Abstract
Description
B.: L. Vaysse et. al., Journal of Biotechnology, Vol. 53, No. 1, S. 41ff., 1997), dass Hydrolasen, v. a. A number of references are known (e.g .: L. Vaysse et. Al., Journal of Biotechnology, Vol. 53, No. 1, pp. 41ff., 1997) that hydrolases, v. a. Lipases or Acyltransferases or lipases with acyltransferase activity the formation of Can catalyze fatty acid hydroxamic acids, with acyl groups of acyl donors such as Fatty acids or fatty acid methyl esters is transferred to hydroxylamine. Also the chemical synthesis using sodium methoxide as a catalyst and z. B. Oleic acid methyl ester and linoleic acid methyl ester is known.
Fatty acid hydroxamic acids of the general form: R-CO-NHOH could in many biotechnological, biochemical and medical fields are used (especially also because of their high potency as a chelating agent and the biologically very active H-NOH group) such as: removal of toxic metals from waste water etc. → generally chelating agents, as Growth factors, food additives, antibiotics, tumor inhibitors, "antifungal" agents, and cell division factors etc. The difficulty is that these substances have so far been commercial are not available.
A chemically similar and related group of biologically highly active substances is the group of hydroxamic acid siderophores, which are formed by many bacteria and fungi to ensure the iron requirement. Iron is complexed in iron III form with extremely high complex formation constants> 10 20 and can be made available to the organisms by reducing iron III to iron II with a simultaneous pH shift that is often present.
Handbook of Microbial Iron Chelates, CRC Press, 1991; Metal lons in Fungi, Marcel Dekker
These hydroxamic acid siderophores, as described in the standard works by Winkelmann et. al .:
Handbook of Microbial Iron Chelates, CRC Press, 1991; Metal ions in Fungi, Marcel Dekker Inc., 1994 and Transition Metals in Microbial Metabolism, Harwood Academic Publishers, 1997 and also in general hydroxamic acids are also known as laccase mediators (WO 96/18770), whereby they are caused by laccases or other oxidoreductases z. B. Peroxidases are likely to be oxidized to highly active NO radicals. These are the actual oxidants z. B. in the delignification / bleaching of cellulose etc. One of the main difficulties in the technological use of this so-called laccase mediator system (LMS) is the relatively high cost of most effective mediators, which are also consumed in the reaction, which is normally stoichiometric the costs further increased.
The aim of the present invention is therefore to provide an oxidation system provide which does not have these and other disadvantages of the prior art.
- c) Hydrolasen v. a. der Klassen 3.1.1.3 Lipasen (Triacylglycerol Lipasen, Triglyceroacyl
dioxy-Funktion enthalten der allgemeinen Form (A),
This was surprisingly achieved by a modular system, characterized in that using module 1, consisting of:
- a) Acyl donors such as fatty acids, fatty acid esters, fats and fatty esters or mixtures thereof, all of which, as described in WO 98/59108 and DE 101 26 988.9, should be used (but are not limited to these) or anilides or similar compounds and
- b) hydroxylamines and / or their derivatives corresponding to formula A (NO, NOH, and RNH-OH compounds, aliphatic, cycloaliphatic, heterocyclic or aromatic) and
- c) hydrolases, especially of classes 3.1.1.3 lipases (triacylglycerol lipases, triglyceroacyl hydrolases) but also enzymes of classes 3.5.1.4 (amidases) or 3.5.5.1 (nitrilases) or acylases or proteinases which are also supposed to be such according to the invention (but not also these are limited) as described in WO 98/59108 and DE 101 26 988.9, a slow and continuous formation of hydroxamic acids but also other NO, NOH and RNH-OH compounds such as aliphatic, cycloaliphatic, heterocyclic or aromatic compounds which contain at least one N-hydroxy, oxime-N-oxy or N, N'-dioxy function of the general form (A),
and that thereby suitable substrates (oxidation precursor substances) for module 2 of the system:
Laccases or other oxidoreductases (according to the invention oxidoreductases of classes 1.1.1-1.97 according to WO 96/18770) are provided, and that this either enables a permanent replication of the oxidants oxidized by laccase (oxidoreductases) or their recycling.
Amidases of anilides or similar compounds and are also suitable Hydroxylamines and / or their derivatives according to formula (A) the corresponding above to generate described NO, NOH, and RNH-OH compounds according to the invention and / or to recycle.
In addition, according to the invention, a combination of module 1 to all possible in WO 96/18770 but also in WU 98/59108 described Laccase Mediator systems or Hydrolase mediated oxidation systems (HOS), consisting of those described in WO 98/59108 or Components mentioned in DE 101 26 988.9: Hydrolase v. a. Lipase, ketone, fatty acid, peroxides and corresponding fatty acid emulsifiers or the others mentioned in DE 101 26 988.9 Oxidation systems such as peroxynitrous acid oxidation system, ferrocene oxidation system, Organosulfonic acid oxidation system and oxidation system based on activated sulfite and superoxide etc. or the combination of the systems mentioned in DE 102 03 135.5. Preferred ligase-type enzymes are lipases from bacteria and fungi, preferably from Candida parapsilosis, Mucor miehei, or Aspergillus spec. used especially those that show acyltransferase activity.

