Description

The present invention relates to the development, formulation and evaluation of herbal cream formulations specifically designed for wound healing. It combines the use of natural ingredients with optimization of formulation properties to provide an effective, safe and patient-friendly alternative to promote wound healing.

Conventional wound healing treatments often rely on synthetic pharmaceutical products, which may have limitations in terms of effectiveness, safety and possible side effects. Given these challenges, there is growing interest in exploring natural alternatives for wound healing. The background of the present invention arises from this need and focuses on the development of an herbal cream formulation specifically designed to promote wound healing.

The formulation is based on the use of herbal extracts known for their therapeutic properties, with a particular focus on Argyreia nervosa extract. Argyreia nervosa is known for its antioxidant, anti-inflammatory and tissue regenerative properties, making it a promising ingredient for wound healing. By combining Argyreia nervosa extract with other carefully selected herbal ingredients, the formulation aims to utilize their synergistic effects to improve the wound healing process.

In addition to the selection of herbal ingredients, important formulation factors such as viscosity, spreadability, thermal stability and pH are also taken into account against the background of the invention. The present invention provides a novel herbal formulation for wound healing, bum healing, skin care and hemorrhoid treatment. The production of a herbal cream based on the herbal formulation is disclosed. The herbal formulation for the cream consists of an extract of Achyranthes aspera. Aloe vera gel. Natural lutein and lutein esters (from Tagetes erecta), Centella asiatica extract, Apamarga Kshar (water-soluble ash of Achyranthes aspera), Citrullus lanatus fruit extract and polyunsaturated essential fatty acids from natural sources as a carrier and cream base. The present invention differs from this in that it relates to the development, formulation and evaluation of herbal cream formulations specifically intended for wound healing.

The invention relates to an easy-to-apply, treatment-supporting, anti-inflammatory, wound-healing herbal additive cream, which contains bitter gourd extract, oil, petroleum jelly, stearic acid, alcohol, isopropyl myristate, glycerin and water in its structure and supports wound healing and the reduction of inflammation. The present invention differs from this as it relates to the development, formulation and evaluation of herbal cream formulations specifically designed for wound healing.

The present invention relates generally to the development, formulation and evaluation of herbal cream formulations specifically designed for wound healing.

One embodiment of the present invention is the extraction of active components from the leaves of Argyreia nervosa.

One embodiment of the present invention is to prepare an Argyreia nervosa cream with wound healing effect. The formulation was carefully prepared using appropriate excipients that complemented the physical properties of the herbal extracts to ensure a stable and effective cream.

In one embodiment of the present invention, the prepared herbal cream for wound healing activity was tested for viscosity, pH, spreadability, extrudability and thermal stability.

BIOLOGICAL SOURCE

The fresh leaves of the Argyreia nervosa plant were collected from the Amaravati area of Maharashtra.

EXAMPLE 1: Method of extraction

When making the herbal extract from the leaves of Argyreia nervosa, the fresh leaves were first dried in the shade to preserve their active ingredients. After drying, the leaves were pulverized using a multi-mill and stored in airtight containers to preserve their quality until further use.

The extraction of Argyreia nervosa leaves was carried out using a Soxhlet apparatus under continuous hot percolation. The powdered leaves weighing 500 g were degreased with petroleum ether to remove any fats present. Subsequently, a series of solvents with increasing polarity, namely petroleum ether and methanol, were used for the extraction process. Each extraction was carried out until the original color of the solvent reappeared, indicating the extraction of the desired compounds from the leaves.

After extraction, the extracts obtained in each solvent were collected separately and subjected to complete drying to remove all traces of solvent. This was done using a distillation system to ensure that the extracts were free of solvents. The concentrated extracts were then processed into a thick paste using a rotary vacuum evaporator and dried under vacuum conditions. These dried extracts were then used for further studies and development of the herbal cream formulation for wound healing.

EXAMPLE 2: Composition

The composition of the herbal cream for wound healing includes the following components and their respective amounts, which are listed in Table 1. The cream is intended for topical use and has been formulated to have enhanced wound healing properties such as: B. antibacterial activity, anti-inflammatory effects and stimulation of cell proliferation and tissue regeneration. The herbal cream formulation for wound healing is free of artificial colors, fragrances and synthetic preservatives and also contains additional natural ingredients such as aloe vera extract, calendula oil or chamomile extract in concentrations of 0.1 to 5%.

The combination of cetostearyl alcohol and cetomacrogol 1000 acts as an effective emulsifier system to achieve optimal stability and consistency; the presence of sodium lauryl sulfate improves the penetration and absorption of the herbal active ingredients into the skin for improved therapeutic effectiveness; Propylene glycol acts as a humectant, promoting moisturization and preventing the wound area from drying out; and the polysorbate 20 helps in the even dispersion of the oil and water phases and ensures the homogeneity of the formulation.

EXAMPLE 3: Manufacturing

The process for preparing the cream formulations is as follows:

In the aqueous phase, purified water is heated to 90°C using a stirrer and then cooled and kept at 60°C. Sodium lauryl sulfate is dissolved in the water heated to 60 ° C, then polysorbate 20 and propylene glycol are added. Methylparaben is then incorporated into the mixture, keeping the temperature in the water bath at 60 °C.

