Es wurde nun gefunden, daß L-Asparaginase in technischem Maßstab unter Anwendung des folgenden Verfahrens gewonnen werden kann: Nach beendetem Bakterienzüchtung wird eine organische Base mit einem Molekulargewicht über 1000 oder deren Salz zur Kulturbrühe gegeben, wodurch die Zellen ausgeflockt und leicht zentrifugierbar werden. Nach Resuspension der Zellen in Wasser werden diese durch Zugabe von Aceton erneut ausgefällt. Dabei werden die Zellen aufgeschlossen und so verändert, daß sich die gebildete L-Asparaginase im nächsten Schritt mit Wasser herauslösen läßt. Die Zellmasse wird abzentrifugiert und in Wasser suspendiert, wobei das Enzym in Lösung geht. Aus dieser Suspension werden durch Zugabe einer der oben genannten Basen die extrahierten Zellen, Nucleinsäuren und Ballastproteine ausgefällt und durch Zentrifugieren entfernt. Aus der überstehenden wässrigen Lösung kann L-Asparaginase durch Zugabe von viel Aceton gefällt werden.