A system according to the invention is thus made available which, with the aid of system module 1: hydrolases, especially lipases and acyl donors (especially fatty acids, fatty acid esters, fats, fatty acid esters) and hydroxylamines or their derivatives (aliphatic / cyclic), corresponding fat / fatty acid / hydroxamic acids or others Fat / fatty acid NO, NOH, and RNH-OH compounds in system module 2:
Laccases (oxidoreductases) provides as an oxidation substrate.
Areas of application of this oxidation or bleaching system according to the invention are intended all the use in pulp bleaching or pulp bleaching, the use in oxidative Treatment of all types of waste water, use in the manufacture of Wood composites, the use as an enzymatic deinking system, the use as oxidative agent in organic synthesis, use as a bleaching system in Detergents, the use as bleaching agents or oxidizing agents in the textile industry (e.g. stone washing and bleaching of fabrics) and use in coal liquefaction of brown or hard coal, but not limited to these applications. The great advantage of the building block system according to the invention is that of cheap preliminary stages and commercially available enzymes e.g. B. hydroxylamine derivatives, fats, lipases and z. B. laccases can be assumed and that the efficiency, z. B. the LMS system strong can be increased, while at the same time greatly reducing the enzyme and Amounts of chemicals, since the hydroxamic acids etc. are slowly and continuously reproduced overreaction is avoided.
So it was completely surprising by the building block system according to the invention found that cellulose delignification (significant reduction in kappa number) can be achieved.
Furthermore, surprisingly, a considerable bleaching effect could be achieved with bleaching of wood pulps and bleaching of fabrics can be found after deinking processes. In the oxidative treatment of waste water, such as waste water from the Manufacture of wood pulp (wood pulp, refiner), from the pulp industry and contaminated with dye Sewage, e.g. B. the textile industry, surprisingly, could also be an oxidative Polymerization of lignin and / or lignin-like substances and / or decolorization be detected.
This oxidative polymerization of lignin and / or lignin-like substances was also able surprisingly when used in the manufacture of wood composites (binder and / or adhesive production by oxidative polymerization of the existing ones Polyphenylpropane body).
In addition, surprisingly, an ink detachment at Deink process (probably caused by swelling of the lignin-containing waste paper fibers) be detected.
Surprisingly, coal liquefaction properties also became apparent during the treatment of brown or hard coal found.
In addition, surprisingly, a high and selective oxidation power was also found in the Use as an "oxidizing agent" in organic synthesis, a strong bleaching power when using bleach in detergents and in general Bleaching of textile fabrics or the special bleaching when used in stone-wash Processes, namely as a substitute for mechanical color removal and / or after bleaching these processes.
In the case of pulp bleaching, the process according to the invention can be used not only for Delignification (bleaching) of sulfate, sulfite, organosolve, etc. Pulp and of Wood pulps are used, but also in the production of cellulose in general, be it from wood or annual plants if defibrillation by the usual Cooking processes (possibly associated with mechanical processes or pressure), d. H. a very gentle cooking up to kappa numbers, which are in the range of approx. 50-120 kappa can, is guaranteed.
In the bleaching of cellulose as well as in the production of cellulose or the treatment of wood pulp, the treatment with the oxidation and bleaching systems according to the invention can be carried out once or repeated several times, either before and / or after washing and extracting the treated material with NaOH etc. or without these intermediate steps also before and / or after all common and known pretreatment and / or post-treatment steps such as acid washing, acid treatment for removing hexonuronic acids, Q stages, alkaline leaching, bleaching stages: such as peroxide bleaching, O 2 -reinforced peroxide stages, pressure peroxide stages, O 2 delignification, Cl 2 bleach, ClO 2 bleach, ClO 2 bleach, ClO 2 bleach, peracid bleaching stages, peracid-enhanced O 2 bleach / peroxide bleaching, ozone bleaching, dioxirane bleaching, reductive bleaching stages, other treatments such as: swelling stages, Sulfonations, NO / NO 2 treatments, nitrusylsulfuric acid treatment. B. Treatments with hydrolases such as cellulases and / or hemicellulases (e.g. xylanase, mannanase etc.) and / or amylases and / or pectinases and / or proteinases and / or lipases and / or amidases and / or oxidoreductases such as e.g. B. laccases and / or peroxidases etc. or several combined treatments take place before and after the above-mentioned HOS systems, LMS systems, the systems according to DE 101 26 988.9 and systems mentioned in DE 102 03 135.5.
This leads to kappa values which can be reduced still further and to substantial increases in whiteness. In particular, an O 2 stage can be used before / and or after the treatment with the oxidation and bleaching systems according to the invention, or, as already mentioned, an acid wash or Q stage (chelate stage) can be carried out.
Im folgenden wird die Erfindung anhand von Beispielen näher erläutert: The invention is explained in more detail below with the aid of examples:

Then the fabric is washed over a nylon sieve (30 µm) and 1.5 hours at 70 ° C, 2% consistency and 8% NaOH extracted per g of pulp.
After washing the fabric again, the kappa number is determined.
Es konnte eine Delignifizierung von 38% erreicht werden. A delignification of 38% was achieved.
