Description

Verfahren zur Gewinnung von Dextransucrase Die Herstellung von Dextran geschieht bekanntlich dadurch, daß man eine Sacch:arose enthaltende Nährlösung mit einer Kultur einer Leuconostocart beimpft. Hierbei bildet sich durch das Enzym Dextransucrase aus der Saccharose durch einen Gärprozeß Dextran. Ein Nachteil dieser Arbeitsweise besteht darin, daß die Konzentration an Saccharose in der Ferrnentationslösung verhältnismäßig niedrig gehalten werden mwß. Insofern sind die hierbei zu handhabenden -Volumenmengen an dem zur Ausfällung des gebildeten Dextrans notwendigen Fällmittel verhältnismäßig groß, so daß das Verfahren insgesamt wenig wirtschaftlich ist.Process for the production of dextran sucrase The production of dextran is known to happen by using a nutrient solution containing sacch: arose inoculated into a culture of a Leuconostocart. Die Erfindung schlägt nun vor, daß das in dem vorgenannten Substrat gebildete Enzym zusammen mit dem in der Nährlösung bereits bei der Fermentation gebildeten Dextran mit Äthylalkohol ausgefällt wird. Auf diese Weise wird eine hohe Enzymausbeute erhalten, wobei noch von besonderer Wichtigkeit ist, daß das bei der erfindungsgemäßen Ausfällung mit Äthylalkohol ebenfalls ausfallende, bereits bei der Fermentation gebildete Dextran eine ausgezeichnete Schutzwirkung auf das Enzym ausübt. Zweckmäßigerweise wird die Fällung bei niedrigen Temperaturen durchgeführt, was sich im Sinne einer Steigerung der Enzymausbeute auswirkt. Das so erhaltene Enzymkonzentrat kann in an sich bekannter Weise zur Erzeugung von Dextran aus Saccharose eingesetzt werden. Beispiel Eine Kultur wird durch Züchten einer Dextran aus Rohrzucker bildenden Leuconostocart, zweckmäßigerweise Leuconostoc mesenteroides, hergestellt, und zwar unter Bedingungen, welche einen hohen Enzym-Behalt in der Kultur ergeben. Nach 96 Stunden, wonach die Enzymaktivität in der Kulturlösung konstant bleibt, wird die Lösung auf -f- 4° C abgekühlt. Gleichzeitig wird ein viermal größeres Volumen 96o/oigen Äthylalkohols auf dieselbe oder niedrigere Temperatur gekühlt. Die Kulturlösung und der Alkohol werden vermischt, worauf die Mischung während einer Stunde bei -I-4° C stehengelassen wird, wobei ein Niederschlag entsteht, der hauptsächlich aus Eiweiß, Enzym und Dextran besteht. Der Alkohol wird absiphoniert und die Fällung bei Zimmertemperatur unter starkem Umrühren in einer 0,2 n Phosphatpufferlösung bei pg 5,5 gelöst. PATENTANSPRUCH: Verfahren zur Gewinnung von Dextransucrase zur Herstellung von Dextranen durch Züchten von Leuconostoc mesenteroides in einem gepufferten, wäßrigen, Saccharose und hydrolysiertes Protein enthaltenden Medium und Ausfällen des gebildeten Enzyms, dadurch gekennzeichnet, daß man das im Substrat gebildete Enzym zusammen mit dem in der Nährlösung bereits bei der Fermentation gebildeten Dextran mit Äthylalkohol ausfällt.
