Die Erfindung betrifft die Herstellung von neuen 9ß,10a-Steroiden der allgemeinen
Formel T .

bedeuten, wobei 0R eine Hydrozy, Alkozy- oder Aoyloxy-

Ein durch das Symbol R2 dargestelltes Halogenatom ist vor-
zugsweise ein Fluor-, Chlor- oder Bromatom, ein durch
das Symbol
% dargestelltes Halogenatom vorzugsweise ein Fluoratom. Eine
ge-
gebenenfalls vorhandene Alkoxygruppe in 2- oder 17-Stellung
ist
bevorzugt eine aliphatische, cycloaliphatische oder araliphatische
Alkylgruppe mit 1 - 10 C-Atomen. Beispiele
solcher Gruppen sind:
Methyl, Aethyl, Propyl, tert. Butyl, Cyclopentyl, Cyclohezyl,
Benzyl,
Cyclopenten-(1)-yloxy, 1'-Aethoxy-cyolopentyloxy und Tetrahydro-
pyranyloxy.
Eine gegebenenfalls vorhandene AGylozygruppe in 2-, 3-,
1?-
oder 21-Stellung leitet sich bevorzugt von einer gesättigten
oder un-
gesättigten aliphatischen, cycloaliphatischen, einer araliphatischen
oder einer aromatischen Carbonsäure mit 1 - 20 C-Atomen
ab. Bei-
spiele solcher Säuren sind: Ameisensäure, $seigsäure, Pivalinsäure,
Propionsäure, Buttersäure, Caproneäure, Oenentheäure, Oeleäure,
Palmitinsäure, Stearinsäure, Bernsteinsäure, Salonsäure,
Benzoe-
säure. Die durch das Symbol Z dargestellten niederen Alkyl-,
Alkenyl-
und Alkinylgruppen enthalten vorzugsweise 1 -
5 C-Atose. Beispiele
für solche Gruppen sind Methyl, Aethyl, Fropyl, iso-Propyl,
Butyl,,
iso-Butyl, Amyl, Yinyl, Allyl, 1'- und 2'-Methallyl, Aethinyl
und
Propargyl.
Die neuen 9ß,10a-Steroide der allgemeinen Formel I können nach
Methoden erhalten werden, wie sie aus der Chemie der Steroid-Normalreihe an sich

A halogen atom represented by the symbol R2 is pre-
preferably a fluorine, chlorine or bromine atom, one by the symbol

The new 9β, 10a-steroids of the general formula I can be obtained by methods such as are known per se from the chemistry of the normal steroid series.
Zur Herstellung von 2-Halogenverbindungen der allgemeinen Formel I
(R2 = Halogen), kann man z.B. ein 2-Oxalyl- oder 2-Hydroxymethylen-9ß,10a-steroid
der Teilformeln (1) bzw. (2)
in denen die gestrichelte Bindung auch hydriert sein kann, mit einem Halogenierungsmittel
wie Perchlorylfluorid, Dichlordimethylhydantoin, N-Brom- oder N-Chlorsuccinimid
oder Chlor, Brom oder Jod umsetzen und anschliessend mit einer Base behandeln (vgl.
z.B. die britische Patentschrift No. 875,822, die U.S.
Patentschrift No. 3,165,543 und No. 3,167,54e und J. Am. Chem.

Soc. 82, 284 1960». Die Verbindungen der 1'silfcrmeln (' "uid
(2) können aus 3-Keto-'ä4-und 3-Keto-g4' b-9ß,l0a-Steroiden durch Umsetzung mit
Oxalestern oder Ameisensu:1=ee@,@tern erhalten werden.
Ein 3-Keto-A 4'6-9ß,10a-Steroid kann auch direkt mit N-Brom-
oder N-Chlorsuccinimid in Dioxan in ein 2-Brom-(bzw. 2-Chlor-)
3-Keto_44,6-9ß,lOa-Steroid überführt werden.
Durch Umsetzung eines 3-Keto-L1lt4#6-9ß,10a-Steroide mit
Chlor oder Brom werden 1,2-Dichlor (bzw..1,2-Dibrom)-3-keto-
d496-9ß,10a-Steroide erhalten, die bei Dehydrohalogenierung,
z.B. durch Behandlung mit Pyridin,2-Chlor"(2-Brom)-3-Keto-A

