Verfahren zur Isolierung von Oxyphenylbenzo-y-pyronderivaten aus Weißdorn
Gegenstand der Hauptpatentanmeldung Sch 20651 IVa/30h ist ein Verfahren zur Gewinnung
von Hydroxyderivaten der Phenylbenzo-y-pyrone durch Extrahieren von Pflanzenteilen
des Weißdorns mit Wasser und/oder organischen, mit Wasser mischbaren Lösungsmitteln,
bei welchem durch Zusatz von Wasser zum erhaltenen Extrakt das Chlorophyll und vorhandene
Wachse und Fett abgeschieden und anschließend im Filtrat unter stufenweiser Veränderung
der Wasserstoffionenkonzentration beginnend im sauren p11-Bereich und im alkalischen
pg-Bereich endend, mit Schwermetalisalzen, die Hydroxyderivate der Phenylbenzo-y-pyrone
getrennt als Salze gefällt und nach Zerlegung der Sch`vermetallsalze isoliert werden.Process for the isolation of oxyphenylbenzo-y-pyrone derivatives from hawthorn

In Weiterentwicklung dieses Erfindungsgedankens konnte ein neues Verfahren
entwickelt werden, mit welchem das besonders wertvolle Vitexin-4'-rhamnosid, Schmp.
215° C, ein Flavonglycosid, das nach dem Verfahren der Hauptpatentanmeldung gemäß
Beispiel 1 in der Fraktion D erhalten ist, in besonders reiner Form erhalten werden
kann. Im Gegensatz zu den üblichen Verfahren zur Gewinnung von Flavonglucosiden
durch Ausfällen mit Bleisalzen in neutralem, saurem oder schwach alkalischem Milieu
erfolgt, wie gemäß der Hauptpatentanmeldung gefunden wurde, eine Abscheidung von
Vitexin-4'-rhamosid nur durch Verwendung basischer Bleisalze in stark alkalischer
Lösung. Die gleichfalls vorhandenen Ballaststoffe und anderen Flavons bzw. Flavonglucosi.de
können vorher durch stufenweise Änderung der Wasserstoffionenkonzentration abgetrennt
werden, so daß schließlich das Vitexin-4'-rhamnosid aus der auf eine stark alkalische
Reaktion gebrachten Lösung und einem gegebenenfalls noch erforderlichen Zusatz von
basischem Bleiacetat ausgefällt wird. Die Bleifällung des Vitexin-4'-rhamnosids
wird durch Filtrieren oder Zentrifugieren entfernt und durch Waschen mit Wasser
von anderen gelösten Stoffen befreit. Der hellgelbe Bleiniederschlag wird mit solchen
Säuren, wie Schwefelsäure, Phosphorsäure oder Schwefelwasserstoff, zerlegt, die
in Wasser oder Wasser-Alkohol-Gemischen praktisch unlösliche Bleiverbindungen bilden
und dadurch von den in Wasser oder Wasser-Alkohol-Gemischen leicht löslichen Vitexin-4'-rhamnosid
abgetrennt werden können. Die das Flavon enthaltende Lösung wird unter vermindertem
Druck von einem Teil des Wassers befreit, bis ein dünnflüssiger Sirup entsteht.
Dem Sirup wird das Vitexin-4'-rhamnosid nun erfindungsgemäß durch wiederholte Extraktion
mit Butyl- oder Amylalkohol entzogen und die Alkanolauszüge im Vakuum vom Lösungsmittef
bei niederer Temperatur befreit. Die Rückstände sind von gelber bis schwach brauner
Farbe und enthalten das Vitexin-4'-rhamnosid bereits in hochkonzentrierter Form.
