Der qualitative und quantitative Nachweis von Eiweiß in biologischen Flüssigkeiten, z. B. im Urin, hat eine große diagnostische Bedeutung, insbesondere bei Nierenerkrankungen, aber auch bei Kreislaufschwäche, Arteriosklerose, Hypertonie, Diabetes. Es sind eine Reihe von Methoden zum Nachweis von Eiweiß bekanntgeworden, ζ. B. die Sulfosalicylsäureprobe, Essigsäure-Kochprobe, Salpetersäure-Ringprobe, Pikrinsäureprobe; alle diese Methoden beruhen auf Fällungsreaktionen und sind ohne Laboratoriumsgeräte nicht durchführbar. Für schnell-diagnostische Zwecke kommen diese Reaktionen daher nicht in Betracht; hierfür benötigt man eine einfache Farbreaktion, welche auch durch ungeschultes Personal routinemäßig durchgeführt werden kann, z. Diese Reaktion nutzt den bei verschiedenen Indikatoren bekannten sogenannten »Proteinfehler« aus: Wird der pH-Wert des Reagenzes unterhalb des Indikatorumschlagbereiches gehalten, so tritt bei Zugabe von Eiweißlösung eine Farbänderung ein, die vom pH-Wert unabhängig ist und durch den Eiweißgehalt bedingt wird; die Farbintensität der entstehenden Farbe ist abhängig von der Konzentration des Eiweiß in der zu prüfenden Lösung. F e i g 1 arbeitete diesen Eiweißnachweis als Tüpfelreaktion aus, wobei er als Reagenzien das Kaliumsalz des Tetrabromphenolphthaleinäthylesters und Essigsäure verwendete. Zur Herstellung von Eiweißtestpapieren auf Basis der Feigl-Reaktion muß natürlich die Essigsäure durch eine nichtflüchtige Säure ersetzt werden; nach den Verfahren der britischen Patentschriften 814 223, 826 066 und 840 362 verwendet man hierfür zweckmäßig eine saure Puffersubstanz, wie z. B. Citratpuffer. Die auf diese Weise erhaltenen Eiweißteststreifen befriedigen jedoch nicht völlig, da die Farbkontraste von negativer und positiver Eiweißprobe in Konzentrationsbereichen unterhalb von 0,1 °/0 Eiweiß sehr wenig differieren; der positive: Test fällt mehr oder weniger stark gelbgrün aus, der negative Test ist grüngelb und ist dunkler als der trockene Teststreifen. Auch läßt die Empfindlichkeit dieser Testpapiere einiges zu wünschen übrig (Nachweisgrenze etwa 0,03 bis 0,01 °/o Eiweiß). Es ist für den Mediziner aber erforderlich, Eiweiß im Harn in derselben Verdünnung nachweisen zu können, wie es mit der Essigsäure-Kochprobe möglich ist; demnach müssen noch 0,005 °/o Eiweiß im Harn einwandfrei diagnostiziert werden können.A color reaction suitable for the detection of protein was first used by F eig 1 et al. Patentschriften wird der saugfähige Träger mit einer Lösung imprägniert, welche die Puffersubstanz zusammen mit dem Farbstoffindikator enthält. Es wurde nun überraschenderweise gefunden, daß man wesentlich empfindlicher reagierende Eiweißtestpapiere erhält, wenn man die Puffersubstanz und den FarbstofF-indikator getrennt, d. h. nacheinander auf den Träger aufbringt. Die hierbei erhaltenen Testpapiere sind schwach gelb gefärbt und geben mit eiweißhaltigem Urin kräftige Blaufärbungen, die entsprechend der Eiweißkonzentration abgestuft sind und sich deutlich von der gelben Färbung des negativen Testes abheben. Es können bis 0,005 °/0 Eiweiß einwandfrei nachgewiesen werden; selbst geringere Eiweißkonzentrationen lassen sich noch feststellen.