9ß,10a-Steroide liefern (vgl. J. Chem. Soc. 1958, 1324).
Ein 6-Brom-3-Keto-d4-9ß,10a-Steroid wird durch Behandlung
mit einem Ohlorierungsmittel, wie Chlor, in ein 2,2-Dichlor-6-
brom-3-keto- d 4-9ß,10a-Steroid übergeführt. Dehydrohalogenierung
mit Lithiumchlorid in Dimethylformamid liefert*dann ein 2-Chlor-
3 Keto- a 1'4'6-9ß,10a-Steroid (vgl. J. Chem. Soc. 1958, 1324).
Ein 2-Chlor-(bzw. 2-Brom)-3-Keto- A 4'6-9ß,10a-Steroid
kann auch
aus einem 3-Acyloxy-2'4'6=9ß,10a-Steroid durch Behandlung mit
einem Chlorierungs- bzw. Bromierungsmittel, wie Chlor oder
Brom,
in Alkohol oder Eisessig in Gegenwart von Kaliumacetat oder
durch Behandlung mit N-Bromsuccinimid erhalten werden. Ausgehend
tOxydation und DehZdrobromierung mit Collidirl
von einem 3-Hydroxy-Lni5-9ß,10a-Steroid kann durch . rotem

ein 2-Brom-3-Keto- A 1,4,6-9ß,10a-Steroid erhalten werden
[vgl.
J. Am. Chem. Soc. M, 3305 (1955)].
Nach einer bevorzugten Arbeitsweise setzt man ein 3-Keto-
h- oder ein 3-Keto-I& 4P6-9ß,10a-Steroid mit einem Oxalsäureester
oder mit einem Ameisensäureester, besonders dem Aethylester, in Lösungsmitteln,
z.B. aromatischen Kohlenwasserstoffen, wie Benzol, Toluol, Xylol, oder in Pyridin,
bei Temperaturen zwischen Raumtemperatur und der Siedetemperatur des Lösungsmittels,
vorzugsweise bei 60 - 70o.in Gegenwart einer Base, wie Natriummethylat oder -äthylat,
Natriumhydrid oder Pyridin zu einer 2-Oxalyl- oder einer 2-Hydroxymethylenverbindung
der oben angeführten Teilformeln (1) und (2) um. Die 2-Oxalyl- bzw. 2-Hydroxymethylenverbindungen
werden dann, gegebenenfalls unter Schutz einer vorhandenen-20-Ketogruppe,mit einem
Halogenierungamittel, z.B. einem N-Haloamid bzw. -imid, wie N-Brom- oder N-Chlorsuccinimid,
N-Bromacetamid, Chloramin T oder Dimethyldichlorhydantoin in einem Lösungsmittel,
wie Aceton, Alkohol oder Dioxan, bei etwa -20o bis +20o in Gegenwart einer Base,
z.B. einem Alkalialkoxyd, wie Natriummethylat, oder einem Acetat, wie Kaliumacetat,
umgesetzt.
Zur Einführung einer 2-Hydroxy- oder 2-Acyloxygruppe
kann man z.B. wie folgt verfahren: Ein 3-Keto-4,5-epoxy-9ß,lOa-Steroid wird mit
Eisessig unter Rückfluss erhitzt, wobei 2-Acetoxy-9ß,10a-Steroide erhalten werden.
Spaltung des Epoxyds durch längeres Erhitzen mit Schwefelsäure in Aceton gibt 2-Hydroxy-9ß,10a-Steroide
(vgl. die amerikanische Patentschrift No. 2,910,487). Eine 2-Acetoxygruppe kann
weiter durch Behandlung von 3-Keto-o'-
6-Brom-9ß,l0a-Steroiden mit Eisessig unter Rückfluss in
amerikanisc
Stickstoffatvmosphäre vgl. die Patentschrift No. 2°857'406),
durch Behandlung von 3-Keto-.d 4- oder 3-Keto-g'6-9ß,l0a-
Steroiden mit Bleitetraacetat in Eisessig (vgl. die amerikani-
sche Patentschrift No. 2,847,428) oder mit N-Bromsuccinimid
in Tetrachlorkohlenstofflgefolgt von der Umsetzung mit Kalium-
