Um ungefähr den gleichen Prozentsatz an Wirksamkeit zu erzielen, muß 10 mg/kg Codein gegenüber 5 mg/kg der Stoffe A, B, C und D angewendet werden, und überdies besitzen letztere eine sechsmal größere Aktivität als 1-Phenylcyclopentan-l-carbonsäure-/}-[(ß'-diäthylamino)-äthoxy]-äthylester. Die organische Schicht wird anschließend mit Natriumsulfat getrocknet, das Lösungsmittel abgedampft und der Rückstand unter vermindertem Druck destilliert. Eine Mischung aus 22,4 g 4-Phenyl-tetrahydropyran
- 4 - carbonsäurechlorid und 15,9 g 5-(N-Diäthylamino)-pentanol-(2)
in 100 ml wasserfreiem Toluol wird 5 Stunden unter Rückfluß erhitzt. Die Mischung wird dann abgekühlt und hierauf mit
100 ml Wasser und 50 ml einer 10%igen wäßrigen Natriumhydroxydlösung versetzt. Die Toluolschicht
wird abdekantiert, dreimal mit je 100 ml Wasser gewaschen, dann wird die Toluolschicht über Natriumsulfat
getrocknet und das Lösungsmittel zur Trockne abgedampft. Durch Destillation des Rückstandes
erhält man den 4-Phenyl-tetrahydropyran-4-(5'-diäthylamino-pentyl-2')-carbonsäureester
vom Kp.0,01 150 bis 152° C.
Der oben erhaltene rohe Rückstand wird in 50 ml Äthanol gelöst, zu dieser Lösung eine heiße Lösung
von 19,2 g Zitronensäure in 100 ml Äthanol hinzugegeben, dann das Ganze abgekühlt und mit einer
kleinen Menge Äther versetzt. Es werden 38,5 g 4 - Phenyl - tetrahydropyran - 4 - (5' - diäthylaminopentyl-2')-carbonsäureestercitrat
vom F. 1000C erhalten.
