den Wirkstoff zu gewährleisten. Die wasserummantelten Zellen
wurden mit Wasser von 38°C temperiert, so dass eine Temperatur
von 32°C an der Permeationsfläche der Haut resultierte.
Das Medium wurde mit einem Magnetrührer bei 500 UpM kontinuierlich
gerührt. 150 μL der gesättigten Anastrozol-Lösung
wurden auf die Haut gegeben. Zu definierten Zeitpunkten wurden Proben
entnommen und in HPLC-Vials gesammelt. Das entnommene Probenvolumen
wurde durch frisches und temperiertes Hautpermeationsmedium ersetzt.
Diese Verdünnung wurde bei der Berechnung der kumulativ
permeierten Mengen an Anastrozol berücksichtigt. Am Ende
des Permeationsexperimentes wurden die Proben mit einer selektiven
HPLC-analytischen Methode mit UV-Detektion auf den Gehalt an Anastrozol
untersucht. Die kumuliert permeierten Mengen wurden gegen die Zeit
in einem kartesischen Koordinatensystem aufgetragen. Die Steigung
des linearen Teils des Profils wurde als steady state Fluss definiert, der
x-Achsenabschnitt als lag-Zeit. Zusätzlich wurden Durchschnittsflüsse
bezogen auf 24 und 48 Stunden berechnet. Das Permeationsprofil von
Anastrozol ist in

Das Massenverhältnis der festen Bestandteile betrug wie folgt: Anastrozol zu Silikon 1:99, bzw. im Falle der Verwendung von Permeationsverstärkern Anastrozol:Permeationsverstärker:Silikon 1:10:89. Nach einer Homogenisierungsphase wurde die Klebstofflösung mit Hilfe einer Rakel auf eine fluorpolymerisierte PET-Folie aufgetragen und der Film einem kontrolliertem Trocknungsprogramm unterworfen, um die Prozesslösemittel Ethylacetat und n-Heptan zu entfernen. Anschließend wurde der getrocknete Film mit einer PE-Membran kaschiert. Das durchschnittliche Gewicht der Anastrozol enthaltenen Adhäsivschicht wurde zu etwa 55 mg pro 10 cm2 bestimmt. Der steady state Fluss konnte zu 2,07 (0,13) μg/cm2/h bestimmt werden und lag somit überraschenderweise nur geringfügig unter dem aus einer gesättigten Lösung mit maximaler thermodynamischer Aktivität (vgl. einer Mischung aus 2-Propanol und Ethylacetat (1:1) gelöst
und anschließend dem in Ethylacetat gelösten Polymer
zugegeben. Nach einer Homogenisierungsphase wurden die TTS analog
Beispiel 2 hergestellt. Die Abwesenheit von Kristallen sowie die
Homogenität der TTS wurden durch polarisationsmikroskopische
Untersuchungen nachgewiesen. Das aus dem Permeationsversuch ermittelte Profil
ist in