acetat),eingeführt werden. Behandlung von 3 Keto-d 4- oder
3-Keto- ƒ4'6-9ß,10cx-Steroiden mit Selendioxyd in tert.
Butanol
und Eisessig gibt 2-Hydroxy-3-keto-i`4- oder 2-Hydroxy-3-
Keto-,6 1' 9 6-9P,10a-Steroide [vgl. J. Am. Vhem. Soe.
80.
1687 (l958)). Aus 3-Keto-,d l'4-9ß,10a-Steroiden werden durch
A
Behandlung mit Osmiumtet ..oxyd in Gegenwart von Pyridin 1,2-
Dihydroxyverbindungen erhalten, die durch Dehydratisierung
in 3-Keto-2-hydrozy ,d 1'4-9p,10a-Steroide übergeführt
werden
können (vgl. die amerikanische Patentschrift No.-3,079,383).
2-Hydroxyverbindungen der allgemeinen Formel I können
ferner durch basische Verseifung von entsprechenden 2-Acetoxy-
verbindungen gewonnen werden. Als lösungamittel kommen dafür
r.B. Alkohole, wie Methanol oder Aethanol, oder wässriges
Dioxan in Betracht, als Basen Alkali- oder Brdalkalicarbonate,
wie Kaliumcarbonat, Kaliumhydrogencarbonat oder Magnesium-
carbonat. 1,2-gesättigte 2-Hydroxyverbindungen der allgemeinen
Formel I können durch Behandlung mit Wismuttrioxyd (vgl. J.
Am. Chem. Soc. 80, 1687 (1958)1 oder auf aikrobiologischem
Weg
amerikanische P$tentsohriftl
(vgl.@e3.087.864) in 1,2-Stellung dehydriert werden.
Die freien 2-Hydroxygruppen können in üblicher Weise verestert
werden, z.B. durch Behandlung mit reaktionafäUgen Säurederivaten,
wie Säureanhydriden, in Gegenwart einer Base.
2-Älkoxyverbindungen der allgemeinen Foriie1 I lassen sich
aus den entsprechenden ?--Hydroxyverbindungen durch Verätherung
erhalten, z.B. durch Umsetzung mit einem Alkylhalogenid, wie
Methyljodid, in Gegenwart einer Base, wie Kaliuin-tert.-butylat.
Eine 2-Alkoxygruppe kann ferner durch Umsetzung einer 2-Brom-
verbindungen mit einem Alkalialkoxyd, z.B. Natriummethylat,
eingeführt werden.
In den so erhaltenen Verbindu%gen können Doppelbindungen
in 1- und (sofern nicht bereits vorhanden) 6-Stellung einge-
führt werden, wozu ebenfalls die aus der Normalreihe der
Steroide bekannten Methoden Anwendung finden können, beispiels-
weise die Behandlung mit Dehydrierungsmitteln, wie 2,3-Dichlor-
oder 2,3-Dibrom-5,6-dicyanobenzochinon oder eine Reaktionsfolge
von Bromierung und Dehydrobromierung. Eine d 6-Doppelbindung
kann weiter mittels Chloranil, eine Al-Doppelbindung auf
mikrobiologischem Weg eingeführt werden.
Ein 3-Keto-@4-System kann weiterhin in ein 3-Acyloxy- d
3'5-System überführt werden, z.B. durch Behandlung mit einem
Acylierungsmittel, wie Ieoprepenylacetat in Gegenwart von p-
2oluolsulfonsäure.
Die erfindungsgemäss erhältlichen neuen 9ß,10a-Steroide
'der Formel I sind hormonal oder antihormonal wirksam. So zeigt
z.B. das 2ß-Fluor-17ß-hydroxy-9ß,l0a-androst-4-en-3-on uterotrophe
Wirksamkeit, das 2ß-Fluor-17a-methyl-17ß-hydroxy-9ß,l0a-androsta-
4,6-dien-3-onjdas 2ß-Chlor-17a-methyl-17ß-acetoxy-9ß,l0a-androsta-
