Abstract
Verfahren zur Identifikation eines Partikels aus einem polymeren Werkstoff, der zwei oder mehr monodispers in dem Werkstoff eingebettete Fluorophore mit Absorptions- und Fluoreszenzmaxima im UV/VIS/NIR-Spektralbereich enthält, wobei die Fluorophore so ausgewählt sind, dass (i) die Absorptionsmaxima der Fluorophore um jeweils mindestens 5 nm voneinander beabstandet sind; (ii) die Fluoreszenzmaxima der Fluorophore um jeweils mindestens 10 nm voneinander beabstandet sind; und (iii) die Absorptionsmaxima und Fluoreszenzmaxima der Fluorophore um jeweils mindestens 5 nm voneinander beabstandet sind in einer Probe, umfassend die Schritte: (i) Bestrahlen von vorzugsweise in Monoschicht vorliegenden Partikeln mit einer oder mehreren Anregungswellenlängen im Überlappungsbereich der Absorptionsspektren der Fluorophore bzw. im Bereich der Absorptionsmaxima der Fluorophore (+/–10% der Extinktion); (ii) Erfassen der Intensitäten der Fluoreszenz an vorgegeben Messpunkten, wobei für jeden im Partikel enthaltenen Fluorophor wenigstens ein Messpunkt vorgegeben wird, der in einem Bereich von mindestens 5% der Intensität des Fluoreszenzmaximums des jeweiligen Fluorophors liegt und wobei die Messpunkte um mindestens 5 nm voneinander beabstandet sind; (iii) Bestimmen eines Intensitätsverhältnisses der an den Messpunkten erfassten Intensitäten; und (iv) Vergleichen des bestimmten Intensitätsverhältnisses mit einem Referenzwert zur Identifikation des Partikels, dadurch gekennzeichnet, dass die Absorptionsspektren der Fluorophore einen gemeinsamen Anregungswellenbereich aufweisen, wobei in diesem Bereich für jeden Fluorophor eine Extinktion von mindestens 0,001 und eine Quantenausbeute von mindestens 0,01 gilt und das Bestrahlen im Schritt (i) unter Verwendung einer einzigen Anregungswellenlänge erfolgt, die im Überlappungsbereich liegt.Method for identifying a particle made of a polymeric material that contains two or more fluorophores embedded in the material with absorption and fluorescence maxima in the UV / VIS / NIR spectral range, the fluorophores being selected such that (i) the absorption maxima of the fluorophores are spaced from each other by at least 5 nm; (ii) the fluorescence maxima of the fluorophores are each spaced apart by at least 10 nm; and (iii) the absorption maxima and fluorescence maxima of the fluorophores are each spaced apart by at least 5 nm in a sample, comprising the steps of: (i) irradiating particles preferably present in monolayer with one or more excitation wavelengths in the overlap region of the absorption spectra of the fluorophores and im Range of absorption maxima of the fluorophores (+/- 10% of the absorbance); (ii) Detecting the intensities of the fluorescence at predetermined measurement points, with at least one measurement point being specified for each fluorophore contained in the particle, which lies in a range of at least 5% of the intensity of the fluorescence maximum of the respective fluorophore and with the measurement points being at least 5 nm apart are spaced; (iii) determining an intensity ratio of the intensities detected at the measurement points; and (iv) comparing the determined intensity ratio with a reference value for identifying the particle, characterized in that the absorption spectra of the fluorophores have a common excitation wave range, in this range an extinction of at least 0.001 and a quantum yield of at least 0.01 applies to each fluorophore and the irradiation in step (i) is carried out using a single excitation wavelength which lies in the overlap region.

