However, this method has the following disadvantages: The acid anhydrides or acid halides, which must be used as starting materials are quite expensive; the process itself is time-consuming, since it involves two stages, and since a solvent must be used, the separation is and Recovery of the solvent and an acid formed as a by-product after the separation of the reaction product is necessary.

Weygand-Hilgetag, Organisch-Chemische Experimentierkunst, 3. Auflage, Leipzig 1964, S. 556 bis 558). Jedoch ist aus der Literatur ersichtlich (vgl. G. Stork et al, J. Am. Chem. Soc. 85, 207 bis 222, und K a t ο et al, Chem. Pharm. However, it can be seen from the literature (cf. G. Stork et al, J. Am. Chem. Soc. 85, 207 to 222, and K a t o et al, Chem. Pharm. Bull. 17 [9], 1889 to 1895, 1969) that from] 3-aminocrotonamide under Using conventional acylating agents no ji-N-acyl compounds can be prepared.
The aim of the invention is to create an improved process for the preparation of pyrimidone derivatives, which uses an inexpensive raw material, a usable product is obtained in one step can be achieved, the use of a solvent is unnecessary and a high yield can be obtained.
It has now been found a process for the preparation of certain pyrimidone derivatives (III) which does not have the above disadvantages.
Gegenstand der Erfindung ist ein Verfahren zur Herstellung von 2-Isopropyl- und 2-Phenyl-6-methyl-4(3H)-pyrimidon durch Acylierung von jS-Aminocrotonamid und anschließendem Ringschluß unter Wasserabspaltung, das dadurch gekennzeichnet ist, daß man 0-Aminocrotonamid mit 1 bis 5 Mol Isobuttersäure-isobutylester oder Benzoesäure-äthylester in Gegenwart von 1 bis 3 g-Atomen metallischem Natrium je MolThe invention relates to a process for the preparation of 2-isopropyl- and 2-phenyl-6-methyl-4 (3H) -pyrimidone by acylation of jS-aminocrotonamide and subsequent ring closure with elimination of water, which is characterized in that one 0-aminocrotonamide with 1 to 5 moles of isobutyl isobutyrate or ethyl benzoate in the presence from 1 to 3 g-atoms of metallic sodium per mole

However, this reaction does not proceed if, in the above-mentioned known process, the acid anhydride or the acid halide are merely replaced by an ester of an organic acid. Surprisingly, however, it was found that when-aminocrotonamide is reacted with an isobutyric acid isobutyl ester or an ethyl benzoate in the presence of sodium as the alkali reagent, the condensation reaction and the ring closure reaction take place simultaneously and very smoothly in one stage.
It is not clear by which reaction mechanism the alkali reagent causes the reaction to proceed, since an ester / amide exchange reaction of this type is not yet known, but it is believed that first in an N-acylation process, the alkoxy group of the ester against the amino group of the j3-aminocrotonamide is exchanged and the effect of the alkali metal in the present reaction has the effect of an alkali catalyst in an ester interchange reaction is similar, so that the actual condensing agent is presumably an alkali metal alcoholate is.

The amount of ester to be used (isobutyl isobutyrate or ethyl benzoate) is theoretically 1 mol per 1 mol of aminocrotonamide, but so that the expensive ß-aminocrotonamide largely reacts with the ester, it is advisable to use the ester in the To use excess. If the ester is to serve as the reaction solvent, up to 5 moles of ester can be used be used. If another inert solvent, e.g. B. benzene or toluene is used, then more than 3 moles of ester are unnecessary.
The amount of sodium metal to be used is in the range of 1 to 3 gram atoms per mole / J-crotonamide.
For carrying out the reaction the metallic sodium with stirring to the isobutyric acid or benzoic acid ethyl ester Unhutvlester added Here, it is preferred to maintain a temperature of 50 0 C. The ^ -aminocrotonamide is then added and reacted with the resulting mixture. However, the order of addition is not so important and does not always have to be adhered to. If an inert solvent is used, as described below, then the order can be changed depending on the type of solvent.
The reaction is carried out at 50 to 7O 0 C, preferably at 50 to 6O 0 C, since a considerable discoloration of the reaction product occurs at a temperature above 70 ° C, which can be difficult to eliminate.
The reaction is complete in 3 to 7 hours.
After the completion of the reaction, the product can be recovered by an ordinary method. For example, the alkali present in the reaction mixture can be mixed with an inorganic acid such as z. B. hydrochloric acid or sulfuric acid, neutralize, filter off the precipitated salt, the resulting mother liquor concentrate under reduced pressure to produce crude crystals and the crude Crystals with a suitable solvent, such as. B. acetone, chloroform, methanol or ethanol, recrystallize.
The pyrimidone derivatives produced by the process according to the invention are valuable intermediates for a wide variety of synthetic organic chemicals.
The invention is explained in more detail with reference to the following examples.
Herstellung von 2-Isopropyl-6-methyl-4(3H)-pyrimidon Preparation of 2-isopropyl-6-methyl-4 (3H) -pyrimidone
Fp. 170 bis 1710C.10 g of metallic sodium were added to 100 g of isobutyl isobutyrate and 20 g E-Aminocrotonamid were added in small portions at a temperature below 5O 0 C. The reaction was carried out with stirring for 5 hours at 50 to 6O 0 C. The reaction mixture was then neutralized with hydrochloric acid under cooling, after which the precipitated sodium was separated by filtration and the resulting mother liquor was concentrated under reduced pressure to recover isobutyl alcohol and unreacted isobutyl isobutyrate. The resulting concentrated residue was recrystallized from acetone, whereby 28 g of 2-isopropyl-6-methyl-4 (3H) -pyrimidone (yield 92%) were obtained. Mp. 170 to 171 0 C.

In this example, the yield of the process according to the invention is examined in a comparative experiment compared to the process described in "A new Synthesis of 2-Alkyl (aryl) -substituted-6-methyl- (3H) -4-pyrimidone", Yakugaku Zasshi, 87 (8), 955 to 960 (1967), given. A comparison is given between those compounds in which the same substituents are contained in the 2-position of the pyrimidone. The minimum values (a) and the maximum values (b) of the yields of the O-N-acyl compounds at the stage of isolation are given in Tables I and II of the cited literature reference. In Table V, the minimum values (c) and maximum values (d) of the yields of the ring-closed compounds from the same kinds of the 0-n-acyl compounds are shown. From this the ranges of the total yields (e) can be calculated as (a ■ c - b ■ d).

It can thus be seen that the yields which can be achieved in the process according to the invention are significantly greater than the yields in the process from the cited literature reference is known.