1,4,6-trien-3-on und das 2ß-Brom-17a-methyl-17ß-acetoxy-9ß,l0a-
androsta-1,4,6-trien-3-on anthonadotrope Wirksamkeit und das
2ß-
Chlor-9ß,l0a-pregna-1,4,6-trien-3,20-dion anti-androgene Wirksam-
keit. Die Verfahrensprodukte können als Heilmittel z.B. in
Form
pharmazeutischer Präparate Verwendung finden, welche sie in
Mischung
mit einem für die enterale oder parenterale Applikation geeigneten
pharmazeutischen, organischen oder anorganischen inerten Träger-
material, wie z.B. Wasser; Gelatine, Milchzucker, Stärke, MagneBium-
stearat, Talk,-pflanzliche Oele, Gummi, Polyalkylenglykole,
Vaseline
usw. enthalten. Die pharmazeutischen Präparate können in fester
Form, z.B. als Tabletten, Dragees, Suppositorien, Kapseln,
oder in
flüssiger Form, z.B. als Lösungen, Suspensionen oder Emulsionen,
vorliegen. Gegebenenfalls sind sie sterilisiert und bzw. oder
enthalten Hilfsstoffe, wie Konservierungs-, Stabilisierungs,
Netz- oder Emulgiermittel, Salze zur Veränderung des osmotischen
Druckes oder Puffer. Sie können auch noch andere therapeutisch
wertvolle Stoffe enthalten.
In den nachfolgenden Beispielen sind die Temperaturen in
Celsiusgraden angegeben.
Beispiel 1
Eine Lösung aus 4,01 g Natrium und 70 ml Methanol wurde
mit 500 ml Benzol versetzt. Danach wurde das überschüssige
Methanol azeotrop abdestilliert. Die resultierende Suspension
wurde mit 13,2 ml Aethylformiat und einer Lösung von 25 g 9ß,
10a-Androst-4-en-3,17-dion in 250 ml Benzol versetzt und 20
Minuten am Rückfluss erhitzt. Nach Kühlen wurde filirriert,
der
Rückstand mit Benzol gewaschen und getrocknet, wobei 24 g
plattenchromatographisch einheitliches Natriumsalz von 2-Hydroxy-
methylen-9ß,l0a-androst-4-en-3,17-dion erhalten wurden, UV:
Ä maz
244 und 303 nm,F = 8300 und 14700.
Zu einer Lösung von 24 g dieser Verbindung in 750 ml Methanol
wurde bei -20o eine Lösung aus 2,1 g Natrium und 85 ml Methanol
gegeben und danach während 80 Minuten Perchlorylfluorid einge-
leitet. Anschliessend wurde nach Entfernen des Kühlbades das
überschüssige Perchlorylfluorid mit Stickstoff aus der Lösung
ausgeblasen. Die Reaktionslösung wurde dann auf die Hälfte
ein-
geengt, mit 47 g wasserfreiem Kaliumacetat versetzt und 90
Minuten
am Rückfluss erhitzt. Nach Filtrieren wurde die Lösung mit
Essig-
ester extrahiert, der Extrakt mit Wasser, 0,5 N Natronlauge
und
Wasser gewaschen, getrocknet und eingedampft. Das erhaltene
gelbe
Oel (9,5 g) wurde an Kieselgel chromatographiert (25 ml-Fraktionen),
Die Fraktionen 195 - 310 enthielten ein Gemisch des 2a- und
des
2ß-Fluor-9ß,l0a-androst-4-en-3,17-dions. Die Fraktionen 311
- 488
enthielten fast reines 2ß-Isomeres, das nach Umkristallisieren
aus Essigester-Isopropyläther reines 2ß-Fluor-9ß,l0a-androst-
4-en-3,17-dion lieferte. Schmelzpunkt:l32 - 1330, UY:%max240
nm,

