Verfahren zur Gewinnung eines in vivo antikoagulierenden-Mittels
aus dem Gift der Viper Ancistrodon Rhodostoma (Boie). Die vorliegende Erfindung
betrifft antikoagulierende Stoffe, insbesondere die Gewinnung des in dem Gift der
malayischen Viper Ancistrodon Rhodostoma (pit viper) vorhandenen def ibrinierenden
enzymartigen Mittels, dessen Herstellung und Charakterisierung in dem Patent ....

Bei dem im Hauptpatent beschriebenen Reinigungsverfahren werden schwach
basische Anionenaustauscher
als Adsorbentien für den Wirkstoff verwendet.
Geeignete Austauschermaterialien sind solche, die eine
ein
tertiäre Aminogruppe an z.B. polymeres Kohlehydrat ge;
bunden enthalten und in dieser Klasse von Adsorbentien hat sich Triäthylaminoäthylcellulose
(TEAE) bisher als das bevorzugte Material erwiesen, Es hat sich nun gezeigt, daß
infolge von Schwankungen der Eigenschaften des Handelsproduktes von Ansatz zu Ansatz
die Triäthylaminoäthylcellulose nicht in jedem Fall gemäß dem in dem Häuptpatent,
insbesondere in Beispiel 1 beschriebenenVerfahren eine genügend deutliche Trennung
zwischen der das Enzym enthaltenden Fraktion und der dieser unmittelbar vorausgehenden
Fraktion ergibt. Infolgedessen ist das nach dieser beschriebenen Arbeitsweise erhaltene
gereinigte Produkt häufig noch mit einem geringen Anteil eines in dem Gift vorhandenen
haemorrhagischen Faktors verunreinigt, dessen größerer Teil in den vorausgegangenen
Elutionsstufen eliminiert wurde. Obwohl die Verunreinigung nicht so groß ist, daß
sie bei der in Betracht gezogenen Dosierung (dose level) des Wirkstoffes gefährlich
werden kann, ist es jedoch wünschenswert, ein so reines Produkt wie möglich zu erhalten,
insbesondere wenn das Produkt durch intramuskuläre oder subcutane Injektion verabfolgt
werden soll. Der erforderliche Reinheitsgrad
kann zwar durch geeignetes
Variieren der Holaritäten der Elutionsmittel in den jeweiligen Stufen des bekannten
Reinigungsverfahrens erzielt.werden. Unglücklicherweise führt jedoch eine Abänderung

Tris/Phosphat ,uffer.._Es kann jedes beliebige geeignete
wässrige Grundlösungsmittel ge-er.hlt werden; es ist jedoch
wünschenswert,.-ein System adequater Aktivitätsstabili-
tät zu wählen,, das eine ausreichend hohe Ionenstärke .
aufweist, um-.anomale Absorptionseffekte auszuschalten,
die mit einigen Proteinen z.B. in reinem ldasser als
Grundlösungsmittel auftreten können. Angenähert isoto-
nische Lösungen wie z.B. der obengenannte Kochsalz/
Phosphat-Puffer sind 'im vorliegenden Falle frei von sol-
chen Wirkungen.
Da die@@Fraktiönierungen durch Gelfiltration auf
einem gegebenen Gel in erster Linie eine
Größe der Kolonne und des aufgegebenen Volumens sind:-;1-
ist e s wünschenswert, das letztere- auf ein so kleines- -
Volumen wie möglich zu reduzieren, um eine größ-tmÖg-
liche Trennung zu erzielen, HiePzu werden zeckm@issiger-
weise Triäthylaminoäthylcelluloseeluäte unter ver-
mindertem Druck bei etwä 30'C konzentriert. Dabei
kann bis zu einer Konzentration von 3m Tris/Phosphats
d.h. bis zu dem 30-fachen des Q,Im Tris/Phosphat
Puff ers vom pH-1-,Tert b ohne Verlust an Aktivität
einge-
engt werden. Das konzentrierte Eluat kann dann einer
Ko-
lonne von geeigneter Gröpe aufgegeben werden. Die hohe
Iönenstärke der aufgegebenen FlÜssigkeit stellt offen-
bar keinen Nachteil dar. Einige gebräuchliche
Verfahren zum Einengen wie Ultrafiltration sind hinge-
gen mit wesentlichen Verlusten an thromhinartiger Akti-
vität verbunden.
Die Erfindung wird anhand des folgenden Beispiels
näher erläutert:

Fraktionierung von Rohgif t mit Triätälaminoäthyleellulose
Die Fraktionierung wurde gemäß dem Verfahren des
Hauptpatentes vorgenommen. Auf die Ki.ution des Proteins
folgte eine U.V.Absorption auf einem kontinuierlich
arbeitenden-"Uvicord recorder" bei 254 m/u sowie
Messungen der einzelnen Fraktionen bei-280 m /u.
2 g des rohen Giftes wurden in 70 cm3 G,0lm Tris/Phos-
phat vöin"pH=Wert 825 gelöst und der unlösliche weisse

Anteil der gefärbten Lösung erscheint jedoch in Fraktion VI, nach
dem Hauptprotein und dem Maximum der thrombinartigen Aktivität, wobei jedoch noch
weiterhin bedeutsame Mengen an Aktivität eluiert werden. Aus.Messungen der optischen
Dichte bei 280,m/u und 440 m/u mit den gefärbten Fraktionen sowie durch Vergleichmit
ähnlichen Messungen an der gefärbten Fraktion VII (1-Aminosäure-. oxydase enthaltendes
Material) kann geschlossen werden, daß der gefärbte Gesamtanteil der Fraktion-VI
nicht proteingebunden ist und wahrscheinlich ein Abbauprodukt von ursprünglich in
dem rohen Gift enthaltenen Flavoproteinen ist.
Fraktionierung der Fraktion°Vh auf. Sephadex G100
Das gesammelte. Material der-Fraktion VI, das aus
2 g rohem Schlgngengif t .erhalaen. wurde . und .bis zu
1200 cm3 entbaltend 70 bis,80 % 'der ursprünglichen
thrombinartigen Aktivität des Rohgiftes ausmachte,

20 mal bei 300C eingeengt. Praktisch das gesamte Kon-
zentrat (32 cm3) wurde einer mit Sephadex G100 gefüll-
ten, 97 cm x 6 cm grossen Kolonne aufgegeben, die
mit 0,1m Natriumphosphat, 0,,1m Natriumchlorid,
pH-Wert 7,0 enthaltend 0,1 ,% Chloroform äquilibriert
worden war. Es wurde mit demselben Lösungsmittel bei.
Raumtemperatur mit 80 cm3/h weitergewaschen und
Fraktionen A 8 cm3 aufgefangen.- Es wurde jeweils
die Absorption bei 280 m/u bestimmt und darauf die
thrombinartige Aktivität kontrolliert. Die Aktivi-
tät in Thrombineinheiten,je cm3.dividi.ert durch,die
optische Dichte bei 280 m/u ergibt ein Maß für die
Stärke.
Das Peak der thrombinartigen Aktivität ist am stärk-
sten bei einem Elutionsvolumen von etwa 990 cm3 und
das Peak des_haemorrhagischen Faktors bei eineng
Volumen von 1290 cm3 mit guter Trennung zwischen
beiden. Die Fraktionen zwischen 952 cm3 und 1075 cm3,
die spezifische Aktivitäten.(Aktivitäten in
Thrombineinheiten pro cm3 dividiert. durch die_opti-
s.che , Dichte in einer f I w cm Zelle. bei .Z80 m/u)

sung, die nach dem Einengen vor dem Beladen der Kolonne
deutlich sichtbar ist, wird auf Seph$dex G100 stark
zurückgehalten, da. sie als Material mit ziemlich
niederem Holekulargewicht wandert Sie wird gut
sowohl von dem 'Peak der tlirombinartigen Aktivität
als auch dem feak des haemorrhagischen Faktors ab-
getrennt.
Die schließlich gesammelte Lösung mit thrombnar-
tiger Aktivität wurde in 1n physiologischer Kochsalz-
lösung zu: einer Aktivität von etwa 70 Thrombineinhei-
ten pro cm3 verdünnt und durch tiem-branf il tration
sternisiert (111pere y. Typ HA) 4 Die sterile. Lösung
wurde in Volumina zu je 1,1 cm3 in Standard 1 cm3 Allglas Ampullen
abgefüllt und verschlossen. Dies ergibt geeignete Einzeldosismengen für intravenöse
