L-( — )-a-Methyl-ß-(3,4-dihydroxy-phenyl)-alanin, welches auch unter der Bezeichnung »a-Methyl-DOPA« bekanntgeworden ist, hat in letzter Zeit eine besondere Bedeutung als blutdrucksenkendes Mittel erlangt. Diese Verbindung in racemischer Form wurde erstmals von Stein et al., J. Am. Chem. Soc, 77, 1955, S. 700 bis 703, bzw. in der entsprechenden USA.-Patentschrift 2 868 818 beschrieben. Nach dem Trocknen im Vakuum bei 1100C schmilzt das Produkt bei 190 bis 1910C. Man verdampft zur Trockne, verrührt den Rückstand bei OC mit wenig Wasser, dem etwas wäßrige Schwefeldioxydlösung zugefügt ist, stellt mit eiskaltem 6 η-Ammoniak, der 0,1 g Natriumbisulfit enthält, auf pH 4,5 ein, rührt noch 1 Stunde bei 00C und saugt den erhaltenen Kristallbrei ab. Die Umkristallisation erfolgt wie unter a) beschrieben. Ausbeute 85% der Theorie. Schmelzpunkt 310 bis 3110C (Zersetzung).50 g of L- () -5- (3,4-dimethoxybenzyl) -5-methylhydantoin-3-acetic acid are refluxed for 6 hours with 300 ecm of 48% hydrobromic acid while simultaneously passing through hydrogen bromide.
