Abstract
Description
The invention relates to a specific vector construct for enzyme expression and to a method for influencing cells transfected with this vector construct in a cell culture rapidly, with little effort and with high specificity, in particular for the purpose of their isolation.
The vector construct according to the invention and the proposed method can be used in particular in molecular biology for the rapid and simple selection of cell clones after transfections.
In this case, vector systems used often contain complete viral genetic information, or parts of the viral genes have been eliminated, for example the genetic information about envelope proteins, enzymes for processing proteins or for packaging the genetic material into the virus envelopes.
The described reporter genes and the antibiotic resistance genes can be used to select the cells containing the viral or synthetic vectors. The selection by means of "cell sorting" on the expression of GFP, however, leads to cell stress due to the methodology; selection by antibiotic resistance is tedious and often does not result in a pure cell population after selection. Therefore, often the selection process must be repeated or it is necessary to constantly add antibiotics.
The invention is based on the object to influence vector-transfected cells quickly and in the simplest possible way, for example, to select them with high purity.
An expression vector is proposed which, in addition to at least one known promoter and also interfaces for inserting use-specific gene sequences according to the invention for the purpose of activating a peptide-inhibited siRNA, contains the genetic information for expressing an enzyme (which cleaves said peptide-inhibited siRNA) which is derived from the Otherwise expression vectors-transfected cells can not be generated. This enzyme is capable of inhibiting the covalent binding of said peptide-inhibited siRNA, which is also introduced into the cells, break open and thus activate the siRNA, which then influences the expression of cellular genes.
The expressed enzyme, for example a protease or peptidase, can be used for the reliable selection of these enzyme-expressing cells over other (non-enzyme-expressing) cells of the cell culture. Thus, in particular, a technically advantageous and low-cost isolation of the above-selected cells is possible when in the expression vector-transfected cells of a cell culture, which are usually grown in a culture vessel and in this growth process attached to the same (which is caused by the expression of specific adhesion molecules becomes), said adhesion-molecule-depression is inhibited. By the expression of the protease or peptidase in the expression vector transfected cells, a special peptide-inhibited siRNA can be activated, which after activation, for example, prevents the expression of said adhesion molecule. Due to the inhibition of the expression of this or these adhesion molecules, the expression vector-transfected cells from the culture vessel, so that they can be easily separated from the other non-enzyme-expressed cells, for example, for their isolation.
Non-expression vector transfected cells are unable to activate the peptide-inhibited siRNA; there is no RNA interference and no cell detachment of culture vessel. In this way, the expression vector transfected cells can be removed with the supernatant and cultured again. The content of the siRNA in the cells decreases after a few hours or days, and there is again an expression of the cell adhesion molecules and thus a re-adhesion of the cells to the culture vessel. Thus, the physiological in vitro growth situation is restored and the expression vector-transfected cells were selected. In the event that the purity of the transfected cells is not sufficiently high, the described selection step can be repeated by introducing peptide-inhibited siRNA.
Should cells have died during these processes, they also float in the cell culture supernatant and are taken together with the expression vector-transfected cells in the described new culture vessel. However, these dead cells will not adhere again and can therefore be removed in the context of routine cell culture.
However, the use of the proposed expression vector to activate a peptide-inhibited siRNA is not limited to the aforementioned selection / isolation of the enzyme-expressing cells. For example, the invention can also find application for the targeted killing of expression vector cells. This is of particular interest in stem cell research in which stem cells are introduced in vivo and may not show the desired effects or form terratomas. If these cells are transfected prior to their introduction with an expression vector of a proposed type, a peptide-inhibited siRNA can be introduced in vivo and activated exclusively in these cells. The target gene of the siRNA could be a cell vital for the survival of the cell, which would lead to the death of the cell. This could reduce side effects of in vivo-introduced stem cells, with the exemplified application not being limited to stem cells.
Furthermore, the described mechanism can be used for diagnostic or physiological considerations.