Die ersten Alkanolauszüge enthalten vor allem noch geringe braungefärbte Verunreinigungen,
die noch in der wäßrigen Phase vorhanden sind. Nach der Extraktion der braungefärbten
Begleitstoffe sind die dann folgenden Auszüge schon äußerlich durch eine hellgelbe
Farbe gekennzeichnet, und die nach dem Abdestillieren des Alkohols erhaltenen Rückstände
haben schon eine hellgelbe Farbe. Für die Darstellung des reinen Glucosids ist gegebenenfalls
noch eine weitere Reinigung erforderlich, und die Destillationsrückstände, die noch
keine Kristallstruktur erkennen lassen, können gleichfalls in die Kristallform übergeführt
werden. Dazu werden die Destillationsrückstände in Wasser zu einer konzentrierten
Lösung aufgelöst, aber aus diesen wäßrigen Lösungen erfolgt die Kristallbildung
nur sehr langsam und nach langem Stehen in der Kälte. Wie aber gefunden wurde, kann
der Vorgang der Kristallisation erheblicher beschleunigt werden, wenn die konzentrierten
wäßrigen Flavonlösungen mit demselben Volumen Essigsäureäthylester überschichtet,
gründlich durchgeschüttelt und dann zum Kristallisieren stehengelassen werden. Nach
kurzer Zeit scheidet sich das Vitexin-4'-rhamnosid in derben Kristallen aus. Der
Essigsäureäthylester wird abgehebert, und die Kristalle können durch Zentrifugieren
von der Mutterlauge abgetrennt werden. Zweckmäßig werden jetzt die Mutterlaugen
mehrerer Ansätze vereinigt, mit dem vorher abgeheberten Essigsäurester wieder
bedeckt
und zum erneuten Kristallisieren stehengelassen. Der beschriebene Vorgang wird noch
einmal wiederholt und die dann. resultierenden Mutter-. laugen im Vakuum bei niedriger
Temperatur stärker konzentriert, mit frischem Essigsäureäthylester bedeckt und erneut
zum Kristallisieren stehe-ngelassen. Auf diese Weise gelingt es, das Vitexin-4'-rhamnosid
fast vollständig zur Kristallisation zu bringen und die Ausbeute von etwa 0,2 bis
0,3 % - auf das Ausgangsmaterial bezogen - zu erreichen. Die von der Mutterlauge
durch Abfiltrieren oder Abschleudern getrennten Kristalle werden wegen ihrer guten
Wasserlöslichkeit zuerst mit Eiswasser und dann mit eiskaltem Äthanol gewaschen.
Das auf vorstehend beschriebene Weise erhaltene Vitexin-4'-rhamnosid stellt gelbe
Prismen dar, die einen Schmelzpunkt von 215° C haben. Durch Umkristallisieren aus
Äthanol ist eine eventuell notwendige Reinigung der Kristalle möglich. Beispiel
60 kg Weißdornblätter werden in einem Druckgefäß zweimal mit je 1001 Methanol auf
60° C erhitzt und 15 Minuten bei dieser Temperatur belassen. Die Droge wird nach
dem Abkühlen abgepreßt und das Methanol bis auf etwa 201 abdestilliert. Die zurückbleibende
dünnflüssige Lösung wird unter Rühren mit 201 Wasser versetzt und nach 24stündigem
Stehen das ausgeschiedene Chlorophyll abgeschleudert. Die Flüssigkeit wird im Vakuum
auf etwa 5 bis 61 eingedampft und mit einer gesättigten Lösung von 3 kg festem Bleiacetat
in Wasser versetzt. Der entstehende Bleiniederschlag wird abgeschleudert und in
der Zentrifuge gründlich mit Wasser gewaschen. Die nach Abtrennung des Bleiniederschlages
erhaltene Lösung sowie die Waschwässer werden vereinigt und mit 2 n-Sch-#vefelsäure
auf eine Wasserstoffionenkonzentration von etwa 4 gebracht. Das sich gut absetzende
Bleisulfat kann durch Filtration abgetrennt werden, und das Filtrat wird sofort
mit Natronlauge auf eine Wasserstoffionenkonzentration von 6 bis 7 gebracht. Diese
praktisch neutrale Lösung wird im Vakuum konzentriert und gegebenenfalls mit Benzol
ausgerührt, um noch vorhandene Chlorophyllreste oder Umwandlungsprodukte des Chlorophylls
zu entfernen. Der dünnflüssige Sirup wird nun so lange mit Buty 1- oder Amy lalkohol
ausgerührt, solange das Extraktionsmittel noch eine positive Reduktionsprobe mit
Zink und konzentrierter Salzsäure durch Rotfärbung ergibt. Die Alkanolauszüge können
laufend im Vakuum abdestilliert und das Destillat zur erneuten Extraktion des Sirups
verwendet werden. Nach vollständiger Extraktion des Sirups, d. h. also bei fast
negativer Reduktionsprobe der Butyl- oder Amylalkoholauszüge wird das Lösungsmittel
restlos im Vakuum abgedampft, wobei ein gelber bis schwach brauner Rückstand bleibt.
Der Rückstand wird dann zu einer konzentrierten Lösung in Wasser aufgelöst und mit
demselben Volumen Essigsäureäthylester versetzt und nach gründlichem Durchrühren
stehengelassen. Nach kurzer Zeit erfolgt die Ausscheidung des Vitexin-4'-rhamnosids
in derben gelben Kristallen. Die organische Phase wird abgehebert und die wäßrige
Phase ent-,veder durch Abschleudern oder durch Abfiltrieren von den Kristallen getrennt.
Durch vorsichtiges Eindampfen der Mutterlaugen im Vakuum und erneuten Zusatz von
Essigsäureäthylester lassen sich noch weitere Mengen des Glykosids gewinnen. Durch
mehrmalige Wiederholung dieses Verfahrens gelingt es, die Ausbeute an Vitexin-4'-rhamnosid
auf 0,2 bis 0,3 % des Ausgangsmaterials zu erhöhen.As a further development of this inventive concept, a new process