Beispiel 2
Eine Lösung von 5,8 g gß,10a-Testosteron in 70 ml Pyridin
wurde mit 20 ml Aethylformiat versetzt und nach Zugabe einer
Lösung aus 940 mg Natrium und 10 ml Methanol 24 Stunden bei
Raumtemperatur gerührt. Das Reaktionsgemisch wurde in Aether
auf-
genommen. Der Extrakt wurde mit 200 ml 20%iger Essigsäure ge-
waschen und sodann mit drei 200 ml 4%iger Natronlauge aus-
gezogen. Der alkalische Extrakt wurde mit Essigsäure angesäuert
und mit Essigester re-extrahiert. Nach Aufarbeiten des Essigester-
extraktes wurden 5,0 g rohes 2-Hydroxymethylen-17ß-hydroxy-9ß,l0a-
androst-4-en-3-on erhalten" UV: %max243 und 304 nm,
t = 8800 und
14200.
Durch Behandlung dieser Verbindung mit Perchlorylfluorid ge-
mäss Beispiel 1 wurde ds.a 2ß-Fluor-17ß-hydroxy-9ß,l0a-androst-4-
en-3-on erhalten. Schmelzpunkt: 164 - 1660 (aus Methylenchlorid-
Isopropyläther), UV: h max242 nm, f= 15000
Beispiel 3
Analog dem im Beispiel 1 beschriebenen Verfahren wurde
17a-Aethinyl-9ß,l0a-testosteron in das Natriumsalz von 2-Hydroxy-
methylen-17a-äthinyl-17ß-hydroxy-9ß,l0a-androst-4-en-3-on über-
geführt. Durch Behandlung dieser Verbindung mit Perchloryl-
fluorid gemäss Beispiel 1 wurde 2ß-Fluor-17a-äthinyl-17ß-hydroxy-
9ß,10a-androst-4-en-3-on erhalten. Schmelzpunkt: 146 - 150°,
UV: Ä max241 nm, = 14200.
Diese Verbindung wurde auch durch Umsetzung des im Beispiel
1
beschriebenen 2ß-Fluor-9ß,l0a-androst-4-en-3-on mit Acetylen-Kalium
in flüssigem Ammoniak erhalten.
Beispiel 4
500 mg des gemäss Beispiel 1 erhaltenen 2ß-Fluor-9ß,10a-
androst-4-en-3,17-dions wurden in 50 ml absolutem Tetrahydrofuran
gelöst und tropfenweise mit einer Suspension von 500 mg Lithium-
aluminiumhydrid in 80 ml Aether versetzt. Nach 50 Minuten wurde
das überschüssige Reagens mit feuchtem Aether zersetzt, das
Reak-
tionsgemisch filtriert und das Filtrat eingeengt. Der Rückstand
wurde in Dioxan aufgenommen und nach Zusatz von 600 mg 2,3-Dichlor-
5,6-dicyanobenzochinon 3 Stunden in Stickstoffatmosphäre gerührt.
Nach Reduktion von überschüssigem Dehydrierungsmittel mittels
S02
und Aufarbeitung wurden 320 mg 2ß-Fluor-17ß-hydroxy-9ß,l0a-androst-
4-en-3-on erhalten, das mit dem gemäss Beispiel 2 erhaltenen
' Präparat identisch War.
Beispiel 5
Analog dem im Beispiel 1 beschriebenen Verfahren wurde aus
17ß-Hydroxy-9ß,l0a-androsta-4,6-dien-3-on das Natriumsalz von
2-Hydroxymethylen-17ß-hydroxy-9ß,l0a-androsta-4,6-dien-3-on
vom
Schmelzpunkt (roh) 244 - 2460 (zerr.); UV:X max288 nm, £ =
11020
erhalten, das mit Perchlorylfluorid 2ac-Fluor-17ß-hydroxy-9ß,1.0a-
androsta-4,6-dien-3-on, Schmelzpunkt 164 - 165o, UV:Xm 295
nm,
Ein 25300, vi D5 = -566o (in Dioxan) und 2ß-Fluor-17ß-hydroxy-9ß,
10a-androsta-4,6-dien-3-on, Schmelzpunkt 146 - 147o,

