The monitoring system is used to determine and document blood pressure, heart rate, respiratory rate, O 2 concentration (finger-pulse oximetry), body temperature and, if possible, the oxygen and carbon dioxide concentration of the respiratory air for the entire duration of the experiment. Before starting the planned application, a venous access is placed on the right and left arms of the subject. One access is used to administer the required study medication (repinotan and one opiate eg Remifentanil (Ultiva ® , G1axoSmithKline) as well controllable opiate), the other contralateral approach is used to take blood samples to determine the drug concentrations (repinotan, opiate, eg remifentanil). Optionally, the analgesic effect of the opiate is registered with a validated pain model.
Durchführung: Jede Dosisstufe besteht aus zwei Studienperioden in denen jeweils randomisiert Behandlung A und Behandlung B durchgeführt werden.
- • Behandlung A: Zum Zeitpunkt 0h wird die Opiatinfusion (z.B. Remifentanil) mit der körpergewichtsbezogen Anfangsinfusionsrate gestartet. Nach jeweils 5 Minuten wird die Infusionsrate verdoppelt bis das Abbruchkriterium (s.u.) erfüllt ist.
- •Behandlung B: Zum Zeitpunkt -5h wird die Infusion mit Repinotan mit einer fest vorgegebenen Infusionsrate (IRepinotan) gestartet. Zum Zeitpunkt 0h wird zusätzlich die Opiatinfusion (z.B. Remifentanil) gestartet. Die Infusionsrate ist mit der unter A beschriebenen Behandlung identisch. Nach jeweils 5 Minuten wird die Infusionsrate verdoppelt bis das Abbruchkriterium (s.u.) erfüllt ist.
