Aus diesen Einsatzbeispielen ergeben sich bestimmte Anforderungen an die Sensoren, sowie Voraussetzungen für ihren Einsatz. Bei Verwendung im Bereich medizinischer Untersuchungen oder biologischer Proben sollte der Sensor, ebenso wie weitere Komponenten einer Messapparatur, steril sein, um die Probe, etwa eine zu überwachende Zellkultur, nicht mit Fremdorganismen zu kontaminieren. Beispielsweise können ein oder mehrere Sensoren in einem Sterilbehälter, etwa einem Einweg-Bioreaktor, eingesetzt werden. Bei wiederholten Messungen über einen langen Zeitraum kann sich eine Drift des Sensors zeigen, welche eine Nachkalibrierung des Sensors erforderlich macht. Ebenso werden manche Sensorarten durch den langen Kontakt mit einer Probe beeinträchtigt, was für längere Messreihen offensichtlich ein Problem darstellt. Eine einfache Nachkalibrierung des Sensors oder eine vorübergehende Entfernung des Sensors aus der Probe ist nicht immer möglich, da dadurch, je nach Einsatz, beispielsweise eine Sterilbarriere durchbrochen werden müsste. Eine Nachkalibrierung in der Probe ist praktisch nicht machbar, oder lediglich unter Beeinträchtigung der eigentlichen Messung. So müsste man in einem Bio-Reaktor zur Nachkalibrierung eines pH-Sensors den pH-Wert im gesamten Bio-Reaktor auf definierte Werte einstellen; diesen pH-Werten wären auch im Bio-Reaktor befindliche Zellen ausgesetzt, was im Sinne der eigentlichen Prozessführung unerwünscht bzw. auch nicht unbedingt möglich ist. Falls ein Sauerstoffsensor nachkalibriert werden soll, so müsste man, um den Nullpunkt (0% Sauerstoff) einzustellen, das gesamte Zellkulturmedium im Bio-Reaktor, und gegebenenfalls auch darin bereits befindliche Zellen, mit Stickstoff begasen, um den Sauerstoff auszutreiben. Zur Einstellung des Luftsättigungswertes müsste der Reaktorinhalt mit Luft begast werden. Abgesehen davon, dass etliche Zellkulturmedien zum Schäumen neigen, was das Begasen an sich schon ungünstig erscheinen lässt, kann eine solche Begasung auch die Zellkultur im Bio-Reaktor schädigen. Auch eine neuerliche Sterilisation des Sensors ist problematisch, da der Sterilisationsprozess den Sensor beeinträchtigen kann, womit die zuvor erfolgte Nachkalibrierung wieder gestört wird. Schließlich ist es auch problematisch, dass Sensoren häufig vor dem Einsatz länger gelagert werden müssen. Manche Sensoren erfordern zum Erhalt der Funktionalität oder zum Zwecke einer schnellen Einsatzbereitschaft einen gewissen Gehalt an Wasser; Beispiele sind etwa enzymbasierte Sensoren wie Glucosesensoren oder optische Kohlendioxidsensoren. Während einer solchen Lagerung trocknet ein solcher Sensor gemeinhin aus und muss vor dem Einsatz längere Zeit, etwa 24 Stunden, gewässert werden. Für die Dialysemembran, etwa aus Celluloseacetat, Polysulfon oder Polyethersulfon, ist eine den diffundierenden Molekülen entsprechende Durchlässigkeitsgrenze, etwa 1-100 kDa zu wählen.