Beispiel 6
Eine Lösung von 200 mg 2ß-Fluor-17ß-hydroxy-9ß,l0a-androsta-
4,6-dien-3-on in 10 ml Aceton wurde bis zur bleibenden Orangefär-
bung mit Jone$-Reagens versetzt. Nach Aufarbeitung wurden 150
mg
2ß-Fluor-9#910a-endrosta-4,6-dien-3917-dion erhalten.
Schmelz-

Dioxan).
Beispiel 7
' 3,42 g 17a-Methyl-17ß-acetoxy-9ß,l0a-androsta-4,6-dien-
3-on wurden,mit 1,5 ml Aethylformiat und 1,1 g Natriummethylat
in 40 ml Benzol 5 Minuten am Rückfluss erhitzt. Nach Abkühlen
des Reaktionsgemisches wurde das ausgefallene Gemisch der Na-
triumsalze der Hydroxymethylenverbindungen abfiltriert und
in
Wasser gelöst. Die Lösung wurde mit 4 N Salzsäure angesäuert
und mit Methylenchlorid extrahiert. Nach Aufarbeiten des Extraktes
wurde an Silicagel chromatographiert, wobei 2-Hydroxymethylen-
17ß-methyl-17ß-acetoxy-9ß,l0a-androsta-4,6-dien-3-on, Schmelz-
punkt:128o (aus Aether), UV:X max293 nm £ = 18300,und
2-Hydroxy-
methylen-17a-methyl-17ß-hydroxy-9ß,l0a-androsta-4,6-dien-3-on,
Schmelzpunkt 135 - 1380 (aus Aether), üV:A max296 nm,
t = 16500"-
erhalten wurden. Aus der letzteren Ver-
bindung wurde gemäss Beispiel 1 2a-Fluor-17a-methyl-17ß-hydroxy-
9ß,10a-androsta-4,6-dien-3-on, Schmelzpunkt: 122 -
1230 (aus

