Stand der TechnikState of the art
The analysis of liquid samples, for example blood or urine, for the detection of diseases is known. For example, a blood analysis can be used in the diagnosis of cancer. This is also referred to as liquid biopsy, whereby the patient is spared a tissue biopsy. Cancer diagnostics focus on the detection of DNA from tumor cells in the blood or directly on the detection of circulating tumor cells (CTCs). In order to be able to isolate and detect certain target cells, for example CTCs, it has hitherto generally been necessary to use a specific marker with which the target cells can to a certain extent be extracted from the sample or can be isolated. However, such markers are often very specific and can only bind to a very specific group of subpopulations, which is why this approach is less suitable for a “universal” cancer test.

Vorteile der ErfindungAdvantages of the invention
The invention provides an apparatus for the dielectrophoretic separation of target particles in a liquid. The functioning of this device is based on the fact that the target particles have a different crossover frequency than other particles in the liquid. The property is used to dielectrophoretically separate the target particles from other particles in the liquid or to separate and / or isolate them from the liquid. This is achieved by applying an alternating electrical field, a frequency being selected which lies between the respective crossover frequencies for the target particles (for example cancer cells, in particular CTCs) and for the other particles (for example healthy blood cells), so that the remaining particles negative dielectrophoretic forces act and positive dielectrophoretic forces act on the target particles. The device comprises a composite of at least two, for example cylindrical, electrical conductors that extend in length. Both electrical conductors are each covered by an insulator layer, the conductors being firmly connected to one another in the longitudinal direction, for example by plasma bonding. In this case, a depression is preferably formed in the longitudinal direction above and below the connection (bond point), to a certain extent a trench, an area with high field strength being able to be formed in this depression when an alternating electrical field is applied to the electrical conductors. To form this recess, the electrical conductors can be oval or circular in cross section, for example, so that such a recess (trench) is formed above and below the connection of the electrical conductors. However, other cross-sectional shapes of the electrical conductors can also be selected. When an AC voltage is applied at a suitable frequency, the target cells will move to the areas with high field strengths in the wells while other particles are rejected. The recessed areas or trenches act as cell retention areas. In this way, the target particles can be separated or isolated. The crossover frequency of the target particles is used to a certain extent as a marker or separation feature in order to isolate the target particles. The particular advantage here is that the target particles are not separated on the basis of specific surface proteins or the like, but that this approach realizes a much more universal method, for example for a general cancer test can be used. The fact that the electrical conductors themselves are each covered by an insulator layer, which is in particular an inert, biocompatible material, does not result in the electrical conductors themselves being contaminated or contaminated with the sample, so that the device can easily be used again is usable.
The liquid or sample itself is preferably human or animal blood. The target particles are preferably cells, in particular circulating tumor cells (CTCs), so that the device can be used in the context of cancer diagnosis. However, other biological cells can also be separated and / or isolated using the device. In principle, the device is suitable for separating any type of target particles in a liquid which are to be separated from other particles, the prerequisite for the function of the device being that the target particles and the other particles differ in their crossover frequency.
A particular advantage of the device is that the device can be used in flowing samples or liquids, that is to say in continuous samples. The device itself can be introduced into the liquid, for example into blood. It is thus possible for the device to be introduced directly into the blood circulation of a person or an animal, in order to remain there in a blood vessel and to accumulate or separate tumor cells in the deep areas (trenches) of the conductor network over a certain period of time , Because the device remains in the body or in the bloodstream, the sample volume that flows around the device is very large. This is a particular advantage of the present device, since the proportion of CTCs is usually very small, for example one cell per milliliter. A large sample volume is therefore required for analysis or diagnostics in order to isolate a sufficiently high number of CTCs. A blood sample of the order of 500 ml, for example, which would be required for a corresponding diagnosis outside the body, is prohibited for medical reasons. The device described here solves this problem in that the device for separating the target particles or the tumor cells can be introduced directly into the bloodstream. Since the electrical conductors or the electrodes that form the composite are located close to one another, the voltage required for generating the alternating electrical field for the dielectrophoresis is only low, so that this approach can be used in vivo without risk to the patient. Nevertheless, the device can also be used outside of a patient's body and used for the analysis of blood or other body fluids.
For example, a hollow needle or cannula can be used to introduce the device into the bloodstream. For this purpose, for example, an access can first be made with a hollow needle, before the device, that is to say in particular the assembly with the electrical conductors, is then inserted into the hollow needle. This is not significantly more uncomfortable for the patient than, for example, taking a blood sample or administering medication via such an access. It also saves the patient from having to take a large blood sample or even a tissue biopsy.
