Acovenoside A shows when administered intravenously in animal experiments and in humans a very strong, with Strophanthin has comparable cardiac effects, but is only very poorly effective when administered orally. The im Genin acovenoside acetylated at the Ci-hydroxyl group A (acovenoside B) is only weakly effective even intravenously. It has now been surprising found that acyl glycosides of the formula I are distinguished by good absorption and enteral effectiveness and can therefore be used for oral therapy of heart failure.
Acta, 33, S. 485 (1950), ist Acovenosid A bereits acetyliert worden.When acovenoside A was first isolated by J. v. E u w and T. Reichstein, Helvetica chim. Acta, 33, P. 485 (1950), acovenoside A has already been acetylated.
The acetyl determination of the product obtained in this way, however, showed a considerably higher value than for one Diacetate would be assumed. As can be seen from the angle of rotation and analysis values, they have Authors only received a mixture that consisted predominantly of triacetate. According to the invention With this method it is now possible for the first time to produce pure diacetylacovenoside. Through gentle acetylation of acovenoside A at room temperature in an inert solvent (for example Dimethylformamide) with acetic anhydride and tertiary amines, a mixture of the mono- and Diacetyl-acovenosids A, which can be separated by multiplicative distribution. Also in the production the remaining acyl derivatives according to the invention of acovenoside A are acylated as gently as possible, so that acylation of the secondary hydroxyl group in the steroid structure is avoided. Other suitable Acylating agents are therefore acid chlorides in pyridine or the free acid and p-toluenesulfonyl chloride in pyridine. The preparation of these new compounds is explained in detail in the following examples.
2 g Acovenosid-A werden in 30 ml Dimethylformamid gelöst. To this are added 2 ml of triethylamine and 10 ml Acetic anhydride. The reaction mixture remains at room temperature for 4 hours. After that, will diluted with ten times the amount of water, extracted by shaking with chloroform, the chloroform phase twice washed with water, dried over sodium sulfate and concentrated. The crude product contains Monoacetyl-acovenosid-A in addition to diacetyl-acovenosid-A, which by multiplicative distribution with the Phase mixture benzene-ethyl acetate-methanol-water (8: 1: 6: 3) are separated.
