure sind als C-3-Verbindungen dem Glycerolgerüst der Triacylglycerole ähnlich. Die sekundäre OH-Gruppe der Tartronsäure trägt eine Fettsäure wie in den Triacylglycerolen, diese Esterbindung kann jedoch wegen der Positionsspezifität der Pankreaslipase nicht angegriffen werden. Die beiden äußeren Positionen haben dagegen "umgekehrte" Esterbindungen: Anstelle des Esters von Fettsäure und Glycerol handelt es sich um einen Ester einer Dicarbonsäure mit langkettigen Alkoholen. Bei einer vollständigen chemischen Hydrolyse der Verbindungen entstehen nur physiologische Komponenten: Fettsäuren, langkettige Alkohole und Malonsäure bzw. Tartronsäure.
- Substratanaloge von Triacylglycerolen mit derartigen Eigenschaften sind in der Literatur nicht beschrieben.
- Ihre mögliche technische und medizinische Anwendung wurde nicht erkannt.
- Beispiel Chemische Synthese von Malonsäure- und Tartronsäuredidodecylester mit üblichen Methoden durch Veresterung von Malonsäure bzw. Tartronsäure mit Dodecanol in Chloroform mit p-Toluolsulfonsäure als Katalysator. Acylierung von Tartronsäuredidodecylester mit Palmitoylchlorid in Chloroform/Pyridin zum Palmitoyl-Tartronsäuredidodecylester.
- Aktivitätsbestimmung von hochgereinigter Pankreaslipase (aus Schweinepankreas) mit dem Standardsubstrat Tributyrylglycerol bei 370 C im Autotitrator. Palmitoyl-Tartronsäuredidodecylester wurde von Pankreaslipase und auch von Carboxylesterase (EC. 3.1.1.1) (aus Schweineleber) nicht gespalten. Zusätze von 0,25, 0,5 und 1,0 molaren Anteilen an Ealmitoyl-Tartronsäuredidodecylester zu 1 Teil Tributyrylglycerol hemmte die Lipaseaktivität um 9, 30 und 50%.
- Äquimolarer Zusatz von Malonsäuredidodecylester zu Tributyrylglycerol hemmtedie Enzymaktivität um rund 30z.
Claims (1)
- Patentanspruch Verfahren zur Hemmung von Lipasen zu technischen, nahrungsmitteltechnischen und medizinischen Zwecken gekennzeichnet durch die Anwendung von Malonsäurediestern und O-Acyl-tartronsäurediestern als substratanaloge Inhibitoren.