In a preferred embodiment of the device, the composite of the electrical conductors can have an additional layer or a thickening region made of a flexible and / or soft material at one end of the composite, namely at the end that is not intended for connection to a voltage source , for example made of soft insulating material. This additional layer can be implemented in the form of a drop, for example, and has the particular advantage that it minimizes the risk of injury to a blood vessel when the device is inserted into the patient's bloodstream. Furthermore, a spacer, in particular an insulating spacer, can be provided at at least one point in the composite, in particular around the composite. This spacer serves to limit the advance of the composite within a hollow needle, by means of which the device can be inserted into the bloodstream. This prevents the compound that is inserted through the hollow needle from being pushed too far into the blood vessel. For this purpose, the diameter of this spacer is expediently larger than the diameter of the access, that is to say the hollow needle.
In a particularly preferred manner, the composite of the electrical conductors is turned on, as a result of which the device can be realized in a small and very space-saving manner. For example, the composite can be turned into a spiral, screw, screw or helical shape. On the one hand, the length of the depression or the trench length between the conductors, which is available for the separation and / or isolation of the target particles, is increased. At the same time, the inconvenience for the patient during use of the device can be reduced even further by the rolled-up shape and the small size of the device.
In one configuration of the device, the assembly of two is connected to one another electrical conductors formed. However, more than two conductors can also be connected to one another. For example, the composite can consist of four interconnected electrical conductors, the individual conductors expediently being connectable crosswise to a voltage source. Increasing the number of electrical conductors in the assembly has the advantage that the total length of the recess areas or trenches between the interconnected conductors is doubled, for example. In the case of two conductors connected to one another, there are two trenches along the length of the composite, whereas with four conductors connected to one another there are four trenches pointing outwards. The increase in the number of conductors is therefore associated with an increase in the effectiveness of the device with the same length of the composite. Increasing the number of conductors in the network further increases the stability of the overall network. A composite of four or more electrical conductors can also be opened in order to reduce the external dimensions of the device.
In a particularly preferred embodiment of the device, the insulator layer of the individual conductors forming the composite is at least partially provided with an unspecific cell-affine coating, for example with fibronectin and / or collagen. This ensures that target particles or target cells that have already been captured continue to adhere to the composite even after the applied voltage has been switched off, so that the device can be easily removed from the blood circulation even after the voltage has been switched off without the adhered target cells detaching from the device , The non-target particles or non-target cells are generally not influenced by the non-specific cell-affine coating, since the non-target particles are repelled by the acting negative dielectrophoretic forces during the applied electrical alternating field.
The invention further comprises a method for the dielectrophoretic separation of target particles in a liquid, the target particles having a different crossover frequency than other particles in the liquid. For this method, a device is introduced into the liquid as described above, the device comprising a composite of at least two electrical conductors connected to one another in the longitudinal direction. An alternating voltage with a suitable frequency is applied to the composite of the device, so that the target particles accumulate in the depressions or trenches of the composite according to their dielectrophoretic properties due to the positive dielectrophoretic forces acting on them. Depending on the concentration and number of target particles in the respective liquid or sample, the method can be carried out over a longer period of time until sufficient target particles have accumulated. The device can then be removed from the liquid, the target particles deposited in the depressions of the composite being isolated and further analyzed. Here, for example, a simple counting or a more detailed analysis can be carried out, for example, using molecular biological immunobiological methods. The device can be used in vitro, ie outside a body, or in vivo. If the target particles are certain blood cells, for example circulating tumor cells, the device can be inserted directly into the bloodstream of a patient, for example. For this purpose, an access is preferably first made with a hollow needle or cannula before the device or the composite is then inserted into the hollow needle and thus enters the patient's bloodstream. The compound can remain in the bloodstream over a longer period of time, for example 30 to 60 minutes, in order to be able to accumulate sufficient target cells. A suitable alternating voltage can preferably be applied during the insertion of the device or the composite into the hollow needle, so that non-specific adhesion of non-target particles or non-target cells is avoided.
A suitable metal conductor is, for example, a metal wire which can have a diameter of approximately 50 μm to approximately 200 μm, for example. Suitable lengths for the composite of two or more electrical conductors are, for example, 5 to 15 cm. If the connection of the electrical conductors is opened, the length can also be significantly longer. The insulator layer is, in particular, an electrically insulating layer made of, for example, polymers. For example, PDMS (polydimethylsiloxane), which is non-toxic and chemically inert, is suitable. PDMS is characterized by a high level of biocompatibility. An alternating current is applied for the voltage supply, for example in a range between 10 and 20 V. Suitable frequencies are in particular in the range between 10 and 200 kHz, the frequency applied in each case being dependent on the crossover frequencies of the target particles and the non-target particles. For example, healthy cells as non-target particles, for example mononuclear blood cells, can have a crossover frequency of more than 120 kilohertz, while the CTCs as target particles have, for example, crossover frequencies between 20 and 100 kilohertz.