For the oil phase, cetomacrogol 1000, hard paraffin, light liquid paraffin and microcrystalline wax are melted together with cetostearyl alcohol. After melting, the temperature of the oil phase is cooled to 60 °C and maintained at this temperature.

The dispersed phase is prepared by transferring the oil phase into the aqueous phase maintained at 60 °C. The mixture is manually stirred at this temperature for 10 minutes and then homogenized at approximately 15,000 rpm for 5 minutes. The temperature is then reduced to 50 °C while continuing to stir by hand and the preparation beaker is placed in a 20 °C water bath.

In the drug phase, Argyreia nervosa extract is mixed with propylene glycol in a beaker until a clear solution is formed. The solution is heated and maintained at 50 °C in a water bath. It is then added to the dispersed phase, keeping the temperature at 50 °C. The drug phase beaker is rinsed with heated propylene glycol, which is then added to the dispersed phase. The mixture is mixed manually for 10 minutes and then homogenized for a further 5 minutes at 50 °C at 15,000 rpm.

The cream formulation prepared using this method has shown promising effectiveness in wound healing, infection prevention and scar reduction. Its natural composition and herbal ingredients make it a favorable option for people looking for alternative wound healing solutions.

EXAMPLE 4: Evaluations

The formulated herbal creams were subjected to various evaluations including physicochemical parameters. The physicochemical parameters of the cream formulations were evaluated to determine their visual properties.

The results showed that Formulation A, Formulation B and Formulation C all had a smooth texture with a brownish-yellow color. In addition, all formulations showed homogeneity, indicating an even distribution of ingredients in the cream.

Determination of viscosity:

The viscosity of Formulations A, B and C was measured with an LV-4 Brookfield viscometer at 6 rpm. Stable readings were obtained 60 seconds after spindle rotation began, and an average viscosity value was calculated from a maximum of three readings.

The results showed that Formulation A had a viscosity of 22,000 centipoise, while Formulation B had a slightly higher viscosity of 24,500 centipoise. Formulation C had the highest viscosity among the three formulations with a value of 26200 centipoise. These viscosity values indicate that the cream formulations have appropriate thickness and consistency, which are crucial for ease of application and effective delivery of the active ingredients.

Thermal stability:

Thermal stability assessment is important to assess the ability of cream formulations to withstand temperature fluctuations during storage and transportation.

The prepared formulations (A and C) were subjected to thermal stability testing by storing them at 37°C for 8 hours.

The results showed that Formulation A and Formulation C had good thermal stability as no phase separation was observed after being stored at 37°C for 8 hours. However, with formulation B, phase separation occurred under the same conditions. This observation suggests that Formulation B may be less stable and its physical properties may change when exposed to higher temperatures. The results illustrate how important it is to ensure adequate thermal stability of cream formulations to ensure their long-term quality and effectiveness.

Distributability:

The spreadability assessment provides information about the ability of the cream formulations to distribute evenly on the skin, which is crucial for effective application and absorption of the active ingredients. The spreadability of the cream formulations was determined by measuring the time required for the top slider to travel 5 cm under the force of a 20 g weight.

The results showed that Formulation A had a mean time of 10 seconds, resulting in a spreadability value of 14.4. Formulation B had a slightly longer average time of 10.6 seconds, which corresponds to a spreadability value of 13.5. Formulation C had the shortest mean time of 9.6 seconds, indicating better spreadability with a value of 15. These results suggest that all formulations have appropriate spreadability, allowing easy application and coverage of the affected area.

pH determination:

The pH of the cream formulations was measured using a pH meter. 5 g of the cream was dispersed in 45 ml of water and pH measurements were taken within 24 hours of preparation. The average pH for each formulation was calculated based on triplicate measurements.

The results showed that Formulation A had a pH of 6.3, Formulation B had a pH of 6.5 and Formulation C had a pH of 6.4. These pH levels are within a range considered compatible with the skin's pH level, which is usually between 4 and 6. The results show that all cream formulations have a pH value that is suitable for maintaining the skin's natural balance and minimizing the potential for irritation or disruption of the skin's barrier function.

Extrudability of the tube:

The extrudability of the cream formulation in the tube was evaluated based on the amount of cream extruded from the tube when finger pressure was applied. The formulation was placed in a clean, collapsible aluminum tube with a 5 mm nose tip opening and pressure was applied to the tube with fingers. The extrudability of the tube was determined by measuring the amount of cream extruded through the tip when pressure was applied.

The results showed that Formulation A had a net weight of 10 g and 6.1 g was extruded, resulting in a percent extrudability of 61%, which was considered sufficient. Formulation B had a net weight of 10g with 7.9g of cream extruded, corresponding to a percent extrudability of 79%, also rated as moderate. Formulation C had a net weight of 10 g with 8.2 g of cream extruded, resulting in a percent extrudability of 82%, which was rated as good. These results show that all formulations had satisfactory extrudability, allowing convenient and efficient removal of the cream from the tube.

Cetomacrogol 1000 at a concentration of 2%; d. light liquid paraffin at a concentration of 14.3%; e. hard paraffin at a concentration of 3.15%; f. microcrystalline wax at a concentration of 2.55%; G. Sodium lauryl sulfate at a concentration of 0.6%. H. Methylparaben at a concentration of 0.15%; i. Propylene glycol at a concentration of 5%; j. Polysorbate 20 at a concentration of 0.1%; and k. purified water with 3% heat loss at a concentration of 60.65%.