Beispiel 9
Zu einer Suspension von 1,19 g Natriummethylat in 120 ml
Benzol wurden bei 25o 5,0 g Oxalsäurediäthylester gegeben.
Die
so erhaltene Lösung wurde mit 3,12 g 9ß,10a-Pregna-4,6-dien-
3,20-dion versetzt und 18 Stunden gerührt. Anschliessend wurde
das Reaktionsgemisch mit 150 ml Aether verdünnt, der Niederschlag
abfiltriert, mit Aether gewaschen und in Vakuum bei 50o getrocknet.
Das so erhaltene Rohprodukt wurde gemäss Beispiel 1 fluoriert.
Aus dem Reaktionsgemisch wurde durch Chromatographie 2ß-Fluor-
9ß,10a-pregna-4,6-dien-3,20-dion, Schmelzpunkt: 156 - 157o,
W: #\ Max 286 nm, t= 26000; isoliert.
Beispiel 10
400 mg 2ß-Fluor-17ß-hydroxy-9ß,l0a-androst-4-en-3-on wurden
mit 500 mg Chloranil in 30 ml tert. Butanol 16 Stunden unter
Stickstoff am Rückfluss erhitzt. Nach Aufarbeitung und
Chromatographie wurden 125 mg 2ß-Fluor-17ß-hydroxy-9ß,l0a=-androsta-
4,6-dien-3-on erhalten, das mit dem gemäss Beispiel 5 erhal-
tenen Präparat identisch war.
Beispiel 11
5 g des gemäss Beispiel 2 erhaltenen 2ß-F'luor-17ß-hydrozy-
9ß,10a-androst-4-en-3-on wurden in 175 ml Benzol gelöst.
Nach
Zusatz von 0,5 g p-Toluolsulfonsäure-wurden aus der Lösung
langsam 600 ml unter fortlaufendem Brsatz des Destillats
durch
frisches Benzol abdestilliert.
Danach wurden 12,5 ml tsopropenylacetat zugegeben und
das Reaktionsgemisoh wurde 2 Stunden am Rückfluss erhitzt.
Nach nochmaligem Isopropenylacetatzusatt und Brhitzen wurden
aus dem Reaktionsgemisch 100 ml abdestilliert, 0,35 ml Pyridin
zugesetzt und der Niederschlag abfiltriett. Aus dem Filtrat
wurden nach Bindampfen und.Chromatographie des Rückstandes
4,5 g
2ß-Fluor-3,17ß-acetoay-9ß,l0a-androsta-3,5-dien erhalten,

Beispiel 12
Eine Lösung von 24 g 17ß-Aeetoxy-9ß,l0a-androst-4-en-3-on
in 170 ml Eisessig wurde bei Raumtemperatur zu einer Lösung
von
35 g Bleitetraacetat in 420 ml Eisessig gegeben. Das
Reaktionsgemisch wurde unter Stickstoff auf 90o erhitzt, nach
3 Stunden mit weiteren 30 g Bleitetraacetat versetzt und noch
18 Stunden gerührt. Danach wurde das Reaktionsgemisch im
Vakuum eingeengt. Nach Aufarbeitung des Rückstandes und
Chromatographie an Kieselgel wurden 3,2 g 2a,17ß-Diacetoxy-9ß.

Beispiel 13
Zu einer Lösung von 1,50 g 17a-Methyl-17ß-acetoxy-9ß,l0a-
androeta-1,4,6-trien-3-on in 3 m1 Methylenchlorid und 40 ml
Asther wurde bei -200 eine Lösung von 370 mg Chlor in 5 ml
Eisessig gegeben. Die Mischung wurde 5 Stunden bei -200 gehalten,
dann auf Wasser gegossen und mit Methylenchlorid
extrahiert. Der
mit Wasser neutral gewaschene Extrakt Wurde getrocknet, ein-
gedampft und der Rückstand mit 10 ml Pyridin 30
Minuten bei
Zimmdrteaperatur gehalten. 8B wurde auf
Salzsäure gegoanen und mit Asther extrahiert. Durch Umkriatal-

mid und 0,7 ml 70 96 Perchlorsäure gegeben. Man hielt die Lösung
5 Stunden bei Zimmertemperatur, goss sie auf Eiswasser und
extrahierte mit Asther. Der Eitrakt wurde mit 2 N NaOH-Lösung
und Wasser neutral gewaschen, mit Natriumsulfat getrocknet
und
eingedampft. Der Rückstand wurde an 250 g Silicagel chromato-
graphiert. Die nach Dümnschichtchromatographie einheitlichen
Benzol-Aceton- (97:3)-Fraktionen wurden aus Aceton-Isopropyläther
umkristallisiert und lieferten 2ß-Brom-17a-methyl-17ß-hydroxy-9ß,
10a-androsta-4,6-dien-3-on. Schmelzpunkt: 119 - 121o.

aus Aceton-Hexan umkristallisiert, 2ß-Chlor-17a-methyl-17ß-acetoxy-
9ß,10a-androsta-4,6-dien-3-oh. Schmelzpunkt: 173 - 1740
