Description
The invention relates to a method for the selective separation of the genetically conditioned and / or formed during ontogenesis human or animal odor complex or other essential information containing complexes of body fluids, tissue and cell extracts, body exudates and other Körperbestandteüen and the concentration of this odor complex a suitable matrix.
Methods for the selective enrichment of the human or animal odor complex or of the substance groups responsible for this complex from body fluids or adequate substances in the liquid phase, which are in the field of application of the invention, are not known.
The object of the invention is to create conditions by means of a selective separation of said complexes, to unambiguously assign human trace material to a person and to identify (objectify) this assignment by equipment analysis, to create conditions for the development of new personal recognition systems and to lay the foundations for new medical diagnostic methods in an extraordinary manner develop sensitive area.
The invention analogously includes the detection and evaluation of animal odor complexes and thus opens up new possibilities for veterinary medicine and animal production.
The invention has for its object to develop a method which makes it possible, located in a liquid or colloidal phase, individual or other essential features and properties-containing substance complex, which also determines the human and animal individual odor of other (disturbing) remove odor-active, odorless, volatile, liquid and solid components, to concentrate it on a suitable matrix many thousands of times and thus make it accessible for a device analysis or other use.
In contrast to the view presented in US Pat. No. 4,451,460 of Apr. 29, 1984, that "components of the basic structure of human odor are due to various secretions, primarily sweat and urine," the invention assumes that the human and animal Art and individual odor not in glands, such as the apocrine sweat glands, but produced inside the organism and is part of the blood serum or plasma, which can be detected by differentiation dogs.
According to the invention the object dddurch solved that the liquid starting material is passed at temperatures of 15-75 ° C over a located in a column in preferably a gel state polymer located until the desired degree of concentration is reached. The gel is then rinsed with a buffer solution and sent for further processing (separation and analysis of the odor complex). The ratio volume of the starting material to gel volume depends on the desired degree of concentration. According to the known laws of the gel filtration processes, it would be expected that the odor-specific substance complex of interest occurs in an independent fraction or coupled with other fractions, as is the case with other odorous substances. In fact, however, a complete uptake of said odor or substance complex by the gel takes place. Thus, there is a new surprising effect, since other not belonging to said complex odor-active or volatile (odorless) substance complexes are not retained by the gel.
Polysaccharide-based polymers and corresponding derivatives, in particular polygalactans such as agarose and other algal galactans, polyglucans such as starch and starch products, dextranes such as Sephadex and Sephacryl, polyacrylamide gels of various structures and compositions, and mixtures of the stated starting materials in a wide range of mixing ratios are used as the gel matrix.
The polymerization of the gels is carried out in buffer solutions, pH range about 3-10, ionic strengths about 0.01 to 0.2 mol / l and a temperature of 60-11O 0 C. Other polymerization conditions resulting in a sufficiently permeable and the specific complex well-absorbing polymer can also be used.
Ausfuhrungsbeispielexemplary
Human blood serum is passed through a gel consisting of 70% agarose and 30% acrylamide in a glass column thermostated at 37 ° C. The gel was prepared with a phosphate buffer pH 8 and an ionic strength of 0.09. After passing through the serum, the Ge! rinsed with ten times the volume of said phosphate buffer and then sent for further use.
