Abstract
Description
Die Verbindung betrifft neue heterozyklische Verbindungen, die im folgenden auch als Jerangolide bezeichnet werden. Eine dieser Verbindungen, und zwar Jerangolid A, ist bereits in J. Anti biotics, 49 (1) (1996) 71-75 vorgeschlagen worden, wobei jedoch weder ein Weg zur mikrobiologischen noch zur vollsynthetischen Gewinnung aufgezeigt wurde. Claims (42)
- (a) Sorangium cellulosum DSM 10 497 (So ce307) in einem Koh lenstoff- und Stickstoff-Quellen sowie Mineralsalze enthal tenden wässerigen Medium aerob fermentiert, wobei man in an sich bekannter Weise
- (b1) in Abwesenheit eines Adsorberharzes fermentiert und nach der Fermentation ein Adsorberharz zugibt und/oder
- (b2) in Gegenwart eines Adsorberharzes fermentiert,
- (c) jeweils das Adsorberharz mit den durch die Fermentation ge bildeten heterozyklischen Verbindungen vom wässerigen Medium abtrennt,
- (d) jeweils das Adsorberharz mit Methanol extrahiert und einer
HPLC-Trennung unter folgenden Bedingungen unterwirft:
Säule: Länge 125 mm und Innendurchmesser 2 mm;
Trennmaterial: Nucleosil 120-5C18 (Macherey-Nagel, Düren)
Trenntemperatur: 70 °C;
Laufmittelfluß: 0,5 ml/min;

Detektion: bei 255,4 nm; und - - im Fall einer Fermentation gemäß (b1) Fraktionen mit einer Retentionszeit Rt (min) gemäß Fig. 1 gewinnt und/oder
- - im Fall einer Fermentation gemäß (b2) Fraktionen mit einer Retentionszeit Rt (min) gemäß Fig. 2 gewinnt und
- (e) die heterozyklischen Verbindungen bzw. die heterozyklische Verbindung isoliert und gewinnt.
- (a) Sorangium cellulosum DSM 10 497 (So ce307) in a carbon and nitrogen sources and mineral salts containing aqueous medium aerobically fermented, whereby in a conventional manner
- (b1) fermented in the absence of an adsorber resin and after the fermentation an adsorber resin is added and / or
- (b2) fermented in the presence of an adsorber resin,
- (c) each separating the adsorber resin with the heterocyclic compounds formed by the fermentation from the aqueous medium,
- (d) the adsorber resin is extracted with methanol and subjected to HPLC separation under the following conditions:
Column: length 125 mm and inner diameter 2 mm;
Separating material: Nucleosil 120-5C18 (Macherey-Nagel, Düren)
Separation temperature: 70 ° C;
Solvent flow: 0.5 ml / min;
Gradient: methanol / water (45/55) from time zero to the 7th minute; then linear increase to methanol / water (70/30) up to the 20th minute; the methanol / water ratio remains constant (70/30) until the 26th minute; linear increase to methanol / water (90/10) up to the 30th minute;
Detection: at 255.4 nm; and - - in the case of a fermentation according to (b1) fractions with a retention time Rt (min) according to FIG. 1 wins and / or
- in the case of fermentation according to (b2), fractions with a retention time Rt (min) according to FIG. 2 are obtained and
- (e) the heterocyclic compounds or the heterocyclic compound is isolated and wins.
- (a) Sorangium cellulosum DSM 10 497 (So ce307) in einem Kohlenstoff- und Stickstoff-Quellen sowie Mineralsalze ent haltenden wässerigen Medium aerob fermentiert, wobei man in an sich bekannter Weise
- (b1) entweder in Gegenwart eines Adsorberharzes fermentiert oder
- (b2) nach der Fermentation ein Adsorberharz zugibt,
- (c) das Adsorberharz mit den durch die Fermentation gebildeten heterozyklischen Verbindungen vom wässerigen Medium ab trennt,
- (d) das abgetrennte Adsorberharz mit Methanol eluiert,
- (e1) das dabei zuerst anfallende stark wasserhaltige Eluat
- - einengt,
- - die anfallende wässerige Phase mit Dichlormethan extrahiert und
- - die Dichlormethanphase trocknet und zu einem Konzentrat einengt
- (e2) das restliche, weniger wasserhaltige Eluat zu einem Konzentrat einengt und mit dem gemäß (e1) angefallenen Rückstand vereinigt,
- (f) die vereinigten Konzentrate mit Heptan extrahiert und von Lipiden befreit,
- (g) den angefallenen Hexan-Extrakt mit Methanol re-extrahiert,
- (h) den methanolischen Extrakt an Kieselgel mit Methanol elu iert,
- (i) das angefallene methanolische Eluat einer HPLC-Trennung an einer Umkehrphase unterwirft, wobei man als Laufmittel ein Methanol/Wasser-Gemisch und danach Methanol verwendet und vier Fraktionen gewinnt,
- (j) die gewonnenen Fraktionen jeweils einer MPLC-Trennung an
einer Umkehrphase insbesondere unter den folgenden Bedin
gungen unterwirft:
Säule: Länge 460 mm und Innendurchmesser 40 mm;
Trennmaterial: YMC-RP 18, 120, 15 bis 25 µm;
Laufmittel: Methylalkohol/Wasser (75/25 oder 80/20);
Detektion: bei 254 nm;
eine oder mehrere Fraktionen mit einem Gehalt an heterozy klischer Verbindung bei 254 nm detektiert und isoliert und - (k) aus einer isolierten Fraktion jeweils eine heterozyklische Verbindung isoliert und gewinnt.
- (a) Sorangium cellulosum DSM 10 497 (So ce307) in a carbon and nitrogen sources and mineral salts ent-containing aqueous medium aerobically fermented, in a manner known per se
- (b1) either fermented in the presence of an adsorber resin or
- (b2) adds an adsorber resin after fermentation,
- (c) the adsorber resin with the heterocyclic compounds formed by the fermentation is separated from the aqueous medium,
- (d) the separated adsorber resin eluted with methanol,
- (e1) the first water-rich eluate
- - constricts,
- - The resulting aqueous phase is extracted with dichloromethane and
- - The dichloromethane phase dries and concentrates to a concentrate
- (e2) the remaining, less water-containing eluate is concentrated to a concentrate and combined with the residue obtained according to (e1),
- (f) the combined concentrates are extracted with heptane and freed from lipids,
- (g) re-extracting the hexane extract obtained with methanol,
- (h) eluting the methanolic extract on silica gel with methanol,
- (i) the resulting methanolic eluate is subjected to HPLC separation on a reverse phase, a methanol / water mixture and then methanol being used as the eluent and four fractions being obtained,
- (j) the fractions obtained are each subjected to MPLC separation at a reverse phase, in particular under the following conditions:
Column: length 460 mm and inner diameter 40 mm;
Separating material: YMC-RP 18, 120, 15 to 25 µm;
Mobile solvent: methyl alcohol / water (75/25 or 80/20);
Detection: at 254 nm;
detected and isolated one or more fractions containing heterocyclic compound at 254 nm and - (k) a heterocyclic compound is isolated from an isolated fraction and wins.