Bei Aggregation der Fluorophore im Partikel treten intermolekulare Wechselwirkungen auf. Es ist mit einem konzentrationsabhängig veränderten spektroskopischen Verhalten zu rechnen. Zudem ist die gemessene Fluoreszenzintensität von der absoluten Gesamtstoffmenge der Fluorophore im Partikel und somit auch von der jeweiligen Größe der Partikel abhängig. Liegen in einer zu untersuchenden Probe eine Anzahl n verschiedener Partikel vor, so müssen diese fluoreszenzspektroskopisch unterscheidbar sein; entsprechend ist bei herkömmlicher Vorgehensweise eine Anzahl n verschiedener und spektroskopisch unterscheidbarer Fluorophore einzusetzen bzw. bei einem vorgegebenen Fluorophor sind Partikel mit abgestuften Konzentrationen des Fluorophors herzustellen. Augenscheinlich nachteilig ist, dass die Anzahl der für die Zwecke tauglichen, in ausreichendem Maße zugänglichen, und in ihren Kosten wirtschaftlich vertretbaren Fluorophore begrenzt ist. Zu Bedenken ist auch, dass ein Austausch einer Komponente (d. h. eines Fluorophors oder einer Komponente des Polymermaterials) des Partikels unter Umständen eine Modifikation des gesamten Systems erfordert, sei es durch eine notwendige Anpassung des Herstellungsweges oder den Austausch weiterer Komponenten, die ansonsten eine die spektroskopische Untersuchung störende Interaktion bedingen könnten. Schließlich ist auch das Identifikationsverfahren für Proben mit einer Vielzahl verschiedener Partikel mit jeweils verschiedenen Fluorophoren aufwendig: Es sind geeignete Anregungswellenlängen für die verschiedenen Fluorophore vorzugeben und die Erfassung der Fluoreszenz ist gleichzeitig für die Vielzahl der verschiedenen Fluorophore zu optimieren. - (ii) die Fluoreszenzmaxima der Fluorophore um jeweils mindestens 10 nm voneinander beabstandet sind; und
- (iii) die Absorptionsmaxima und Fluoreszenzmaxima der Fluorophore um jeweils mindestens 5 nm voneinander beabstandet sind.

Eine zu untersuchende Probe kann also verschiedene Arten von Partikeln enthalten, die sich jeweils im Verhältnis der erfassten Fluoreszenzintensitäten und damit relativen Anteilen der Fluorophore unterscheiden, ansonsten jedoch einen identischen Aufbau besitzen. Natürlich können daneben auch Partikel mit unterschiedlichen Fluorophoren verwendet werden. Die in dem polymeren Werkstoff verteilten Fluorophore sind monodispers verteilt, liegen also im Idealfall als homogene Lösung der Fluorophore in der polymeren Matrix vor, oder sind, wenn es sich um eine Dispersion handelt, als Farbstoffteilchen weitgehend gleicher Größe in der polymeren Matrix eingebettet. Die Absorptionsspektren der Fluorophore weisen einen Überlappungsbereich auf, wobei in diesem Bereich für jeden Fluorophor eine Extinktion größer 0,001 und eine Quantenausbeute größer Null ist. Hierdurch kann – in noch näherer erläuterter Weise – das Identifikationsverfahren durch Anregung mit nur einer Wellenlänge eingeleitet werden. Das erfindungsgemäße Identifikationsverfahren von Partikeln der vorgenannten Ausführungsform in einer Probe. umfasst die Schritte:
- (i) Bestrahlen von vorzugsweise in Monoschicht vorliegenden Partikeln mit einer oder mehreren Anregungswellenlängen im Überlappungsbereich der Absorptionsspektren der Fluorophore bzw. im Bereich der Absorptionsmaxima der Fluorophore (+/–10% der Extinktion);
- (ii) Erfassen der Intensitäten der Fluoreszenz an vorgegebenen Messpunkten, wobei für jeden im Partikel enthaltenen Fluorophor wenigstens ein Messpunkt vorgegeben wird, der in einem Bereich von mindestens 5% der Intensität des Fluoreszenzmaximums des jeweiligen Fluorophors liegt und wobei die Messpunkte um mindestens 5 nm voneinander beabstandet sind;
- (iii) Bestimmen eines Intensitätsverhältnisses der an den Messpunkten erfassten Intensitäten; und
- (iv) Vergleichen des bestimmten Intensitätsverhältnisses mit einem Referenzwert zur Identifikation des Partikels.

Die Apparatur zur Durchführung der spektroskopischen Untersuchung als auch das Identifikationsverfahren selbst muss nur einmalig auf ein ganz spezifisches spektrales System eingestellt/optimiert werden, um eine Vielzahl unterschiedlicher Partikel zu identifizieren.
